LncRNA CASC2 Plays an Indicative Role in Diagnosing Bladder Cancer

Abstract Background: Bladder cancer is a common cancer of urinary system, with high incidence and mortality. LncRNA CASC2 as a tumor suppressor has been reported to be involved in many human tumors. In this study, we aimed to explore the diagnostic value of CASC2 for bladder cancer patients.Methods: qRT-PCR was used to detect the expression level of CASC2 in 140 bladder cancer patients and 90 healthy volunteers. The differences of CASC2 expression between the cancer group and healthy group were analyzed using student’s t test. The correlation of CASC2 expression with clinical characteristics of the bladder cancer patients was estimated with Chi-square test. In addition, ROC curve was plotted to evaluate the diagnostic value of CASC2 for bladder cancer patients.Results: Serum CASC2 level was lower in bladder cancer patients than that in healthy group (P<0.05). The expression level of CASC2 was significantly associated with histological grade (P=0.000), TNM stage (P=0.000), and lymph node metastasis (P=0.001). The area under the ROC curve (AUC) was 0.864 and the optimal cutoff value was 0.955, suggesting the diagnostic value of CASC2 for bladder cancer. The diagnostic sensitivity was 77.8% and specificity was 85.7%.Conclusion: CASC2 may be a novel biomarker for early diagnosis of bladder cancer.

Abstract Background MiR-221 has been identified to play an important role in tumorigenesis and progression. In the present study, we aimed at to investigate the expression pattern of serum miR-221 and evaluate its diagnostic value in cervical cancer. Methods Quantitative real-time polymerase chain reaction (qRT-PCR) was used to measure the expression pattern of miR-221 in cervical cancer patients and healthy controls. The association of miR-221 with clinicopathological data was analyzed with χ2 test. Then receiver operating characteristic (ROC) curve was built to evaluate the diagnostic value of serum miR-221 by calculating the area under the ROC curve (AUC). Results The results indicated that the miR-221 expression level was statistically elevated in cervical cancer patients compared with healthy individuals. The increased miR-221 expression was significantly associated with lymph node metastasis (P = 0.026) and FIGO stage (P = 0.028). ROC curve suggested that serum miR-221 had a high diagnostic value in differentiating cervical cancer patients from healthy controls with AUC of 0.932 (95%CI: 0.903–0.960) corresponding with sensitivity of 77.6% and specificity of 94.8%. Conclusions Taken together, the expression level of miR-221 is increased in cervical carcinoma and it may serve as a promoting bio-marker in the diagnosis of cervical cancer patients.

Abstract Background: Present study aimed to estimate the diagnostic efficacy of urine Metastasis-associated in colon cancer 1 (MACC1) for bladder cancer patients.Methods: Urine MACC1 expression was measured in bladder cancer patients and controls via quantitative real-time polymerase chain reaction (qRT-PCR) assay. Chi-square test was used to assess the correlation of urine MACC1 levels with clinicopathological features of the patients. The diagnostic performance of urine MACC1 was evaluated by receiver operating characteristics (ROC) curve.Results: Urine MACC1 was obviously elevated in bladder cancer patients compared with controls (P<0.001). What’s more, abnormal urine MACC1 expression was significantly correlated with differentiation (P=0.013), tumor stage (P=0.023), lymph node metastasis (P=0.033) and distant metastasis (P=0.026). Furthermore, according to ROC analysis, urine MACC1 could distinguish bladder cancer patients from controls with an AUC (the areas under the ROC curve) of 0.909. The cut-off value of urine MACC1 for bladder cancer diagnosis was 1.275, with 79.8% specificity and 87.3% sensitivity.Conclusion: Urine MACC1 may be act as a biomarkers for diagnosis of bladder cancer.

Abstract Background: Lung cancer is one of the most common causes of cancer death among all the malignancies worldwide. Evidences suggest that the incidence and mortality of lung cancer has been on the rise. MicroRNA-449a (miR-449a) as one important member of microRNAs, has been demonstrated acting as a tumor suppressor in lung cancer. In this study, we sought to assess the relationship between miR-449a expression level and diagnostic value of lung cancer.Methods: In this present research, quantitative Real-Time PCR was applied to detect the miR-449a expression in 116 lung cancer patients and 41 healthy volunteers. The diagnostic value of miR-449a in lung cancer patients was determined by receiver operating characteristic (ROC) curve.Results: MiR-449a was significantly down-regulated in lung cancer patients compared with healthy control (P<0.05). In addition, miR-449a expression was associated with sex (P=0.004), tumor size (P=0.000), TNM stage (P=0.006) and metastasis (P=0.036). However, there was no correlation with age, smoking history and histological type of lung cancer patients (all P>0.05). In the ROC analysis, the results showed that the area under the ROC curve (AUC) was 0.902 with the sensitivity of 94.8% and specificity of 78.0%, and the optimum cutoff value was 2.255.Conclusion: MiR-449a expression was down-regulated in lung cancer patients, and it could be an efficient diagnostic biomarker in lung cancer patients.

