Background: Enhancing a compound’s biological activity is the central task for lead optimization in small molecules drug discovery. However, it is laborious to perform many iterative rounds of compound synthesis and bioactivity tests. To address the issue, it is highly demanding to develop high quality in silico bioactivity prediction approaches, to prioritize such more active compound derivatives and reduce the trial-and-error process. Methods: Two kinds of bioactivity prediction models based on a large-scale structure-activity relationship (SAR) database were constructed. The first one is based on the similarity of substituents and realized by matched molecular pair analysis, including SA, SA_BR, SR, and SR_BR. The second one is based on SAR transferability and realized by matched molecular series analysis, including Single MMS pair, Full MMS series, and Multi single MMS pairs. Moreover, we also defined the application domain of models by using the distance-based threshold. Results: Among seven individual models, Multi single MMS pairs bioactivity prediction model showed the best performance (R2 = 0.828, MAE = 0.406, RMSE = 0.591), and the baseline model (SA) produced the most lower prediction accuracy (R2 = 0.798, MAE = 0.446, RMSE = 0.637). The predictive accuracy could further be improved by consensus modeling (R2 = 0.842, MAE = 0.397 and RMSE = 0.563). Conclusion: An accurate prediction model for bioactivity was built with a consensus method, which was superior to all individual models. Our model should be a valuable tool for lead optimization.

Women’s rugby (rugby league, rugby union and rugby sevens) has recently grown in participation and professionalisation. There is under-representation of women-only cohorts within applied sport science and medicine research and within the women’s rugby evidence base. The aims of this article are: Part 1: to undertake a systematic-scoping review of the applied sport science and medicine of women’s rugby, and Part 2: to develop a consensus statement on future research priorities. This article will be designed in two parts: Part 1: a systematic-scoping review, and Part 2: a three-round Delphi consensus method. For Part 1, systematic searches of three electronic databases (PubMed (MEDLINE), Scopus, SPORTDiscus (EBSCOhost)) will be performed from the earliest record. These databases will be searched to identify any sport science and medicine themed studies within women’s rugby. The Preferred Reporting Items for Systematic Reviews and Meta-analyses extension for Scoping Reviews will be adhered to. Part 2 involves a three-round Delphi consensus method to identify future research priorities. Identified experts in women’s rugby will be provided with overall findings from Part 1 to inform decision-making. Participants will then be asked to provide a list of research priority areas. Over the three rounds, priority areas achieving consensus (≥70% agreement) will be identified. This study has received institutional ethical approval. When complete, the manuscript will be submitted for publication in a peer-reviewed journal. The findings of this article will have relevance for a wide range of stakeholders in women’s rugby, including policymakers and governing bodies.

Objective.Current clinical trial designs for pharmacologic interventions in rheumatoid arthritis (RA) do not reflect the innovations in RA diagnosis, treatment, and care in countries where new drugs are most often used. The objective of this project was to recommend revised entry criteria and other study design features for RA clinical trials.Methods.Recommendations were developed using a modified nominal group consensus method. Canadian Rheumatology Research Consortium (CRRC) members were polled to rank the greatest challenges to clinical trial recruitment in their practices. Initial recommendations were developed by an expert panel of rheumatology trialists and other experts. A scoping study methodology was then used to examine the evidence available to support or refute each initial recommendation. The potential influence of CRRC recommendations on primary outcomes in future trials was examined. Recommendations were finalized using a consensus process.Results.Recommendations for clinical trial inclusion criteria addressed measures of disease activity [Disease Activity Score 28 using erythrocyte sedimentation rate (DAS28-ESR) > 3.2 PLUS ≥ 3 tender joints using 28-joint count (TJC28) PLUS ≥ 3 swollen joint (SJC28) OR C-reactive protein (CRP) or ESR > upper limit of normal PLUS ≥ 3 TJC28 PLUS ≥ 3 SJC28], functional classification, disease classification and duration, and concomitant RA treatments. Additional recommendations regarding study design addressed rescue strategies and longterm extension.Conclusion.There is an urgent need to modify clinical trial inclusion criteria and other study design features to better reflect the current characteristics of people living with RA in the countries where the new drugs will be used.

University students who sought hormonal emergency contraception (EC) in the Tampere Student Health Station during the period 1.9.2000-31.12.2001 received a questionnaire on their use of it. Of the total, 114 responded (67%). The aim of this study was to ascertain why the respondents did not want to get pregnant now and when it would be suitable for them to get children. Free answers to these questions were analysed using a collective consensus method. The main reasons for not wanting pregnancy now were un? nished studies and the non-steady character of the relationship. Almost all planned to become pregnant some day in the future when their life situation was appropriate, usually at the age of about 30 years.

