A micropropagation procedure for juvenile and adult savory (Satureja obovata Lag.) explants is described. Pretreatment of the nutlets with gibberellic acid (0.57 mm) did not improve in vitro germination. Optimum shoot proliferation of juvenile and adult material was obtained on medium containing 2.22 μm N6-benzyladenine. Rooting and acclimatization of juvenile shoots were accomplished in vivo, while adult shoots were rooted in vitro after 3 days of exposure to 4.92 μm indole-3-butyric acid followed by subsequent transfer to auxin-free medium. More than 95% survival of adult rooted plants was obtained during the acclimatization phase. Chemical names used: gibberellic acid (GA3); N6-benzyladenine (BA); indole-3-butyric acid (IBA); isopentenyladenine (2iP).

ABSTRACT The anterior, middle, and posterior hypothalamus, the cortex cerebri, the anterior hypophysis as well as the diaphragm of adult ovariectomized rats were incubated in vitro with tritiated 17β-oestradiol. The uptake of tritiated oestradiol was differentially distributed intracerebrally with higher accumulation in the anterior hypothalamus and the hypophysis. Lowering the temperature of the incubation medium caused a reduction in the uptake of radioactivity by the anterior hypothalamus as compared to that found in other brain tissues. Tritiated oestradiol taken up in vitro by the anterior hypothalamus and the hypophysis tended to be retained after further incubation in a steroid-free medium. The addition of non-radioactive 17β-oestradiol to the medium inhibited the uptake of tritiated oestradiol by these tissues. Moreover, pretreatment with non-radioactive 17β-oestradiol in vivo prevented the preferential accumulation of tritiated oestradiol in vitro in the anterior hypothalamus and the hypophysis. These results indicate that oestradiol is preferentially taken up in vitro by the anterior hypothalamus and the hypophysis of the rat.

A previous study by McLean and co-workers reported that regenerating axons of the rabbit vagus nerve were unable to sustain axonal transport in vitro for several months after nerve injury. In contrast, we found that sensory axons of the rat sciatic nerve were able to transport 3H-labeled protein into their regenerating portions distal to the site of injury within a week after injury when placed in vitro. Transport in vitro was not significantly less than transport in axons maintained in vivo for the same period. Transport occurred in the medium that was used by the McLean group, but was significantly reduced in calcium-free medium. When axon regeneration was delared, only small amounts of activity were present in the nerve distal to the site of injury, showing that labeled protein normally present in that part of the nerve was associated with axons and was not a result of local precursor uptake by nonneural elements in the sciatic nerve. We were not able to explain the failure of McLean and co-workers to demonstrate transport in vitro in regenerating vagus nerve, but we conclude that there is no general peculiarity of growing axons that makes them unable to sustain transport in vitro.

The genus Cleome is widely distributed in drier areas of the tropics and subtropics. Cleome dendroides and C. rosea are Brazilian native species that occur mainly in Atlantic Forest and sandy coastal plains, respectively ecosystems negatively affected by human impacts. Cleome spinosa is frequently found in urban areas. Many Cleome species have been used in traditional medicine, as C. spinosa. In the present work, was investigated C. dendroides, C. rosea and C. spinosa germinative behavior under in vivo conditions, as well as was established suitable conditions to in vitro germination and seedling development. The in vivo germination was performed evaluating the influence of temperature, substrate and light. It was observed that only C. spinosa seeds presents physiological dormancy, which was overcome by using alternate temperatures. The substrate influenced significantly the germination of C. rosea and the seeds of C. dendroides showed the highest germination percentages in the different conditions evaluated. The post-seminal development stages under in vivo and in vitro conditions were defined. It was observed that the development was faster under in vitro than in vivo conditions. An effective methodology for in vitro germination, enabling the providing of material to experiment on plant tissue culture was established to C. dendroides and C. spinosa.

A tree species having a unique stem morphology has been found to grow at Sam Poo Kong temple, Gunung Batu, Semarang city, and at Pager Wunung  Darupono Conservation Park, Central Java, about 30 km away from the temple. Based on plant key reference identification the name of this tree species is Bauhinia scandens Willd, a liana from Leguminosae family. This species has a local name of ‘pohon rantai’ due to the chain shape of the stems. It flowered in August and the fruits matured in October. In vitro germination of the seeds collected from the Conservation Park was not successful and only one out of 30 seeds germinated in vivo after 12 weeks. Further studies should be conducted on conservation and propagation of this unique species.