Abstract Background: Prostate cancer is the the second leading cause of cancer-related death in male. NF-κB inhibitor alpha gene (NFKBIA), is found dys-expressed in the progression of tumors. The aim of this study was to investigate the expression of NFKBIA and its diagnostic value in prostate cancer patients.Methods: The mRNA expression of NFKBIA was detected with quantitative real-time polymerase chain reaction (qRT-PCR) assay and the association of its expression with clinical characteristic was analyzed via Chi-square test. Receiver operating characteristic (ROC) curve was built to evaluate the diagnostic value of serum NFKBIA.Results: The mRNA expression level of NFKBIA was significantly increased in prostate cancer patients compared with healthy controls (P<0.01). The high expression of NFKBIA was significantly associated with pathological T stage (P=0.010) and differentiation (P=0.019). ROC curve showed that serum NFKBIA had a high diagnostic value in differentiating prostate cancer patients from healthy individuals. The area under the ROC curve (AUC) value was 0.873 corresponding with a sensitivity of 85.6% and a specificity of 78.3%.Conclusions: The expression of NFKBIA was up-regulated in serum of prostate cancer patients and it may be a useful diagnostic bio-maker for the early detection of this cancer.

Abstract Background: NIN1/RPN12 binding protein 1 homolog (NOB1) gene was reported to play a key role in the oncogenesis and prognosis of carcinomas. The aim of the present study was to investigate the expression of NOB1 and its clinical significance in lung cancer, and further explore the diagnostic value of NOB1 in lung cancer patients.Methods: Quantitative real-time polymerase chain reaction (qRT-PCR) analysis was used to characterize the expression of NOB1 in 97 lung cancer patients and 55 healthy controls. The associations of NOB1 mRNA expression with clinicopathological factors of lung cancer patients were analyzed by Chi-square test. The receiver operating characteristics (ROC) curve was used to evaluate the diagnostic value of NOB1 in lung cancer.Results: The expression levels of NOB1 mRNA in lung cancer samples were significantly up-regulated compared with healthy controls (P<0.001). And the NOB1 expression was significant correlated with differentiation (P=0.005) and invasion depth (P=0.008). No associations were found between NOB1 expression and other factors (All P>0.05). ROC curve revealed that the area under ROC curve (AUC) was 0.885, with a sensitivity of 76.3% and a specificity of 89.1%. And the cutoff value of NOB1 was 2.225.Conclusion: NOB1 is a sensitive and specific diagnostic marker for lung cancer, and provide a new therapeutic target for lung cancer treatment.

Abstract Background The purpose of this study was to evaluate the diagnostic and prognostic significance of miR-211-5p in atherosclerosis (AS) by detecting the expression level in serum of patients with AS. Methods A total of 85 healthy controls and 90 asymptomatic AS patients participated in this study. The expression level of miR-211-5p in all subjects were measured by qRT-PCR. Spearman correlation coefficient was used to evaluate the correlation of miR-211-5p with CRP and CIMT. The ROC curve was established to assess the diagnostic value of miR-211-5p in AS. The Kaplan–Meier survival curve and multivariate COX regression analysis were used to evaluate the prognostic significance of miR-211-5p in AS. Results The expression levels of miR-211-5p in AS patients were significantly lower than in healthy controls (P < 0.001), and miR-211-5p showed a significant negative correlation with CRP (r =  − 0.639, P < 0.001) and CIMT (r =  − 0.730, P < 0.001). The AUC of the ROC curve was 0.900, the specificity and the sensitivity were 84.7% and 78.9%, respectively, which indicating that miR-211-5p had diagnostic value for AS. Survival analysis showed that patients with low miR-211-5p expression were more likely to have cardiovascular end-point events (Log rank P = 0.013). Conclusion Serum miR-211-5p could be used as a new biomarker for the diagnosis of AS, and the low expression of miR-211-5p is associated with the poor prognosis of AS.

