Iran’s slow COVID-19 vaccination will see long impacts

Significance The slow pace of vaccination and the arrival of the Delta variant caused a deadly new pandemic crisis in Iran. However, the new government of President Ibrahim Raisi, like its predecessor, has resisted lockdowns as a solution. Impacts The five waves of COVID-19 will likely result in higher antibody prevalence across the Iranian population than in less-affected countries. The dominance of the Sinopharm vaccine, which is less protective against Delta, will increase the importance of booster shots. The development of domestic vaccines will be used to validate the doctrine of the ‘resistance economy’. Vaccine development could create export opportunities and leave the country more secure in the face of future biological threats.

COVID-19 vaccine development has been widely awaited, but concerns around acceptance and political polarisation prevail. We sought to determine the willingness of Australians to take a (then prospective) COVID-19 vaccine, compared with their previously recorded opinions about other vaccines. We also sought to determine reasons for hesitancy, levels of support for possible government mandates, and the political basis of support. We surveyed 1200 Australians, including 898 participants in a panel previously asked in 2017 about vaccines and mandates. In all, 66% of respondents indicated they would take a coronavirus vaccine, less than the 88% who in 2017 agreed that vaccines are safe, effective, and necessary. Also, 70% of the respondents who indicated hesitancy were concerned about the safety of the vaccine if it was developed too quickly, and 73% of all respondents agreed that the government should require a coronavirus vaccine for work, travel, and study. This is lower than the 85% who agreed with the childhood vaccine mandate in 2017, but slightly higher than the number of respondents who indicated that they would definitely be willing to take the coronavirus vaccine themselves. Older respondents, higher income respondents, and respondents who vote for major parties were all significantly more likely to take a coronavirus vaccine and to support government requirements.

ABSTRACTStaphylococcus aureusis a commensal bacterium associated with the skin and mucosal surfaces of humans and animals that can also cause chronic infection. The emergence of antibiotic-resistant strains such as methicillin-resistantS. aureus(MRSA) and strains causing chronic intramammary infections (IMI) in cows results in severe human and livestock infections. Conventional approaches to vaccine development have yielded only a few noneffective vaccines against MRSA or IMI strains, so there is a need for improved vaccine development. CD4 T lymphocytes are required for promoting gamma interferon (IFN-γ) mediated immunoglobulin isotype switching in B lymphocytes to produce high-affinity IgG antibodies and IFN-γ-mediated phagocyte activation for an effective resolution of bacterial infection. However, the lack of known CD4 T cell antigens fromS. aureushas made it difficult to design effective vaccines. The goal of this study was to identifyS. aureusproteins recognized by immune CD4 T cells. Using a reverse genetics approach, 43 antigens were selected from theS. aureusNewman strain. These included lipoproteins, proteases, transcription regulators, an alkaline shock protein, conserved-domain proteins, hemolysins, fibrinogen-binding protein, staphylokinase, exotoxin, enterotoxin, sortase, and protein A. Screening of expressed proteins for recall T cell responses in outbred, immune calves identified 13 proteins that share over 80% sequence identity among MRSA or IMI strains. These may be useful for inclusion in a broadly protective multiantigen vaccine against MRSA or IMI.

Object. Transthoracic endoscopic T2–3 sympathectomy is currently the treatment of choice for palmar hyperhidrosis. Compensatory sweating of the face, trunk, thigh, and sole of the foot was found in more than 50% of patients who underwent this procedure. The authors conducted this study to investigate the associated intraoperative changes in plantar skin temperature and postoperative plantar sweating. Methods. One hundred patients with palmar hyperhidrosis underwent bilateral transthoracic endoscopic T2–3 sympathectomy. There were 60 female and 40 male patients who ranged in age from 13 to 40 years (mean age 21.6 years). Characteristics studied included changes in palmar and plantar skin temperature measured intraoperatively, as well as pre- and postoperative changes in plantar sweating and sympathetic skin responses (SSRs). In 59 patients (59%) elevation of plantar temperature was demonstrated at the end of the surgical procedure. In this group, plantar sweating was found to be exacerbated in three patients (5%); plantar sweating was improved in 52 patients (88.1%); and no change was demonstrated in four patients (6.8%). In the other group of patients in whom no temperature change occurred, increased plantar sweating was demonstrated in three patients (7.3%); plantar sweating was improved in 20 patients (48.8%); and no change was shown in 18 patients (43.9%). The difference between temperature and sweating change was significant (p = 0.001). Compared with the presympathectomy rate, the rate of absent SSR also significantly increased after sympathectomy: from 20 to 76% after electrical stimulation and 36 to 64% after deep inspiration stimulation, respectively (p < 0.05). Conclusions. In contrast to compensatory sweating in other parts of the body after T2–3 sympathetomy, improvement in plantar sweating was shown in 72% and worsened symptoms in 6% of patients. The intraoperative plantar skin temperature change and perioperative SSR demonstrated a correlation between these changes.

