THE ACTIVITY OF ESSENTIAL OILS OBTAINED FROM SPECIES AND INTERSPECIES HYBRIDS OF THE Mentha GENUS AGAINST SELECTED PLANT PATHOGENIC BACTERIA

Plant essential oils of six aromatic herb species and interspecies hybrids of the family Lamiaceae – chocolate mint (Mentha piperita × ‘Chocolate’), pineapple mint (Mentha suaveolens ‘Variegata’), apple mint (Mentha × rotundifolia), spearmint (Mentha spicata), orange mint (Mentha × piperita ‘Granada’) and strawberry mint (Mentha × villosa ‘Strawberry’) – were investigated for antimicrobial effects against plant pathogenic bacteria: Agrobacterium tumefaciens, Pseudomonas syringae pv. syringae and Xanthomonas arboricola pv. corylina. The screening was carried out in vitro on agar plates filled with the target organism. All essential oils screened exhibited a higher level of antibacterial activity against A. tumefaciens and X. arboricola pv. corylina than streptomycin used as a standard in all tests. The antimicrobial effect of streptomycin and five mint oils was at the same level for P. syringae pv. syringae. There were no significant differences in the influence of the chocolate mint oil on the growth inhibition of all bacteria tested. Plant essential oils from pineapple mint, apple mint, spearmint and strawberry mint showed the weakest antimicrobial activity against P. syringae pv. syringae and the strongest towards A. tumefaciens and X. arboricola pv. corylina. The essential oils from strawberry mint, pineapple mint, spearmint and apple mint had the strongest effect on A. tumefaciens, and the lowest inhibitory activity was exhibited by the chocolate mint and orange mint essential oils. X. arboricola pv. corylina was the most sensitive to the strawberry mint, pineapple mint and spearmint oils. The chocolate mint oil showed the greatest activity against P. syringae pv. syringae.

Numerous scientific research studies all over the world have addressed the problem of agriculture in the 21st century as being particularly sensitive to climate change, which has caused phytopathogenic bacteria to spread. Therefore, there is a clear and urgent need to contain this kind of risk in agricultural production (both conventional and organic farming). The objective of this study was to determine the antibacterial activity of 30 essential oils (EOs) against three harmful plant pathogenic bacteria of agricultural importance, Erwinia amylovora, Xanthomonas campestris pv. campestris and Pseudomonas syringae pv. syringae. The study included in vitro testing, using an agar-diffusion assay. The EOs of Ceylon cinnamon (leaf and bark), oregano, clove bud and palmarosa revealed antibacterial activity against the test bacteria, and the maximum mean inhibition zone diameters of 35 mm was found against E. amylovora and X. campestris pv. campestris (highly sensitive reaction), while it was smaller in the case of P. syringae pv. syringae, from 18.25-26.25 mm (sensitive to very sensitive reaction). Maximum diameter of the zone of inhibition (35 mm) was obtained using basil and peppermint against E. amylovora, and rosemary, blue gum and camphor tree against X. campestris pv. campestris. Not a single EO inhibited P. syringae pv. syringae with the resulting total diameter zone of 35 mm, and this test bacteria was resultingly classified as the least susceptible bacterium of the three tested. EOs of lemongrass, aniseed, ylang ylang, silver fir, lemon, dwarf mountain pine, bay laurel and scots pine caused sensitive reaction of the tested bacteria. Peppermint, black cumin, Indian frankincense, bergamot orange, common juniper, bitter orange and neem produced variable reactions from total to weakly or no inhibition at all. Weakly activity was found in niaouli and Atlas cedar. Eastern red cedar, patchouli, Indian sandalwood and ginger caused no reaction of any of the test bacteria. The results offer a basis for further work based on in vivo testing for the purpose of developing ?natural pesticides? for control of phytopathogenic bacteria, thus giving a significant contribution to reducing yield losses in agriculture and sustainable development.