To explore exosomal miR-375 expression in gastric cancer patients and its relationship with patient prognosis. A total of 53 patients diagnosed with gastric cancer in our hospital from May 2014 to May 2016 were included as the gastric cancer group, and 46 healthy women who came to our hospital for physical examination during the same period were enrolled as the healthy group. Exosomal miR-375 expression level was detected using qRT-PCR, and the diagnostic performance and prognostic significance of exosomal miR-375 in gastric cancer were explored. The gastric cancer group showed increased exosomal miR-375 expression than the healthy group (P< 0.05); Kaplan-Meier survival analysis exhibited that serum exosomal miR-375 has an AUC of 0.778, sensitivity of 69.57%, and specificity of 75.47%, whereas Cox regression analysis showed that the miR-375 expression in exosomes was an independent risk factor affecting the prognosis of gastric cancer patients (P< 0.05). Patient with gastric cancer showed upregulated miR-375 expression in serum exosomes. Serum exosomal miR-375 was found to has positive sensitivity and specificity in the diagnosis of gastric cancer, which may be associated with poor prognosis of gastric cancer patients.

Breast cancer (BC) is one of the most common lethal diseases in women worldwide. Recent evidence has shown that covalently closed Circular RNA (circRNA) deregulation is observed in different human malignancies and cancers. Lately, circRNAs are being considered as a new diagnostic biomarker; however, the mechanism and the correlation of action between circRNAs and BC are still unclear. In the present study, we try to investigate the expression level of hsa_circ_0005046 and hsa_circ_0001791 in BC. By using quantitative real-time polymerase chain reaction (qRT-PCR), expression profiles of candidate circRNAs were detected in 60 BC tissue and paired adjacent normal tissues. Furthermore, the clinicopathological relation and diagnostic value were estimated. Our results showed the higher expression levels of hsa_circ_0005046 and hsa_circ_0001791 in BC tissues compared to paired adjacent normal tissues with P value ( P < 0.0001 ) for both circRNAs, and the area under the receiver operating characteristic (ROC) curve was 0.857 and 1.0, respectively; in addition, a total 10 miRNAs that can be targeted by each candidate circRNAs was predicted base on bioinformatics databases. Taken together, for the first time, the results of our study presented high expression levels of hsa_circ_0005046 and hsa_circ_00017916 in BC; although there was no direct correlation between the high expression level of both circRNAs with clinic pathological factors, except hsa_circ_0001791 association with estrogen receptors (ER), high ROC curve in expressed samples indicated that both circRNAs could be used as a new diagnostic biomarker for BC. Moreover, miRNAs selection tools predicted that miR-215 and mir-383-5p which have a tumor suppressor role in BC can be targeted by our candidate circRNAs to affect the PI3K/AKT pathway; in conclusion, further studies are required to validate the oncogene role of our candidate circRNAs through the PI3k pathway.

Abstract Background Prostate cancer is one of common cancers around the world, and in our country the incidence and mortality of PCa are both increasing. More and more reports have revealed that SOX9 is involved in various human cancers. In this study, we aimed to explore the relationship between SOX9 expression and diagnostic value of PCa patients. Methods In this study, quantitative real-time PCR (qRT-PCR) was performed to determine the expression of SOX9 of the 131 PCa patients and 74 healthy volunteers. And receiver operating characteristic (ROC) curve was used to determine the diagnostic value of SOX9 for PCa patients. Results The results of qRT-PCR showed that the expression of serum SOX9 in PCa patients was higher than that in healthy controls (P < 0.05). And the expression of SOX9 was significantly associated with PSA (P = 0.001), differentiation (P = 0.000), and lymph node metastasis (P = 0.000). Besides, the area under the ROC curve (AUC) was 0.966 with the sensitivity of 93.2% and specificity of 87.8% respectively. The optimal cutoff value of SOX9 was 2.34. Conclusions Our results found that SOX9 is a novel oncogene for PCa, and may be a novel and effective biomarker for the diagnosis of patients with PCa.

Abstract Background Cholangiocarcinoma (CCA) is one of the most aggressive malignancies. Late diagnosis may be responsible for the high mortality. MicroRNA-106b (MiR-106b) is accepted as an important regulator in various human malignancies. The current study was aimed to investigate the diagnostic value of miR-106b in CCA. Methods Serum levels of miR-106b in CCA patients and healthy control were evaluated by quantitative real-time polymerase chain reaction (qRT-PCR). Chi-square test was used to analyze the association of miR-106b with the clinicopathological features. To evaluate the diagnostic value of miR-106b in CCA, the ROC curve was constructed. Results The expression of miR-106b was significantly increased in CCA samples compared with the healthy controls (P < 0.001). The overexpression of miR-106b was remarkable correlated with the lymphatic node metastasis (P = 0.038), clinical stage (P = 0.017) and differentiation (P = 0.009). ROC curve suggested that miR-106b was an effective diagnostic biomarker in CCA with the AUC of 0.913. The optimal cutoff value was 2.525, with the sensitivity of 89.7% and the specificity of 79.3%. Conclusions MiR-106b functions as an oncogene in CCA, which may be an potential diagnostic biomarker for CCA.
