Preparation of Polypeptide-Dye Multilayers by an Electrostatic Assembly Process

ABSTRACTTo develop novel optical thin films, we have prepared self-assembled polypeptide films by an electrostatic process. The films were placed on a glass slide previously silanized by an amino silane and given a positive charge by immersion in aqueous acid. Subsequent immersion of the slide in aqueous anionic solutions of either poly(L-glutamic acid), congo red, copper phthalocyanine tetrasulfonic acid or p-nitroaniline-modified poly(L-glutamic acid) resulted in deposition of the anions on the surface. Following anionic immersion, the slides were dipped into a cationic poly(L-lysine) solution. Alternate dipping into anionic and cationic solutions yielded multilayers. The thin films were characterized by optical absorption and circular dichroism. The optical density increased with dipping cycles. Circular dichroism measurements of the thin films showed induced dichroism of the congo red and phthalocyanine-containing films, suggesting formation of a locally ordered dye-polypeptide complex. Solution circular dichroism measurements of the polypeptides indicated a coil conformation, while poly(Lglutamic acid)/poly(L-lysine) complexes showed circular dichroism spectrum characteristic of a β-sheet.

AbstractSoft-x-ray magnetic circular dichroism (MCD) is the difference between the absorptivity or reflectivity of left and right circularly polarized soft-x-rays at the magnetically interesting L2,3- edges of 3d transition metals or the M4,5-edges of the 4f rare earth elements. Thanks to its large absorption cross-section and strong MCD effect, this technique has become a powerful new means for probing, in an element- and site-specific manner, the magnetic properties of ultra-thin films and multilayers. Soft-x-ray MCD experiments, recently conducted at the Dragon beamline, are utilized to demonstrate the recent progress in this technique and its applications in the research of magnetic thin films.

The polarization properties responsible for the classical "green birefringence" of the amyloid-Congo red complex have been determined by a new optical method, phase modulation microspectrophotometry. This method now makes possible the measurement of one optical property at a time (birefringence, optical rotation, linear dichroism and circular dichroism throughout the visible spectrum) in complex specimens in which visible contrast in polarized light is the result of a mixture of polarization effects. The green birefringence is explained by a combination of optical effects, the strongest of which are dispersion of birefringence and linear dichroism superimposed on the smaller effects of circular dichroism and optical rotatory dispersion. The interaction of the planar dye molecules with the amyloid protein induces an extrinsic Cotton effect.
