Use of in vitro assays to assess the potential antigenotoxic and cytotoxic effects of saffron (Crocus sativus L.)

The present study aimed to estimate and compare, using in vitro assays, the antioxidant activities of alcoholic extracts of saffron stigma and spath as by-products of the saffron crop. The antioxidant activity was evaluated using in vitro assays such as total antioxidant capacity, DPPH free radical, Scavenging of H2O2, FRAP and β-carotene bleaching assay. The results allowed us to evidence the presence of products with antioxidant and bioactive capacities in spaths and stigmas of Crocus sativus. The response of each extract depends on the type of the test. Extract from spaths protect β-carotene from bleaching more than the extract of stigmas and extract from stigmas revealed a significant reducing power activity than that of spaths. As a result, we can consider these extracts especially that of spaths like a new source of antioxidant compounds.

Thalidomide is an old well-known drug that is still of clinical interest, despite its teratogenic activities, due to its antiangiogenic and immunomodulatory properties. Therefore, efforts to design safer and effective thalidomide analogs are continually ongoing. Research studies on thalidomide analogs have revealed that the phthalimide ring system is an essential pharmacophoric fragment; thus, many phthalimidic compounds have been synthesized and evaluated as anticancer drug candidates. In this study, a panel of selected in vitro assays, performed on a small series of phthalimide derivatives, allowed us to characterize compound 2k as a good anticancer agent, acting on A2058 melanoma cell line, which causes cell death by apoptosis due to its capability to inhibit tubulin polymerization. The obtained data were confirmed by in silico assays. No cytotoxic effects on normal cells have been detected for this compound that proves to be a valid candidate for further investigations to achieve new insights on possible mechanism of action of this class of compounds as anticancer drugs.

O3 dissolved in water (or ozonized water) has been considered a potent antimicrobial agent, and this study aimed to test this through microbiological and in vitro assays. The stability of O3 was accessed following modifications of the physicochemical parameters of water, such as the temperature and pH, with or without buffering. Three concentrations of O3 (0.4, 0.6, and 0.8 ppm) dissolved in water were tested against different microorganisms, and an analysis of the cytotoxic effects was also conducted using the human ear fibroblast cell line (Hfib). Under the physicochemical conditions of 4 °C and pH 5, O3 remained the most stable and concentrated compared to pH 7 and water at 25 °C. Exposure to ozonized water resulted in high mortality rates for Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Enterococcus faecalis, and Candida albicans. Scanning electron micrograph images indicate that the effects on osmotic stability due to cell wall lysis might be one of the killing mechanisms of ozonized water. The biocidal agent was biocompatible and presented no cytotoxic effect against Hfib cells. Therefore, due to its cytocompatibility and biocidal action, ozonized water can be considered a viable alternative for microbial control, being possible, for example, its use in disinfection processes.

Background:: Tumors of the childhood are considered to be grave and devastating pathologies, with high mortality rates. Current therapeutic options like cytotoxic drugs and radiotherapy target both healthy and malignant cells, thus resulting in long-term neurological and intellectual sequelae and endocrinological disorders. Objectives:: In this study, we focused on the anticancer potency of crocins, the main constituents of Crocus sativus L, stigmas. Crocins were first extracted using organic solvents from the dried stigmas and then were identified using the HPLC analysis. Materials and Methods:: TE-671 cells were treated with the extract of crocins using a range of concentrations between 0.25-mg/ mL and 16 mg/mL. Viability of the cells was measured at 24h, 48h, 72h and 96h. In addition, we have examined the expression levels of the p53 gene using Real-Time Reverse Transcription PCR. Results:: Results showed that crocins exerted significant cytotoxic and anti-proliferative effects in a concentration and time - dependent-manner on TE-671 cells. Furthermore, p53 manifested similar expression pattern as the anti-proliferative effect of crocin. Conclusion:: Our data demonstrate that crocins could be a novel promising agent for the improvement of tumor treatment.

Crocus sativus L. is a male sterile vegetatively propagated plant. Its flower produces stigmas that when dried, constitute the source of a spice commonly known as Saffron. Slow vegetative propagation and diseases limit the production and the development of saffron. “In vitro” culture could be an effective method to overcome these limitations by improving the quantity and the quality of the planting materials. In this work, Crocus sativus L. segments corms of cultivar from the region of Taliouine (Southeast of Morocco) were used for the propagation through indirect organogenesis. To optimize the in vitro growth conditions, we have used the Murashige and Skoog medium (MS medium), supplemented with 2.4-dichlorophenoxyacetic acid (2.4-D) and with 6-benzylaminopurin (BAP) at combination of various concentrations. Our results showed the formation of callus in 85.42% of explants that grow in a culture medium supplemented with 2,4-D combined with BAP, at a concentration of 1mg/l each. In addition, we observed that increasing the concentration of BAP in the culture medium to 1.5mg/l improved the rate of shoots initiation (0.81). In the meantime, we noted that a combination of BAP (8mg/l) and Naphthalene acetic acid (NAA; 2mg/l) has significantly improved the rate of the formation of advanced shoots (6.65). Finally, the shoots that developed were transferred to an induction medium of roots and corms. As a result, we observed that 50% of shoots tested in ½ MS medium supplemented with 2.4-D and of BAP (1 mg/l each) and 5% sucrose, formed corms. Our study provides a first database for in vitro culture of Moroccan saffron cultivars.
