EXTRA-LARGE TRIBOLIUM CONFUSUM: A NEW AUTOSOMAL RECESSIVE MUTANT

A new mutant of Tribolium confusum Jacquelin duVal (Coleoptera: Tenebrionidae), extra-large (designated xl), was isolated in mating competition tests with red-eye (re) and wild-type (+). Crosses showed that it was autosomal recessive gene with subvital effects. The pupal weights averaged 6.1 and 7.3 mg for males and females, respectively, about twice the weights of the ancestral wild-type. The generation time (egg to adult) was approximately 8 to 9 weeks compared with about 4 weeks for the wild-type. This increase resulted from a lengthening of the larval stage since the durations of the egg and pupal stages were within the ranges of the wild-type. Mean longevity of xl males and females was reduced to 8.5 and 6.0 weeks, respectively at 26.7 ± 1 °C and 60% RH.

AbstractThe radiosensitivity of a wild-type (+/+) laboratory strain of Tribolium confusum Jacquelin duVal was compared with that of an extra-large (xl/xl) strain known to differ in only a single locus. Differences in both longevity and fecundity between the two strains after irradiation were very highly significant (p < 0.001), but real differences between strains in these biological characteristics may have produced the differential response. When differences in longevity and fecundity were corrected for the unirradiated control, there were only minor differences in response between the two strains. Thus, physical size alone apparently did not greatly influence the comparative radiosensitivity for the two strains of this species.

Often, investigators disregard the sex ratio when selecting test insects for exposure. Demonstrated differences in susceptibility between male and female insects require evaluation of the sex ratio in studies on insecticidal effectiveness.

Fucosidosis is a rare lysosomal storage disorder characterized by deficiency of α-L-fucosidase with an autosomal recessive mode of inheritance. Here, we describe a 4-year-old Chinese boy with signs and symptoms of fucosidosis but his parents were phenotypically normal. Whole exome sequencing (WES) identified a novel homozygous single nucleotide deletion (c.82delG) in the exon 1 of the FUCA1 gene. This mutation will lead to a frameshift which will result in the formation of a truncated FUCA1 protein (p.Val28Cysfs*105) of 132 amino acids approximately one-third the size of the wild type FUCA1 protein (466 amino acids). Both parents were carrying the mutation in a heterozygous state. This study expands the mutational spectrum of the FUCA1 gene associated with fucosidosis and emphasises the benefits of WES for accurate and timely clinical diagnosis of this rare disease.

Biological control has been reducing the use of chemical products against insect pests, specially predatory Pentatomidae. Species of this group can present high variations in their life cycle as a result of their diet. Thus, the objective of this research was to study nymph development and reproduction of Podisus distinctus (Stäl, 1860) (Heteroptera: Pentatomidae) fed on Bombyx mori L., 1758 (Lepidoptera: Bombycidae) larvae (T1), compared to those fed on Tenebrio molitor L., 1758 (Coleoptera: Tenebrionidae) (T2) and Musca domestica L., 1758 (Diptera: Muscidae) larvae (T3) at a temperature of 25 ± 0.5ºC, relative humidity of 70 ± 2%, and photophase of 12 h. Predators fed on B. mori showed duration of the nymph phase (18.68 ± 1.02) similar to those fed on T. molitor (18.32 ± 1.49). Pre-oviposition and oviposition periods and number of egg masses, besides eggs and nymphs per female, were higher with B. mori (5.83 ± 2.02; 15.00 ± 7.40; 8.42 ± 1.84; 296.69 ± 154.75; and 228.55 ± 141.04, respectively) while longevity of males and females of P. distinctus was 25.76 ± 16.15 and 35.00 ± 16.15 days with T. molitor, and 20.57 ± 13.60 and 23.46 ± 12.35 days with B. mori, respectively.

Loss-of-function mutations in DJ-1 are associated with autosomal recessive early onset Parkinson’s disease (PD), yet the underlying pathogenic mechanism remains elusive. Here we demonstrate that DJ-1 localized to the mitochondria-associated membrane (MAM) both in vitro and in vivo. In fact, DJ-1 physically interacts with and is an essential component of the IP3R3-Grp75-VDAC1 complexes at MAM. Loss of DJ-1 disrupted the IP3R3-Grp75-VDAC1 complex and led to reduced endoplasmic reticulum (ER)-mitochondria association and disturbed function of MAM and mitochondria in vitro. These deficits could be rescued by wild-type DJ-1 but not by the familial PD-associated L166P mutant which had demonstrated reduced interaction with IP3R3-Grp75. Furthermore, DJ-1 ablation disturbed calcium efflux-induced IP3R3 degradation after carbachol treatment and caused IP3R3 accumulation at the MAM in vitro. Importantly, similar deficits in IP3R3-Grp75-VDAC1 complexes and MAM were found in the brain of DJ-1 knockout mice in vivo. The DJ-1 level was reduced in the substantia nigra of sporadic PD patients, which was associated with reduced IP3R3-DJ-1 interaction and ER-mitochondria association. Together, these findings offer insights into the cellular mechanism in the involvement of DJ-1 in the regulation of the integrity and calcium cross-talk between ER and mitochondria and suggests that impaired ER-mitochondria association could contribute to the pathogenesis of PD.

In Neurospora crassa, strains of opposite mating type generally do not form stable heterokaryons because the mating type locus acts as a heterokaryon incompatibility locus. However, when one A and one a strain, having complementing auxotrophic mutants, are placed together on minimal medium, growth may occur, although the growth is generally slow. In this study, escape from such slow growth to that at a wild type or near-wild type rate was observed. The escaped cultures are stable heterokaryons, mostly having lost the mating type allele function from one component nucleus, so that the nuclear types are heterokaryon compatible. Either A or a mating type can be lost. This loss of function has been attributed to deletion since only one nuclear type could be recovered in all heterokaryons except one, but deletion spanning adjacent loci has been directly demonstrated in a minority of cases. Alternatively when one component strain is tol and the other tol+ (tol being a recessive mutant suppressing the heterokaryon incompatibility associated with mating type), escape may occur by the deletion or mutation of tol+, also resulting in heterokaryon compatibility. An induction mechanism for escape is speculated upon.
