This study was carried out to investigate the effects of light spectra, additional UV-A, and different growth regulators on the in vitro germination of Senecio cineraria DC. Seeds were surface-sterilized and inoculated in MS medium to evaluate the following light spectra: white, white plus UV-A, blue, green, red or darkness. The maximum germinability was obtained using MS0 medium under white light (30%) and MS + 0.3 mg L-1 GA3 in the absence of light (30.5%). S. cineraria seeds were indifferent to light. Blue and green lights inhibited germination. Different concentrations of gibberellic acid (GA3) (0.1; 0.4; 0.6; 0.8; 1.0 and 2.0 mg L-1) and indole-3-acetic acid IAA (0.1; 0.3 and 1.0 mg L-1) were evaluated under white light and darkness. No concentration of GA3 enhanced seed germination percentage under white light. However, when the seeds were maintained in darkness, GA3 improved germination responses in all tested concentrations, except at 1.0 mg L-1. Under white light, these concentrations also increased the germination time and reduced germination rate. Germination rate, under light or darkness, was lower using IAA compared with GA3.

Asymbiotic seed germination of Eulophia nuda Lindl. was observed on Knudson C medium. About 90% seeds germinated within 8-10 weeks and formed green protocorms in 11-12 weeks. Effect of BA and IBA was studied on plantlet development from protocorms. BA shows the best results with respect to number and length of shoots. Maximum number (6.45±1.36) and length (3.90±0.99) was observed on MS medium supplemented with 4.44μM.BA. Maximum root growth was also observed on same medium (4.8±0.99 number of roots and 1.43±0.13cm length). The regenerated plantlets were successfully acclimatized and transferred to earthen pots. The results presented here show that in vitro seed germination and plantlet development in Eulophia nuda Lindl., an endangered orchid, can be achieved at a higher rate by this method.Int J Appl Sci Biotechnol, Vol 3(2): 243-247 DOI: http://dx.doi.org/10.3126/ijasbt.v3i2.12476

The experiment was designed to evaluate the effect of growth regulators on leaf explant of Gerbera for callus induction. Various kinds of plant growth regulators such as 6-Benzylaminopurine (BAP), α-Naphthalene acetic acid (NAA), 2, 4-Dichlorophenoxyacetic acid (2, 4-D), Indole-3-acetic acid (IAA) were used to initiate cultures. These were added to Murashige and Skoog medium in different combinations and concentrations. Leaf explants cultured on MS medium supplemented with BAP+ 2, 4-D+ IAA in T4 treatment & BAP+ 2,4-D in T5 treatment showed the best results for callus induction. On the other hand callus was induced early in the combination of BA+ 2,4-D + IAA hormone in T5, T9 & T8 treatment respectively. The rate of callus induction was very low in BA + NAA combinations but it was much earlier.

Peppermint (Mentha piperita), sweet basil (Ocimum basilicum), and coriander (Coriandrum sativum) are important medicinal plants in the pharmacological industry. These plants are produced in commercial scale but their seeds exhibit low germination percentages under favorable germination conditions. Enhancing seed germination is thus crucial for improving the production of these plants. The influence of gibberellic acid (GA3), indole-3-acetic acid (IAA), indol-3-butyric acid (IBA), and naphthalene acetic acid (NAA) on seed germination of the three plants were investigated. The seeds were soaked in each plant growth regulator at 50, 100, and 150 mg·L−1 for 24 hours at 25 ± 2 °C. Seed germination was checked daily for 20 days and germination parameters including final germination percentage (FGP), corrected germination rate (CGRI), and number of days lapsed to reach 50% of FGP (GT50) were recorded. The phosphorus and protein contents were determined in germinated seedlings on day 21 of culture. All plant growth regulators enhanced seed germination as compared with control. However, GA3 improved seed germination more than IAA, IBA, and NAA. GA3 at 100 mg·L−1 significantly increased the FGP from 22.3% and 33.3% (control) to 74% and 65.6% for peppermint and sweet basil, respectively. Low concentration of GA3 at 50 mg·L−1 increased the FGP for coriander from 27% to 52.3%. GA3 also increased CGRI, GT50, phosphorus, and protein contents in germinated seedlings as compared with control. Seeds of peppermint, sweet basil, and coriander possess a physiological dormancy that could be elevated by GA3 presowing treatment. This study established a successful methodology for optimizing seed germination to satisfy the demand for the medicinal parts of these plants in the pharmacological industry.