Abstract The present study reports an efficient in vitro propagation system for Turnera ulmifolia using nodal segments as explants. Turnera ulmifolia (Passifloraceae) is an important garden plant with multipotent medicinal values. Effective shoot proliferation was achieved on agar gelled MS medium (Murashige and Skoog, 1962). The maximum number of shoots (8.3 ± 0.57) per initial explant was obtained on MS medium supplemented with 8.88 mM of 6-benzylaminopurine (BAP) and 0.54 mM of α-naphthalene acetic acid (NAA). The highest number of shoots (59.5 ± 2.10) proliferated on semi-solid MS medium (with agar) augmented with 2.22 mM of BAP and 2.32 mM of kinetin (Kin) along with 0.54 mM of NAA. Longer (4-5 cm) and healthy shoots were rooted (12.0 ± 0.10 roots per shoot) on half-strength MS medium fortified with 9.84 mM of indole-3 butyric acid (IBA). The in vitro regenerated plantlets were hardened in the greenhouse and transferred to the field. Significant developmental changes were observed in the foliar micromorphology of in vitro raised plantlets when these were transferred to the field. The stomatal index was gradually reduced (26.72 to 21.25) in the leaves from in vitro to field environments. But, vein-islets and veinlet terminations (13.4 and 7.6) were increased (39.7 and 18.4) respectively from in vitro to in vivo grown plants. Simple, unicellular, less frequent and underdeveloped trichomes were observed with the leaves of in vitro plants but fully developed trichomes recorded in the field transferred plants. The study could help in understanding the response and adaptation of tissue culture raised plantlets towards changed environmental conditions.

Fibres of three cotton cultivars (Gossypium hirsutum H-4, H-8 and G. arboreum G. Cot-15) were analysed for growth in terms of fibre length and dry weight and endogenous gibberellic acid (GA3) content thrice during 1997–2000, at Rajkot. The development of cotton fibre was divided into four distinct growth phases but overlap between elongation and secondary thickening was considerable which suggests that both these phases are independent of each other. During fibre elongation, GA3 content remained low and increased after a decrease in the rate of fibre elongation in all three genotypes. The long staple cultivar (H-4) showed highest endogenous GA3 content followed by the middle one (H-8) and the short staple cultivar (G. Cot-15). In in vitro studies when GA3, NAA or GA3+NAA was supplemented to the media, increase in fibre length of the short staple cultivar was maximum, followed by the middle one and the long staple cultivar. Both in vivo and in vitro findings suggest that GA3 is one of the important factors that determine fibre length.

Experiments were conducted on tissue proliferation (TP) development and in vitro and ex vitro growth of tissues from plants with (TP+) and without TP (TP-). In 1993 the increase in TP in one-, two-, and three-yr-old `Holden' and `Besse Howells' was 3%, 52%. and 32% and 10%, 26% and 21%, respectively. No differential mortality was observed. Shoot tip cultures initated from TP+ and TP- `Montego' showed 10-12 mo were required for miniaturiziation and multiplication in TP- shoot tips and 4 mo in TP+ shoot tips. TP- cultures require 10 uM 2-iP for normal shoot proliferation; whereas TP+ cultures had to be transferred to hormone-free medium after 6 mo to maintain normal shoot morphology. Cutting propagation from TP- and TP+ plants older than 5 yr, showed persistence of morphological aberrations associated with TP+ plants.

Multiple adventitious shoot formed from internodal segments of Cephaelis ipecacuanha cultured 25 days on Gamborg basal medium (GAMBORG et al., 1968) supplemented with 6.66mM 6-benzylaminopurine there was a maximum of nine shoots per segment and an average of five shoots per segment formed. The presence of gibberellic acid in the subculture media promoted shoot elongation in all treatments. The shoots attained 3cm in height and rooting of 100% after 35 days of culturing upon Murashige and Skoog's basal medium (MS), added with 4.92mM indole-3-butyric acid, 0.87m gibberellic acid and 0.1% activated charcoal. Further growth was accelerated after the transfer to 1/2 MS without growth regulators. Rooted plantlets transferred to potting soil could be successfully established.