Essential oils are volatile substances from plants and many of them have antimicrobial activity. For that reason, they have become known as a useful alternative to chemical preservatives and pesticides. In this study, we tested essential oils of four aromatic plants. Cinnamon (Cinnamomum zeylanicum), thyme (Thymus vulgaris), oregano (Origanum vulgare) and clove (Syzygium aromaticum) essential oils were investigated for their composition and antimicrobial effect against plant pathogenic bacteria (Pectobacterium spp. and Pseudomonas spp.). Both are commonly associated with diseased fruit trees in orchards and gardens. The chemical composition of the tested essential oils was identified by gas chromatography coupled with mass spectrometry. The cinnamon essential oil was most effective form tested oil. The experimental results indicated that the wild strains of tested bacteria are more resistant to essential oils than commonly used laboratory strains. In conclusion, certain essential oils could be used for the control of postharvest bacterial pathogens. The findings of the present study suggest that the essential oils have a potential to be used as antimicrobial agents.

The in vitro effects of plant essential oils (EOs) against pathogenic bacteria are well known, yet few studies have addressed the effects of these compounds against pathogens associated with ready-to-cook foods. Experiments were conducted to determine the effectiveness of oregano and nutmeg EOs on the growth and survival of Yersinia enterocolitica and Listeria monocytogenes in broth culture and in Iranian barbecued chicken. Ready-to-cook Iranian barbecued chicken was prepared according to the common practice with 1, 2, and 3 μl/g of oregano and nutmeg EOs. The test and control (without EOs) samples were inoculated with Y. enterocolitica and L. monocytogenes to a final concentration of 6 to 7 log CFU/g and stored at 3, 8, and 20°C. Microorganisms were counted just before and at 24, 48, and 72 h after storage based on growth on Yersinia selective agar supplemented with cefsulodine, igrasan, and novobiocin and on Listeria selective agar supplemented with nalidixic acid and acriflavin. In the broth culture system, the nutmeg EO had a greater effect on L. monocytogenes (MIC = 0.20 μl/ml) than did the oregano EO (MIC = 0.26 μl/ml). However, the oregano EO had a greater effect on Y. enterocolitica (MIC = 0.16 μl/ml) than did the nutmeg EO (MIC = 0.25 μl/ml). In ready-to-cook Iranian barbecued chicken, the log CFU per gram of both bacteria after up to 72 h of incubation was not decreased significantly by various combinations of oregano and nutmeg EOs (1, 2, and 3 μl/g) and storage temperatures (3, 8, and 20°C) when compared with control samples (without EOs). Although examination of spices in culture media can yield accurate microbiological data, without complementary tests in foods these data are of limited value for assessing food safety.

The motile plant pathogenic bacteria Erwinia carotovora pv. carotovora, Pseudomonas syringae pv. phaseolicola, and Xanthomonas campestris pv. campestris were strongly attracted to conidia of Bipolaris sorokiniana, B. victoriae, and to sclerotia of Macrophomina phaseolina and their exudates in vitro and in soil, but not to phosphate buffer or buffer–soil mixtures. Bacteria accumulated radioactivity within 1 h after being placed in exudates from 14C-labelled conidia of B. sorokiniana. After 5 h, radioactivity of the 14C-labelled exudate was reduced to 29–54% of that in the original medium. Exudates from fungal propagules may act as attractants and substrates for motile plant pathogenic bacteria in soil.