Scarification treatments (a control, a 10-minute vacuum, or a 1.5-minute ultrasound), different media (modified Norstog and Van Waes) and growth regulators [benzyladenine (BA) at 0, 1, 1.5, or 2 mg·L-1 and 6-(r,r-dimethylallylamino)-purine riboside (2iPR) at 0, 1, 1.5 or 2 mg·L-1] were used in combination to increase seed germination of Cypripedium calceolus var. parviflorum. Seeds treated with ultrasound had higher germination (58.0%) than those treated with vacuum (27.4%) or controls (19.2%). Germination rates increased with 2iPR level and reached a maximum between 1.5 and 2 mg·L-1. Seeds on Van Waes medium, which were not transferred to fresh medium after germination, had a severe browning problem causing many protocorms to die. Those on Norstog medium continued to grow into seedlings with less browning. Germination rates of Calopogon tuberosus × Calopogon `Adventure' and Liparis liliifolia were determined on the different media and growth regulator treatments. Multiple shoots of Calopogon developed from single seeds on media containing growth regulators. Flower buds formed in vitro on Calopogon in media containing 1 mg·L-1 or higher BA 5 months after germination. L. Iiliifolia seeds in Norstog medium had a higher proportion of germination than those in Van Waes medium.

The present work was carried out to investigate the effects of gamma radiation on regeneration of Capsicum annuum L. var Kulai via in vitro. Seeds of C. annuum were irradiated with various doses of gamma ray (0, 20, 40, 60, 80, 100, 200, 300, 400, 500, and 600 Gy) emitted from the Caesium-137 source at the rate of 4.31 Gy per minute. Irradiated seeds grown on MS medium without hormone for hypocotyl and cotyledon preparation as explant for in vitro regeneration. Seed germination rate revealed significant variation between treatments, and seeds started to germinate between 6 to 17 days. Irradiated seeds between 0-60 Gy were observed to germinate in less than 10 days. All explants including hypocotyl and cotyledon were cultured on MS medium with different concentrations of BAP in combination with AgNO3 to observe the response of these explants to different hormone concentrations. From the observation, calluses were induced in 90% of hypocotyl and cotyledon explants in all treatments. The characteristics of calluses were varied with greenish friable, greenish compact, yellowish watery, yellowish friable and yellowish compact. In other treatments, calluses were found in purple, bright yellow and yellowish orange. On the other hand, shoot regeneration was observed in treatment between 40-100 Gy. In conclusion, gamma radiation gave impact on seed germination, seedling growth performance, in vitro callus formation and shoot regeneration of Capsicum annuum var. Kulai

The in vitro seed germination which results in the production of disease-free seedlings and greenhouse germination of the seeds of Mansonia altissima was investigated in order to establish a better way of germination of the timber species. Five levels of GA3 treatment were used in in vitro germination with three replicate and two seeds were inoculated in each of the jam bottle. Whereas, in greenhouse germination, five levels of different treatments were used, replicated three times and each Petri plate contained 15 seeds. The experiment was repeated twice and the data from each experiment was put together and used for the statistical analysis. The results showed that seeds germination occurred eight days after inoculation in in vitro but in the case of greenhouse germination, it took only five days. For in vitro rapid germination of Mansonia altissima, the MS medium should be supplemented with 1.0 μm of GA3. Equally, in greenhouse germination, the seeds need to be soaked in 1.0 mM of GA3 for 24 hours. Alternatively, in the absence of GA3, the seeds can be soaked in water for 24 hours before broadcasting the seeds on the seedbed for germination, as this will help to identify nonviable seeds.

ABSTRACT The Brazilian guava (Psidium guineense Swartz) is seed-propagated and, being native to the Caatinga biome, may frequently have uneven germination.Thus, we aimed to evaluate the synchronization of the in vitro seed germination of three accessions of the Brazilian guava, using water, polyethyleneglycol (PEG 6000), and potassium nitrate (KNO3) at different potentials and times of osmotic priming. Seeds from three accessions of the Brazilian guava (Y85, Y93,and Y97) from the UNEB/BA Germplasm Active Bank were subjected to the following pretreatments: -0.6, -1.0, -1.4, and -1,8 MPa PEG 6000; 10 and 20% KNO3 for 24h; 10 and 20% KNO3 for 48h; water for 24 and 48h; and non-primed seeds as the control. The experimental design was therefore a 10x3+1 factorial scheme. We assessed the germination percentage (G), mean germination time (MGT), germination speed (GS), and germination speed index (GSI). Data was subjected to analysis of variance followed by a means test (Duncan at 5% probability) and regression. There was interaction between the priming treatments and accessions for all evaluated features, except G. PEG 6000 decreased the MGT (from 6 to 8 days) and increased GS and GSI of seeds from all three accessions at potentials -1.0 to -1.5 MPa.Water-priming had a positive effect on MGT, GS, and GSI of accession Y85 seeds. KNO3 negatively affected germination of seeds from all three accessions. Thereby, we could synchronize seed germination of accessions Y85 and Y97 with PEG 6000.