Introduction: Many studies have illustrated climate change effects on social variables and health. This study aimed to identify the SDH associated with climate change. Methods: The present study is a review study with a systematic search. The keywords related to climate change and social variables were searched in the Web of Science and PubMed databases until June 2018. In the initial search 12097 articles were obtained and after the elimination of duplicate and non-related articles, 5932 articles remained After studying the abstracts, 342 articles were excluded based on entry and exit criteria (studies that were not related to climate change and SDH) and 43 articles remained. In the next phase, the full text of the articles was evaluated by two evaluators individually and the consensus method was used. 23 papers were finally included in the study. Results: According to the review, social variables related to climate change were divided into 5 different categories of variables, including structural variables related to climate change (socioeconomic status), variables related to social status and work conditions (access to health services, unemployment, immigration, inequality, education, work condition, food security), variables related to social relationships and social networks (social movements, urban warfare, riot, group protests, interpersonal violence), individual variables related to lifestyle (place of living; city/village), and individual variables (age, race, gender) based on social determinants of health. Conclusions: Climate change has a wide range of social outcomes.. Various groups that are vulnerable to climate change include women, elderlies, children, villagers, and workers.

In this paper we propose an integrated approach in order to increase the precision of feature point matching. Many different algorithms have been developed as to optimizing the short-baseline image matching while because of illumination differences and viewpoints changes, wide-baseline image matching is so difficult to handle. Fortunately, the recent developments in the automatic extraction of local invariant features make wide-baseline image matching possible. The matching algorithms which are based on local feature similarity principle, using feature descriptor as to establish correspondence between feature point sets. To date, the most remarkable descriptor is the scale-invariant feature transform (SIFT) descriptor , which is invariant to image rotation and scale, and it remains robust across a substantial range of affine distortion, presence of noise, and changes in illumination. The epipolar constraint based on RANSAC (random sample consensus) method is a conventional model for mismatch elimination, particularly in computer vision. Because only the distance from the epipolar line is considered, there are a few false matches in the selected matching results based on epipolar geometry and RANSAC. Aguilariu et al. proposed Graph Transformation Matching (GTM) algorithm to remove outliers which has some difficulties when the mismatched points surrounded by the same local neighbor structure. In this study to overcome these limitations, which mentioned above, a new three step matching scheme is presented where the SIFT algorithm is used to obtain initial corresponding point sets. In the second step, in order to reduce the outliers, RANSAC algorithm is applied. Finally, to remove the remained mismatches, based on the adjacent K-NN graph, the GTM is implemented. Four different close range image datasets with changes in viewpoint are utilized to evaluate the performance of the proposed method and the experimental results indicate its robustness and capability.


<em>Abstract</em>.—Stream fish are expected to be influenced by climate change as they are ectothermic animals living in lotic systems. Using fish presence–absence records in 1,110 stream sites across France, our study aimed at (1) modeling current and future distributions of 35 stream fish species, (2) using an ensemble forecasting approach (i.e., several general circulation models [GCM] × greenhouse gas emission scenarios [GES] × statistical species distribution models [SDM] combinations) to quantify the variability in the future fish species distribution due to each component, and (3) assessing the potential impacts of climate change on fish species distribution and assemblage structure by using a consensus method that accounted for the variability in future projections.

“Health Consensus for the Assessment of Chronic Care Programs” (HC-ACP) is an internet based application created to promote and facilitate the participation of health professionals in the definition of a set of indicators for the assessment of chronic care and management of areas of improvement in this field. The first prototype of the application has been applied twice, first in the region of Catalonia, and in a second project in the context of the whole Spanish Health System. HC-ACP has collected contributions from more than 800 health professionals from around Spain including profiles in the fields of management, health care professional, health planning and quality assessment, allowing sharing and aggregate knowledge and clinical experience from a wide range of points of view. After a process of literature review and panel meetings with professionals who proposed a wide list of indicators, the HC-ACP application was used to select a minimum set of indicators following a participative process based on Health Consensus, an online Real Time Delphi method. The first part of this chapter is devoted to expose paradigms that define the interdisciplinary research field of the method, the second part of the chapter presents the Health Consensus method, and finally the third part exposes a detailed description of the HC-ACP application and the followed process. Besides the relevance and utility of the Health Consensus method, the action-research conducted to build the application proves the efficiency and effectiveness of getting health professionals really involved in the processes of defining the models to assess the healthcare system. The online method proposed has been accepted by participants who have expressed high levels of satisfaction during the participation process.