Strawberry is a highly desirable fruit with a unique taste and a good source of bioactive compounds beneficial to human health. However, it has a short post-harvest shelf life, mainly due to the soft rot caused by Rhizopus stolonifer. This study aimed to evaluate the antimicrobial properties of essential oils (EOs) of Mentha piperita, Cymbopogon martinii, Cinnamomum camphora, and Mentha spicata using spore germination and micro-well dilution assays, and to test the effects of the vapor-phase application of M. spicata and C. martini on the incidence and severity of soft rot in strawberry artificially inoculated with R. stolonifer. In in vitro tests, C. martinii and M. spicata EOs were the most effective, inhibiting more than 95% of the spore germination. Additionally, in the microwell dilution test, these EOs had the lowest minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) (5 and 10 μg/mL, respectively), and for the microwell dilution test, the lowest MIC (5 μg/mL and 10 μg/mL, respectively) and MFC (10 μg/mL for both). High in vivo inhibitory effects of M. spicata and C. martinii EOs were observed at 10% concentration, with 100 and 78% reduction, respectively, in the R. stolonifer-induced spoilage. Our results suggest that C. martinii and M. spicata EOs can be used as efficient natural fungicides and can be an alternative to synthetic fungicides for preserving fresh strawberries from soft rot.

AbstractPlant extracts and essential oils (EOs) are characterized by their antibacterial properties against various bacterial pathogens, including staphylococci. Some strains of these bacteria are resistant against the adverse effects of the environment including antibiotics, e. g. methicillin-resistantStaphylococcus aureus(MRSA). EOs alone cannot substitute for antibiotics but their treatment may be useful to intensify and strengthen the effects of antibiotics on pathogenic staphylococci. In this work, we tested the antibacterial effects of the essential oils ofMenthaspecies with menthol as one of the effective substances against different strains ofS. aureus. Twoin vitromethods were used, the qualitative disc diffusion assay and the quantitative minimal inhibitory concentration (MIC) of selected essential oils. Peppermint oil fromMentha piperita, spearmint oil fromMentha spicatavar.crispaand cornmint oil fromMentha arvensiswere tested in this study against the various strains ofStaphylococcus aureus, including methicillin resistantStaphylococcus aureus(MRSA). The oils were dissolved in DMSO (dimethyl sulfoxide) and diluted at the following ratios: 1 : 1, 1 : 2, 1 : 5, and 1 : 10. Based on the results determined by the agar disc diffusion test, the highest antibacterial properties were observed in spearmint oil againstS. aureusCCM 4223 at 1 : 2 ratio where the inhibition zone varied at a range of 35.67 ± 6.81 mm. We determined also the MIC of all the oils where concentrations of the oils were as follows: 1 %; 0.5 %; 0.25 %; 0.125 % and 0.0625 %. The lowest concentrations of essential oils that possessed inhibitory effects on the growth ofS. aureusvaried between 0.125 % and 0.25 %.

Abstract A total of 235 Phytophthora infestans isolates were collected from five regions of the Czech Republic during the growing seasons 2012–2014 and 2016 and examined using the in vitro amended agar method for their sensitivity to metalaxyl-M (MFX), propamocarb-HCl (PCH), and dimethomorph (DMM). A majority of the isolates (50%) were sensitive to MFX. Resistant isolates were found in all four years of the survey; they represented 30% of the samples. The EC50 values of PCH in inhibiting mycelial growth of 65% of the overall isolates were higher than 100 μg ml−1, which indicates the occurrence of insensitivity to PCH in the Czech P. infestans populations. DMM was very effective, and the mycelial growth of all isolates tested was completely suppressed at the concentration of 0.1 μg ml−1. Furthermore, the efficacy of 12 plant essential oils was tested against 20 isolates of P. infestans using the in vitro amended agar method. Essential oils of Cymbopogon winterianus, Litsea cubeba, Mentha spicata, Pelargonium graveolens, Syzygium aromaticum, and Thymus vulgaris were observed to have the highest antifungal activity against P. infestans, with minimal inhibitory concentrations less than or equal to 1 μl ml−1.

