SUMMARYThe current trend of antimony (Sb) unresponsiveness in the Indian subcontinent is a major impediment to effective chemotherapy of visceral leishmaniasis (VL). Although contributory mechanisms studied in laboratory-raised Sb-R parasites include an up-regulation of drug efflux pumps and increased thiols, their role in clinical isolates is not yet substantiated. Accordingly, our objectives were to study the contributory role of thiols in the generation of Sb unresponsiveness in clinical isolates. Promastigotes were isolated from VL patients who were either Sb responsive (n=2) or unresponsive (n=3). Levels of thiols as measured by HPLC and flow cytometry showed higher basal levels of thiols and a faster rate of thiol regeneration in Sb unresponsive strains as compared with sensitive strains. The effects of antimony on generation of reactive oxygen species (ROS) in normal and thiol-depleted conditions as also their H2O2 scavenging activity indicated that in unresponsive parasites, Sb-mediated ROS generation was curtailed, which could be reversed by depletion of thiols and was accompanied by a higher H2O2 scavenging activity. Higher levels of thiols in Sb-unresponsive field isolates from patients with VL protect parasites from Sb-mediated oxidative stress, thereby contributing to the antimony resistance phenotype.

A novel strategy for the synthesis of zinc thioporphyrazine nanospheres is presented. The uniform spheres were fabricated through a polymerization reaction without using any template or emulsifier, which result from the covalent links between zinc tetra(2,3-bis(pentenylthio)porphyrazine (ZnPz) possessing eight long flexible chains in periphery and each with one terminal olefin group. A possible mechanism to explain the self-assembly of ZnPz nanospheres was also illuminated through monitoring nanosphere formation by means of transmission electron microscopy (TEM). More importantly, ZnPz nanospheres can effectively activate dioxygen in aqueous media under simulated sunlight irradiation, and showed significantly improved photocatalytic activity for the degradation of Rhodamine B (RhB) as compared to monomeric ZnPz. Furthermore, the reactive oxygen species derived from ZnPz nanospheres under light irradiation were determined by electron paramagnetic resonance (EPR) technology, indicating that singlet oxygen ([Formula: see text]O[Formula: see text] and hydroxyl radical (•OH) are the major reactive oxygen species. The facile synthetic methodology and the enhanced photocatalytic performance of the ZnPz nanospheres endow this novel material with the potential of being an efficient biomimetic photocatalyst for wastewater treatment.

Jixueteng, the dried stem of Spatholobus suberectus Dunn (Leguminosae), is a traditional Chinese herbal medicine that is commonly classified as a herb that promotes blood circulation and can be used to treat blood stasis. The aim of this study was to examine the reactive oxygen species (ROS) scavenging activity of Jixueteng and other herbal medicines. The ROS scavenging activities of the water extracts of Jixueteng, Cnidium officinale and Salvia miltiorrhiza were examined using an electron spin resonance (ESR) technique and faint luminescence measurement. The ESR signal intensities of the superoxide anion (O2•-) and hydroxyl radical (HO•) were reduced more by Jixueteng than the other herbal medicines we tested. High photon emission intensity to hydrogen peroxide (H2O2) and HO• was observed in Jixueteng using the XYZ chemiluminescence system that was used as faint luminescence measurement and analysis. The results of the present study revealed that the ROS scavenging activity of 8% Jixueteng was the strongest among the herbal medicines we tested. It has been reported that Jixueteng includes various polyphenols. In the ROS scavenging activity by Jixueteng, it is supposed that the antioxidant activity caused by these polyphenols would contribute greatly. In conclusion, a water extract component of Jixueteng had potent free radical scavenging activity and an antioxidative effect that inhibited the oxidative actions of O2•-, H2O2 and HO•. Therefore, Jixueteng represents a promising therapeutic drug for reactive oxygen-associated pathologies.