ABSTRACTThe expression of many antigens, stimulatory molecules, or even metabolic pathways in mycobacteria such asMycobacterium bovisBCG orM. smegmatiswas made possible through the development of shuttle vectors, and several recombinant vaccines have been constructed. However, gene expression in any of these systems relied mostly on the selection of natural promoters expected to provide the required level of expression by trial and error. To establish a systematic selection of promoters with a range of strengths, we generated a library of mutagenized promoters through error-prone PCR of the strong PL5promoter, originally from mycobacteriophage L5. These promoters were cloned upstream of the enhanced green fluorescent protein reporter gene, and recombinantM. smegmatisbacteria exhibiting a wide range of fluorescence levels were identified. A set of promoters was selected and identified as having high (pJK-F8), intermediate (pJK-B7, pJK-E6, pJK-D6), or low (pJK-C1) promoter strengths in bothM. smegmatisandM. bovisBCG. The sequencing of the promoter region demonstrated that it was extensively modified (6 to 11%) in all of the plasmids selected. To test the functionality of the system, two different expression vectors were demonstrated to allow corresponding expression levels of theSchistosoma mansoniantigen Sm29 in BCG. The approach used here can be used to adjust expression levels for synthetic and/or systems biology studies or for vaccine development to maximize the immune response.


Significance Developed economies have implemented fiscal support measures worth over 16% of their GDP to combat the pandemic's impacts. To pay off the resulting debt and to ensure sufficient revenue in the face of a fragile and uneven recovery, new taxes and higher rates are being considered. Impacts Levying a wealth tax on productive firms risks starving them of capital for the future. If levied too high or too long, people would sell assets to pay, depressing asset prices, reducing investment and prompting outflows. Criticisms of wealth taxes mean that a one-off surcharge on investment income above a certain threshold would be easier and more popular.

PurposeThe vulnerability of worn-out textures in the face of natural disasters is one of the most significant challenges that have forced planners and urban managers to intervene in these structures. In this context, the new-urbanism, or the new urbanization, movement is one of the most novel approaches. This paper aims to organizing one of the worn-out neighborhoods in Rudsar, northern Iran with an approach toward the new urbanism.Design/methodology/approachThe procedure adopted in this research is a combination of both quantitative and qualitative practices with an applied approach. Therefore, through utilizing the analytical hierarchy process (AHP) and space syntax methods, the principles of the new urbanism are examined using the Expert Choice and Depthmap software. Subsequently, the appropriate priorities are mentioned for organizing the neighborhood with the new-urbanism approach using the building information system (BIM) and strengths, weaknesses, opportunities and threats (SWOT) techniques.FindingsThe findings of this study indicate that among the main characteristics of the new urbanism, walkability is recognized as the most important factor. Afterward, the components of maintaining the traditional structure of neighborhoods and connectivity were in the second and third ranks, respectively, of importance. Accordingly, by identifying the societal potential of roads according to their spatial configuration, it is possible to boost walkability, as well as economic prosperity in these areas.Originality/valueThe combination and correlation of the four utilized methods in this research can be adopted in the future studies as a new outlook of the mixed methods in the field of urban studies.

Purpose The purpose of this paper is to establish the place of the printed book in the era of technological advancement with the assumption that the print media is facing imminent death in the face of readily available and convenient online information. Also the paper aims to assess how the development of new technologies have affected the production, circulation and readership of the printed book, especially among the young generation. Design/methodology/approach Explanatory study was used with closed-ended approach to collect data from 50 students of the Technical University of Kenya and 5 key informant interviews with selected book publishers in Nairobi. The uses and gratification theory was used to explore the knowledge-seeking behavior among the respondents. Findings Findings showed that more than 80% of the respondents preferred the internet to the printed book, which, according to them, has no future in the face of technological advancement. Book publishers, on the other hand, felt that the printed book has a bright future among specific audiences who are committed to it, and especially those in the rural areas who have no access to the internet. While they agreed that the internet has posed a major challenge to the sales and readership of the printed book significantly, it is helping in marketing the printed book as opposed to killing it. New bookshops in Nairobi and modern libraries in high schools, tertiary institutions and universities demonstrate that the printed book is not dying soon. Research limitations/implications The researcher experienced challenges in data collection as the respondents were busy preparing for final examinations and hence many of them were not willing to spare time to fill the questionnaire. To solve this, the researcher had to spend more time to collect data as opposed to if the students were free and ready to participate in the study without any pressure. Practical implications The findings can be used as a basis for further research to widen the scope that can help bring a wider perspective to the topic. The results can also inform policy guidelines on the topic and also contribute to the body of knowledge. Social implications The topic touches on social phenomena that are affecting a number of young people and their information-seeking habits in the era of digital revolution. The way the young generation seek and use information should be of interest not only to academic staff but also to policymakers. Originality/value The paper is original based on primary data that was collected by the researcher from the respondents. It is backed by secondary data to bridge the gap between theory and practice.