Pholidota articulata is an epiphytic orchid mostly used in ornamental cut/pot flower and in traditional medicine. As it has high ornamental and medicinal values, its population from natural habitats is decreasing, therefore, it is listed in the Appendix-II of Convention on International Trade in Endangered Species (CITES). The objective of the present study is to obtain the in vitro plants of P. articulata from seed culture to conserve its germplasm. The in vitro seed germination was carried out in different strengths of Murashige and Skoog (MS) and Knudson C (KnC) medium supplemented with various plant hormones. On the half-strength of MS medium, seeds were started to germinate after 4 weeks of primary culture and they were developed into protocorms with first leaf primordium earlier than on the other medium. Therefore, in vitro developed protocorms were sub-cultured on the half-strength of MS medium supplemented with different concentrations of 6-benzylaminopurine (BAP), gibberellic acid (GA3) and α-naphtalene acetic acid (NAA). They were successfully developed into shoots on the 1.5 mg/l BAP supplemented half-strength of MS medium. Later, they were inoculated on the half-strength of MS medium supplemented with different concentration of α-napthalene acetic acid (NAA), indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) for the root formation, where IBA supplemented medium was found effective for the development of roots. Thus, this study provides a reliable protocol for non-symbiotic seed germination and plant production, and reveals that seed-derived protocorms are good explants for the in vitro mass propagation for conservation and sustainable utilization in horticulture.

The experiment was undertaken with an objective to investigate the effect of various concentrations of plant growth regulators, i.e., Gibberellic acid (GA3) and Indole 3-acetic acid (IAA) on seed germination of Macrotyloma uniflorum. Seeds were soaked for 12 hours in different concentrations substances (1.0, 2.0, 3.0 and 4.0 mg/L) of IAA, GA3 and control set was soaked only in distilled water.  Three replicates of each treatment with fifty seeds per replicate were arranged for precise physiological analysis. Significant variation was found in all aspects after analysis of variance (ANOVA) of each mean value. After two weeks of seed soaking, it was noted that germination percentages were significantly accelerated by lower concentrations (1 and 2 mg) of used hormones. Amongst the two potential growth regulators, 2 mg/L was found most effective because it showed highest germination percentage for IAA (93%) and GA3 (88%). A great deal of information relating to seed germination practices shows that these plant growth regulators were efficient in overcoming dormancy leading to rapid seed germination. IAA was selected as best hormone in this study, which showed highest seed germination (93%).

Abstract Background: In Ethiopia, species used for timber production are limited to few species. So, it is becoming necessary to find innovative ways of maintaining and improving the genetic quality of species such as Milicia excelsa by domesticating them into a wider scale of ecological and social environment. Hence, the main objective of this study was to investigate the seed germination biology and behavior of Milicia excelsa under in-vitro laboratory conditions so as to promote the domestication and establishment of seed production areas (SPA) in-situ. Methods: Seeds collected from Benchi district of Benchi-Maji zone of southwestern Ethiopia were subjected to germination biology in-vitro to determine the germination responses of M. excelsa seeds under laboratory conditions. So, a total of 1200 seeds were randomly selected, treated and prepared using four treatments procedures with six replications for paper method and 3 replications for sand method, and tested using paper methods in CRD design. So, 600 seeds were tested using paper and sand methods each where 25 seeds were tested for each replication in paper and 50 seeds for each replication in sand method. The germination process was lasted between 19 March 2020 and 15 May 2020. Result: The mean germination percentage of seeds of M. excelsa was noticed to be lower than 20%. This is below the expected (45%) germination rate of mature and healthy seed lot. Overall, seeds that were not treated using any pre-sowing treatments has shown relatively better germination percentage (19%) followed by seeds rubbed by hand (15%). Analysis of one-way t-test shows that the effects untreated (control –T1) and rubbing by hand treatments (T4) is significant at p < 0.05 compared to the effects of the other pre-sowing treatments (T2- seeds only washed with normal tap water, and T3- seeds washed and soaked in hot water for 15 minutes) on the germination capability of seeds of M. excelsa . However, no significance difference was observed between control (T1) and hand scarification (T2). Conclusions: The average germination of M. excelsa seeds combined for all treatments is lower than 20% which is lower germination when comparing with other similar research reported in other countries. Therefore, it required to include and test other pre-sowing treatments including tissue culture micro-propagation of seeds to maximize the germination of M. excelsa seeds helping to get enough seedlings required for the domestication project.
