The present study aims to optimize the natural deep eutectic solvent (NADES) as a green solvent-based microwave-assisted extraction (MAE) of total polyphenols content (TPC) from Mitragyna speciosa Korth Havil leaves using response surface methodology (RSM). Preparation of natural deep eutectic solvent (NADES) as a green solvent was performed by melting the two-component of malic acid and glucose using a magnetic stirrer. The leaves sample was extracted using the NADES-MAE method with various extraction conditions with four factors and three levels (Box Behnken Design) and optimized using RSM with licensed software of Design Expert V12. TPC was determined using a Folin-Ciocalteau reagent and absorbance was measured on a UV-VIS spectrophotometer at 770 nm and gallic acid as a standard. This study was obtained the optimum extraction conditions include: NADES ratio (malic acid: glucose) of 1:2 g/g, 50% microwave power, a solvent-sampel ratio of 12:1 mL/g, and extraction time for 7 minutes. The optimum conditions were obtained according to the equation formula: TPC = 165.17 – 33.97X1 + 38.36X2 – 6.08X3 + 16.12X4 – 62.77X1X2 + 79.61X12X2, where a R2 value = 0.7102 with a TPC prediction of 402.087±43.81 mg GAE/g sample. These conditions can be used to extract TPC from this plant efficiently, quickly, easily, and environmentally friendly.

This study aims to determine the effect of the use of malic acid-glucose based microwave assited extraction (MAE) methods on total polyphenol extraction from kadamba leaves (Mitragyna Speciosa Korth). Samples that have been dried in the extraction using solvents (green solvent) Malic Acid-Glucose with a ratio of 1: 1 and 1: 3 g/g in several conditions including, microwave strength 50% for 10 minutes with a sample ratio and a solvent of 1:10 g/mL. Then the extract solution obtained was measured in total polyphenol levels using the folin ciocalteu (FC) reagent and the absorbance was measured at a wavelength of 791 nm using a UV-vis spectrophotometer. Based on the results of the study obtained levels of polyphenols from each condition of the solvent used is 258mg GAE/g sample (1:1 g/g solvent) and 292mg GAE /g sample (1: 3 g / g solvent). From this study it can be concluded that the method of malic acid-glucose based microwave assited extraction can be used to extract polyphenol compounds from Kadamba leaves quickly, easily and efficiently.

Kadamba (Mitragyna speciosa Korth Havil) merupakan salah satu tumbuhan endemik di Asia Tenggara, di Indonesia, tumbuhan ini banyak di jumpai di pulau Kalimantan. Tumbuhan ini dipercaya oleh masyarakat dapat mengobati berbagai macam penyakit. Namun, karena kandungan alkaloidnya, tumbuhan ini dilarang digunakan sebagai bahan baku obat herbal oleh Badan Pengawas Obat dan Makanan. Oleh karena itu, perlu dilakukan pengembangan metode ekstraksi untuk menarik senyawa metabolit sekunder target dan meminimalkan senyawa yang tidak diinginkan yaitu dengan menggunakan metode ekstraksi berbantu mikrowave dengan pelarut hijau (Natural Deep Eutectic Solvent) yang dioptimasi menggunakan response surface methodology (RSM). Simplisia kering daun Kadamba diekstraksi menggunakan metode ekstraksi berbantu mikrowave dengan pelarut hijau (asam sitrat-glukosa) dengan berbagai kondisi ekstraksi dengan empat faktor dan tiga level (Box Behnken Design) yang dioptimasi menggunakan RSM dengan aplikasi perangkat lunak Design Expert versi 12 berlisensi. Penetapan kadar polifenol total dilakukan menggunakan reagen Folin-Ciocalteau dan diukur absorbansinya pada spektrofotometer UV-Vis pada panjang gelombang 770 nm, serta asam gallat sebagai standar. Berdasarkan hasil penelitian, diperoleh kondisi optimum pada perbandingan rasio pelarut NADES (asam sitrat : glukosa) 5:1 g/g, rasio pelarut-sampel sebesar 1:20 g/mL, waktu ekstraksi selama 20 menit dan kekuatan gelombang mikro sebesar 30% dengan kadar polifenol total prediksi sebesar 314,924 ±35,95 mg GAE/g sampel, sedangkan dari hasil proses verifikasi (scale-up confirmation) yaitu dengan meningkatkan jumlah sampel yang digunakan sebanyak sepuluh kali lipat dengan Kadar polifenol total diperoleh  sebesar 427,12 ±35,95 mg GAE/g. Penelitian ini merupakan langkah awal dalam pengembangan metode ekstraksi untuk memperoleh senyawa target secara cepat, mudah, efisien, dan ramah lingkungan.

This research was conducted to determine the influence of the use of the choline chloride-sorbitol based microwave assisted extraction (MAE) method against the total extraction of polyphenols from the leaf Kadamba (Mitragyna speciosa Korth). Simplisia extracted using solvent (green solvent) choline chloride-sorbitol with a ratio of 1:1; 1:2 and 1:3 g/g with sample and solvent ratio of 1:10 g/mL, microwave strength is 50% watt and performed for 10 minutes, the extract solution has been obtained and then done determination of total levels of polyphenols by using folin ciocalteu reagents and further measured its absorption with the UV-VIS spectrophotometer device at a wavelength of 770 nm. Based on the results of the study, obtained the levels of polyphenols from each comparison of solvent used against the sample of 334 mg of GAE/g samples (1:1 g/g solvent); 372 mg GAE/g samples (1:2 g/g solvents) and 299 mg of GAE/g samples (1:3 g/g solvents). Based on the results of the study can be concluded that the method of choline chloride-sorbitol based microwave assisted extraction (MAE) can be used to extract the polyphenols from Kadamba leaves easily, quickly and efficiently.

