The Role of Sample Size to Attain Statistically Comparable Groups – A Required Data Preprocessing Step to Estimate Causal Effects With Observational Data

Propensity score methods provide data preprocessing tools to remove selection bias and attain statistically comparable groups – the first requirement when attempting to estimate causal effects with observational data. Although guidelines exist on how to remove selection bias when groups in comparison are large, not much is known on how to proceed when one of the groups in comparison, for example, a treated group, is particularly small, or when the study also includes lots of observed covariates (relative to the treated group’s sample size). This article investigates whether propensity score methods can help us to remove selection bias in studies with small treated groups and large amount of observed covariates. We perform a series of simulation studies to study factors such as sample size ratio of control to treated units, number of observed covariates and initial imbalances in observed covariates between the groups of units in comparison, that is, selection bias. The results demonstrate that selection bias can be removed with small treated samples, but under different conditions than in studies with large treated samples. For example, a study design with 10 observed covariates and eight treated units will require the control group to be at least 10 times larger than the treated group, whereas a study with 500 treated units will require at least, only, two times bigger control group. To confirm the usefulness of simulation study results for practice, we carry out an empirical evaluation with real data. The study provides insights for practice and directions for future research.

Abstract Background Propensity score methods are increasingly being used in the infectious diseases literature to estimate causal effects from observational data. However, there remains a general gap in understanding among clinicians on how to critically review observational studies that have incorporated these analytic techniques. Methods Using a cohort of 4967 unique patients with Enterobacterales bloodstream infections, we sought to answer the question “Does transitioning patients with gram-negative bloodstream infections from intravenous to oral therapy impact 30-day mortality?” We conducted separate analyses using traditional multivariable logistic regression, propensity score matching, propensity score inverse probability of treatment weighting, and propensity score stratification using this clinical question as a case study to guide the reader through (1) the pros and cons of each approach, (2) the general steps of each approach, and (3) the interpretation of the results of each approach. Results 2161 patients met eligibility criteria with 876 (41%) transitioned to oral therapy while 1285 (59%) remained on intravenous therapy. After repeating the analysis using the 4 aforementioned methods, we found that the odds ratios were broadly similar, ranging from 0.84–0.95. However, there were some relevant differences between the interpretations of the findings of each approach. Conclusions Propensity score analysis is overall a more favorable approach than traditional regression analysis when estimating causal effects using observational data. However, as with all analytic methods using observational data, residual confounding will remain; only variables that are measured can be accounted for. Moreover, propensity score analysis does not compensate for poor study design or questionable data accuracy.

Aims: It is common in the context of evaluations that participants have not been selected on the basis of transparent participation criteria, and researchers and evaluators many times have to make do with observational data to estimate effects of job training programs and similar interventions. The techniques developed by researchers in such endeavours are useful not only to researchers narrowly focused on evaluations, but also to social and population science more generally, as observational data overwhelmingly are the norm, and the endogeneity challenges encountered in the estimation of causal effects with such data are not trivial. The aim of this article is to illustrate how register data can be used strategically to evaluate programs and interventions and to estimate causal effects of participation in these. Methods: We use propensity score matching on pretreatment-period variables to derive a synthetic control group, and we use this group as a comparison to estimate the employment-treatment effect of participation in a large job-training program. Results: We find the effect of treatment to be small and positive but transient. Conclusions: Our method reveals a strong regression to the mean effect, extremely easy to interpret as a treatment effect had a less advanced design been used (e.g. a within-subjects panel data analysis), and illustrates one of the unique advantages of using population register data for research purposes.

Selection bias is a problem for mathematics education researchers interested in using observational rather than experimental data to make causal inferences about the effects of different instructional methods in mathematics on student outcomes. Propensity score methods represent 1 approach to dealing with such selection bias. This article describes general principles underlying propensity score methods and illustrates their application to mathematics education research using 2 examples investigating the impact of problem-solving emphasis in mathematics classrooms on students' subsequent mathematics achievement and course taking. Limitations of the method are discussed.