ABSTRACTStaphylococcus aureusis a major cause of morbidity and mortality worldwide.S. aureuscolonizes 20 to 80% of humans at any one time and causes a variety of illnesses. Strains that are resistant to common antibiotics further complicate management.S. aureusvaccine development has been unsuccessful so far, largely due to the incomplete understanding of the mechanisms of protection against this pathogen. Here, we studied the role of different aspects of adaptive immunity induced by anS. aureusvaccine in protection againstS. aureusbacteremia, dermonecrosis, skin abscess, and gastrointestinal (GI) colonization. We show that, depending on the challenge model, the contributions of vaccine-inducedS. aureus-specific antibody and Th1 and Th17 responses to protection are different: antibodies play a major role in reducing mortality duringS. aureusbacteremia, whereas Th1 or Th17 responses are essential for prevention ofS. aureusskin abscesses and the clearance of bacteria from the GI tract. Both antibody- and T-cell-mediated mechanisms contribute to prevention ofS. aureusdermonecrosis. Engagement of all three immune pathways results in the most robust protection under each pathological condition. Therefore, our results suggest that eliciting multipronged humoral and cellular responses toS. aureusantigens may be critical to achieve effective and comprehensive immune defense against this pathogen.IMPORTANCES. aureusis a leading cause of healthcare- and community-associated bacterial infections.S. aureuscauses various illnesses, including bacteremia, meningitis, endocarditis, pneumonia, osteomyelitis, sepsis, and skin and soft tissue infections.S. aureuscolonizes between 20 and 80% of humans; carriers are at increased risk for infection and transmission to others. The spread of multidrug-resistant strains limits antibiotic treatment options. Vaccine development againstS. aureushas been unsuccessful to date, likely due to an inadequate understanding about the mechanisms of immune defense against this pathogen. The significance of our work is in illustrating the necessity of generating multipronged B-cell, Th1-, and Th17-mediated responses toS. aureusantigens in conferring enhanced and broad protection againstS. aureusinvasive infection, skin and soft tissue infection, and mucosal colonization. Our work thus, provides important insights for future vaccine development against this pathogen.

Purpose Coastal and floodplain areas are on the frontline of climate change in Bangladesh. Small-scale coastal and floodplain fishing communities of the country face a host of cross-scale stressors continually, some induced by climate change, and they have developed coping and adaption strategies based on customary social and experiential learnings. This paper aims to examine the coping and adaptation strategies that small-scale fishing communities undertake in the face of stresses including climate change and variability. Design/methodology/approach This research takes a nuanced ethnographic-oriented approach based on around two-year-long field study in two coastal and floodplain fishing villages, represented by two distinct ethnic groups. The study adopts direct observational methods to denote the ways small-scale fishing communities address the arrays of stressors to construct and reconstruct their survival and livelihood needs. Findings It was observed that fishers’ coping and adaptation strategies comprise a fluid combination of complex overlapping sets of actions that the households undertake based on their capitals and capabilities, perceptions, socio-cultural embeddedness and experiential learnings from earlier adverse situations. Broadly, these are survival, economic, physiological, social, institutional and religiosity-psychological in nature. Adaptation mechanisms involve some implicit principles or self-provisioning actions that households are compelled to do or choose under given sets of abnormal stresses to reach certain levels of livelihood functions. Originality/value Based on empirical field research, this paper recognizes small-scale fishers’ capability and adaptability in addressing climate change-induced stresses. Policymakers, international development planners, climate scientists and social workers can learn from these grassroots-level coping and adaptation strategies of fishing communities to minimize the adverse effects of climate change and variations.

Purpose – The purpose of this paper is to show, in a practical manner, that discursive procedures (mediated by online platforms or publication systems) enhance cultural understanding of distinct social entities such as Europe and Russia. Design/methodology/approach – Joint special issues convening authors from east and west in an equilibrated manner give an opportunity for in-depth review processes, in the course of which different paradigms of global evolution are perceived and reflected. Findings – If handled with care, information technologies are able to strongly support such stepwise consensus finding. The areas of futurology, forward looking, global megatrends, global evolution, globalistics and global studies are prime cases for the patient alignment of world views and multi-paradigmatic approach towards (ultimately) holistic and second-order science. Originality/value – Even in the face of diverse political realities a series of conferences, lectures and web-supported exercises for the authoring of articles are able to consistently merge actors in academia and policy consulting.