The response surface methodology using the Box-Behnken design was established to describe supercritical carbon dioxide assisted extraction of phyllanthin from Phyllanthus amarus Schum and Thonn leaves prior to HPLC analysis. The effects of extraction pressure, temperature, modifier concentration and extraction time on the yield of phyllanthin were investigated. By solving the regression equation, the optimum conditions were as follows: extraction pressure 23.2 MPa, temperature 40 °C, methanol as modifier at a concentration of 10 % and time 90 min. Under these conditions, the phyllanthin yield was 12.83 ± 0.28 mg g-1, which was in good agreement with the predicted values. Modifier concentration and extraction time showed a significant effect on the phyllanthin yield.

This study aims to determine the effect of citric acid-glucose based microwave assited extraction (MAE) method on total polyphenols content from (Peperomia pellucida [L.] Kunth) herbs. Dried samples were extracted using green solvent glucose citric acid with a ratio of 1: 1 g/g, 3: 1 g/g, and 5: 1 g/g, respectively, 30% microwave power for 5 minutes, and 1:5 g /mL solid-liquid ratio. Then the obtained extract solution was performed determination total polyphenols content using Folin-Ciocalteu reagent, and the absorbance was measured using a UV-Vis spectrophotometer at 791 nm. Based on the results, obtained total polyphenols content according to the extraction condition namely 214.405 mg GAE/g sample (3: 1 g /g solvent), 84.833 mg GAE /g sample (1: 1 g /g solvent) and 53,9451 mg GAE/g sample (5: 1 g /g solvent), respectively. This study is a preliminary data about the use of citric acid-glucose based microwave assisted extraction method on total polyphenols content from this plant quickly, easily and efficiently.

The investigation presented herein had as its scope the development of an integrated process for the efficient extraction of polyphenols from hop. For this purpose, a novel, natural deep eutectic solvent (DES) was synthesised, composed of glycerol and L-alanine, and the process was optimised by deploying a response surface methodology based on a Box–Behnken design. The variables considered were the DES/water proportion, the liquid-to-solid ratio and the stirring speed. Under the optimised conditions, the yield in total polyphenols achieved was 118.97 ± 8.27 mg gallic acid equivalents per g of dry mass. Ultrasonication, incorporated into the process as a pretreatment step, was shown to significantly change the kinetic pattern of polyphenol extraction and contributed to attaining higher yields only at 80 °C, whereas at lower temperatures a supressing effect was observed. Furthermore, increasing temperature was negatively correlated with the second-order extraction rates, evidencing a slow-down of the extraction rate at elevated temperatures.

Objective: Melinjo (Gnetum gnemon L.) seeds are known to contain resveratrol, which are classified as a phenolic compound of the stilbenoid. Melinjoseeds have high water content, making them unstable to be stored for prolonged periods with open packaging at room temperature. The present studyaimed to explore the use of ultrasonic-assisted extraction with a betaine-based natural deep eutectic solvent (NADES) for resveratrol extraction fromirradiated melinjo seeds.Methods: The best betaine-based NADES component was identified among betaine-urea, betaine-lactic acid, and betaine-malic acid. Optimization ofextraction methods was performed using the best NADES and extraction variables such as time of extraction, water percentage, and sample: solventratio. The outcome of extraction was evaluated by measuring resveratrol content using high-performance liquid chromatography and the results wereanalyzed using response surface methodology.Results: The best betaine-based NADES was found to be betaine-lactic acid, yielding a resveratrol content of 0.3344 mg/g powder. The optimumextraction was achieved in 10 min with 60% water and a sample: solvent ratio of 1:10, yielding a resveratrol content of 0.227 mg/g powder.Conclusion: Betaine-based NADES can be purposed as an alternative solvent for resveratrol extraction from irradiated melinjo seeds.

The consumer and cosmetic industries have recently placed a greater emphasis on ecofriendly solvents for botanical extraction, including natural deep eutectic solvents (NADES). In this study, NADES were prepared for Morus alba callus extraction. The efficiency of extraction from the NADES and methanol was investigated by comparison of the stilbenoids yield and anti-melanogenesis activity. Prior to testing the irritability of a suitable NADES on the reconstructed human epidermis (RhE), the effect of the selected NADES on stilbenoids stability was determined. The results showed that the highest yields of stilbenoids were obtained from choline chloride-glycerol mixtures (Ch1G2) and methanol extracts, with no significant difference in yields (5.06 ± 0.05 and 6.32 ± 0.40 mg/g callus dry weight, respectively). The NADES extracts of M. alba callus showed comparable anti-melanogenesis activity compared to methanol. In term of stability, stilbenoids in Ch1G2 remained stable after six months of storage at 4 °C except resveratrol. Furthermore, Ch1G2 had no irritation effect on RhE. Thus, based on the findings of this study, Ch1G2 is an intriguing green solvent alternative for the extraction of M. alba callus and may be advantageous for the preparation of skin-lightening cosmetics.