The anti-diabetic effect of ethanolic and aqueous extracts of Imperata cylindrical  rhizomes was investigated in alloxan-induced diabeties in rats. Diabetes was induced by a single 150 mg/kg intraperitoneal dose of alloxan. Rats were divided into five groups with six rats in each group i.e. the normal control group, diabetic control group, standard group (glibenclamide, 10mg/kg, p.o.), Test-I group (200 mg/kg ethanolic extract) and Test-II group (200 mg/kg aqueous extract). The above concerned groups were inoculated on 21st day. On the last day of the experiment, fasted rats were killed by cervical dislocation. The body weight was measured at the initial day and final day. The blood samples were collected for estimation of glucose. The loss of body weight in control group, but recovery was observed in drug treated group. The serum glucose level was significant increased in diabetic rats. However, significant improvement was observed in treated group. The biochemical parameters such as HDL and proteins level were decreased in the control group but maintained in drug treated group. LDL, cholesterol, triglyceride creatinine and urea were significant increase in control group however, reduced level in drug treated group. The present study concluded that ethanolic and aqueous extracts of I. cylindrical  rhizome showed an appreciable effect in reducing the hyperglycemia and the complications associated with diabetes. However, aqueous extract is found more significant in decreasing blood glucose level in comparison to the ethanolic extract. The study results justify the traditional use of the plant as anti-diabetic.

PURPOSE: To study the effect of alcoholism on intestinal healing and postoperative complications in rats METHODS: One hundred and sixty rats were divided into two groups: control and treated. The control group received water and the treated group 30% ethanol. After 180 days, colotomy with anastomosis were performed. After, the groups were divided into four subgroups: 20 rats for study at the following moments: 4th, 7th, 14th and 21st postoperative. The analyzed parameters were: weight gain, breaking strength, tissue hydroxyproline, postoperative complications and histopathological study RESULTS: Weight gain was greater in the control group (p<0.05). When all the subgroups were clustered, breaking strength was significantly greater in the control (p<0.05). Histopathology and hydroxyproline dosage did not show differences. There were five surgical site infections in the treated group while the control group showed two (p>0.05). Nine fistulas occurred in the treated group whereas the control group two (p<0.05). There were three deaths in the control group and seven in the treated group (p>0.05). CONCLUSIONS: Treated group undergo a malnutrition process that is revealed by lower weight gain. Impaired intestinal healing as indicated by smaller breaking strength. There were a larger number of postoperative complications in the treated animals.

Background and Aim: Recently, many medicinal plants have received considerable attention in the medical field because of their role in the wound healing potential. This study aimed to determine the effectiveness of H. tuberosus powder on the healing pathway of full-thickness cutaneous wounds in a mouse model. Materials and Methods: H. tuberosus powder was prepared by a freeze-drying process using a lyophilizer and its active ingredients were evaluated by high-performance liquid chromatography (HPLC), while its antibacterial properties were evaluated by agar well diffusion assay. The percentage wound contraction was also assessed. Thirty mice were used, which were divided equally into two groups, a control group and a treated group. A full-thickness wound, 1 cm×1 cm in size, was established on the dorsal aspect of the thoracolumbar region, into which H. tuberosus powder was topically applied in the treated group. In contrast, the control group was left without any treatment. The animals were euthanized on days 7, 14, and 21 after wounding for histopathological study. Results: The agar well diffusion method indicated the antibacterial activity of H. tuberosus, while the HPLC results indicated that the active ingredients of H. tuberosus powder mainly consisted of three major kinds of fatty acid. In addition, the macroscopic results of wound contraction rate and the histopathological outcomes of the healing process were significantly (p≤0.05) enhanced in the treated group compared with those in the control group. Conclusion: H. tuberosus powder acts as an antibacterial agent with the ability to enhance the wound healing process.
