Manililla is a mango variety whose residues contain bioactive compounds such as polyphenols and flavonoids, with high added value. The use of environmentally friendly extraction technology would be of great relevance; hence, this study aimed to evaluate the effect of solvent relation, sonication time and amplitude on the ultrasound-assisted extraction of total polyphenols in Manililla mango residues (peel, endocarp and kernel) and antioxidant activity. An experimental design 23 with a central point was used to evaluate the curvature behavior of the process variables. Conventional maceration was used as a control. The better conditions were obtained at the central point using 50% ethanol in water, 60% amplitude and 20 min of sonication time. We obtained values of up to 1814 mg GAE/100 g, 469 mg GAE/100 g and 672 mg GAE/100 g of total polyphenols and 1228 mg QE/100 g, 653 mg QE/100 g and 880 mg QE/100 g of total flavonoids for peel, endocarp and kernel, respectively. Mangiferin was quantified in ultrasound-assisted extraction at 150 mg/g in peel and 0.025 mg/g in the kernel, but it was not detectable in maceration. An antioxidant capacity of 87%, 14% and 83% inhibition for peel, endocarp and kernel, respectively, were obtained. Peel and kernel were the residues with higher potential as extraction material, while endocarp was not.

Chlorogenic acid has been proved to have cardiovascular protection, antibacterial, antiviral, hemostatic, and hypolipidemia effects. Modern scientific research on the bioactivity of chlorogenic acid has been extended to the fields of food, medicine, health care and daily-use chemical industry. The aim of this research was to optimize the extraction conditions for chlorogenic acid from Eucommia ulmoides (Eucommiaceae) leaves. The significant variables were screened and optimized by a combination of Plackett-Burman test and Box-Behnken design. Optimum extraction parameters with ethanol concentration of 50%, solvent pH value of 3, and particle size of 60 mesh were determined according to variance analysis and contour plots. Under these conditions, the yield of chlorogenic acid was up to 4.36 mg/g, which was basically consistent with the theoretical prediction value of 4.50 mg/g. This study also proved the potential antioxidant activity of E. ulmoides leaves. The optimal extract of E. ulmoides leaves rich in chlorogenic acid showed the highest antioxidant activity in the FRAP method, which was 219.8 μM Trolox equivalents (TE) per g extract weight (EW) (μM TE/g EW). The DPPH method gave a similar value (168 μM TE/g EW) to the ABTS method (152 μM TE/g EW). The established extraction process was efficient in the recovery of chlorogenic acid from E. ulmoides leaves, encouraging its valorization as a cheap and sustainable alternative for the isolation of chlorogenic acid.

AbstractThe total content of polyphenols and flavonoids determined in the same plant and their corresponding antioxidant activities may vary widely, depending on the extraction conditions applied. This study was conducted to optimise the extraction conditions of phenolics and flavonoids from the mistletoe plant. Various extraction methods, i.e. ultrasound-assisted extraction technology, maceration, maceration with stirring, accelerated solvent extraction (ASE), and extraction under reflux were evaluated for their percentage extraction of polyphenols (TPC) and flavonoids (TFC) from Viscum album subsp. abietis. In addition, the anti-radical activity of extracts was analysed using the 2,2-diphenyl-1-picrylhydrazyl method. The effects of temperature, solvent type, and concentration on the phenolic extraction efficiency and antioxidant activity were studied using chemometric and statistical methods. The results showed that the extracts of V. album subsp. abietis contained large amounts of polyphenols and flavonoids (up to 57.673 mg g−1 and 9.955 mg g−1 of dry extract, respectively) and exhibited potent antioxidant activity, hence representing promising sources of powerful antioxidants. Due to its high extraction efficiency and considerable saving of time and solvent, ASE was more effective than the other extraction techniques. Extracts prepared with water-polar solvent mixtures displayed the highest TPC, TFC, and antioxidant activity, while organic polar solvents were the least efficient extractants.

The use of green marine seaweed Ulva spp. as foods, feed supplements, and functional ingredients has gained increasing interest. Microwave-assisted extraction technology was employed to improve the extraction yield and composition of Ulva pertusa polysaccharides. The antioxidant activity of ulvan was also evaluated. The impacts of four independent variables, i.e., extraction time (X1, 30 to 60 min), power (X2, 500 to 700 W), water-to-raw-material ratio (X3, 40 to 70), and pH (X4, 5 to 7) were evaluated. The chemical structure of different polysaccharides fractions was investigated via FT-IR and the determination of their antioxidant activities. A response surface methodology based on a Box–Behnken design (BBD) was used to optimize the extraction conditions as follows: extraction time of 43.63 min, power level of 600 W, water-to-raw-material ratio of 55.45, pH of 6.57, and maximum yield of 41.91%, with a desired value of 0.381. Ulvan exerted a strong antioxidant effect against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and showed reducing power in vitro. Ulvan protected RAW 264.7 cells against H2O2-induced oxidative stress by upregulating the expression and enhancing the activity of oxidative enzymes such as superoxide dismutase (SOD) and superoxide dismutase (CAT). The results suggest that the polysaccharides from U. pertusa might be promising bioactive compounds for commercial use.

The interest in berries from food manufacturers is huge, which is explained by good organoleptic characteristics and high biological activity of berries. One way to preserve the properties of berries throughout the year is to obtain extracts. Determining the conditions and type of optimal extraction technology to obtain biologically active substances from plant materials is an important stage in the production of new natural preventive additives. The aim of this work is to study the chemical composition and antioxidant activity of blueberry extracts (Vaccinium myrtillus L.) obtained by various methods: maceration, ultrasonic and microwave extraction. The use of ultrasonic extraction as a method of intensifying the process of extracting biologically active compounds from blueberries is very effective. This proves an almost 2-fold increase in the flavonoid content in the extracts. However, the content of anthocyanins in the extracts of blueberries is not the highest for ultrasonic extraction (a decrease by 3.3 times compared with maceration), in all likelihood due to the instability of this class of compounds in this type of processing. Among the extracts of blueberries obtained by three technologies (maceration, microwave and ultrasound treatment), it is the ultrasound extract that has the highest antioxidant activity (9.5±0.1 mg/cm3, 18.18 mmol±0.24 Fe2+/1 kg, 58.6%), determined by three methods: DPPH method, FRAP method, method for assessing antioxidant properties using a model system with linoleic acid. Ultrasonic extraction was chosen as a technology for obtaining a concentrated blueberry extract.

In this study, pomegranate peel as a traditional natural remedy was extracted and encapsulated in proniosomal systems in order to improve its stability against harsh environmental conditions. Pomegranate peel was extracted by using sonication as a green extraction technology and the antioxidant activity of the obtained extract was evaluated to be 85.37% by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals. Proniosomal powder was prepared based on the slurry method with a mixture of non-ionic surfactants namely span 60 and tween 20 in combination with cholesterol as a bilayer stabilizer. Proniosome-derived niosomes were achieved by hydration of the powder with water. The obtained vesicles were evaluated for their particle size, morphological observations, entrapment efficiency, cytotoxicity assay, DPPH antioxidant activity and, physical stability at 4 °C for 28 days. The results demonstrated that the proniosome-derived niosomes were of small size (198.16 nm for unloaded and 411.3 for extract loaded), quite homogeneous (PDI = 0.188 for unloaded and 0.216 for loaded) with highly negative charged spherical vesicles and showed appropriate physical stability during the time of storage. The encapsulation efficiency was 68.43±0.24% and the cytotoxicity assay proved that the formulations were not toxic against 3T3 fibroblast cells in the applied concentration.