Abstract In the framework of the performed studies, the antibacterial activity of the following fungicides was evaluated: Miedzian 50 WG (active substance - a.s. 50% copper oxychloride), Ridomil MZ Gold 68 WG (a.s. 3.8% metalaxyl-M and 64%, mancozeb), Euparen Multi 50 WG (a.s. 50% tolylfluanid), Captan 80 WG [a.s. 80% N-(captan)], Dithane Neotec 75 WG (a.s. 75% mancozeb). The evaluation also concerned the essential oils: lavender, sage, lemon balm, clove, and a preparation based on thyme oil (BioZell). Each preparation and compound was tested against the following bacterial pathogens: Erwinia amylovora, Xanthomonas arboricola pv. corylina, X. arboricola pv. juglandis, Pseudomonas syringae pv. syringae, Agrobacterium tumefaciens (presently Rhizobium radiobacter). Each preparation and compound was tested at a concentration of 1,000 ppm of active substance. Copper oxychloride was also tested at a concentration of 1,500 ppm. Among the tested fungicides, metalaxyl-M with mancozeb, mancozeb alone, and copper oxychloride inhibited all of the tested strains of pathogenic bacteria. Tolylfluanid did not inhibit any of the bacteria used. Out of the investigated essential oils, the strongest inhibitors of bacteria were: sage, cloves, and BioZell. The protective activity of the above mentioned fungicides was also evaluated in vivo. They were assessed against fire blight on apple blossoms and pear fruitlets, against bacterial canker on sweet cherry fruitlets, and against crown gall on sunflower seedlings (the test plant). All fungicides were applied at the same concentrations as those in the in vitro tests. Only copper oxychloride was found to show protective activity against the studied diseases. This result indicates that the antibacterial properties of the other fungicides did not correspond with their activity on the plant organs used in the in vivo experiment.

Five essential oils isolated from plants originating from Serbia and ten combinations of the selected essential oils were assayed to test their inhibitory and fungicidal activity against Trchoderma aggressivum f. europaeum Samuels & W. Gams using two distinctive methods: microdilution and fumigant macrodilution methods. The strongest activity was demonstrated by spearmint (Mentha spicata L.) and thyme (Thymus serpyllum L.) oils at the minimum inhibitory concentration (MIC) of 6.25 ?l ml-1 using microdilution, and 0.16 ?l ml-1 of air using fumigant macrodilution method. The antifungal activity of basil (Ocimum basilicum L.) and peppermint (Mentha piperita L.) was medium, while the oil extracted from St. John?s wort (Hypericum perforatum L.) exhibited the lowest activity. None of the selected essential oils exhibited fungicidal effect at minimal fungicidal concentrations (?FC) of over 25 ?l ml-1 or 0.32 ?l ml-1of air, using micro- and macrodilution, respectively. When microdilution was used, the strongest antifungal activity was demonstrated by two oil combinations: spearmint-thyme and spearmint-peppermint, having MIC and MFC values of 3.75 ?l ml-1. The lowest activity was demonstrated by the basil-St. John?s wort essential oil combination, at 30 ?l ml-1 MIC, and MFC exceeding 30 ?l ml-1. The obtained results indicate possible synergistic effects of essential oils and their components.

ABSTRACTGlucosinolates (GSLs) are secondary metabolites found inBrassicavegetables that confer on them resistance against pests and diseases. Both GSLs and glucosinolate hydrolysis products (GHPs) have shown positive effects in reducing soil pathogens. Information about theirin vitrobiocide effects is scarce, but previous studies have shown sinigrin GSLs and their associated allyl isothiocyanate (AITC) to be soil biocides. The objective of this work was to evaluate the biocide effects of 17 GSLs and GHPs and of leaf methanolic extracts of different GSL-enrichedBrassicacrops on suppressingin vitrogrowth of two bacterial (Xanthomonas campestrispv. campestris andPseudomonas syringaepv. maculicola) and two fungal (AlternariabrassicaeandSclerotiniascletoriorum)Brassicapathogens. GSLs, GHPs, and methanolic leaf extracts inhibited the development of the pathogens tested compared to the control, and the effect was dose dependent. Furthermore, the biocide effects of the different compounds studied were dependent on the species and race of the pathogen. These results indicate that GSLs and their GHPs, as well as extracts of differentBrassicaspecies, have potential to inhibit pathogen growth and offer new opportunities to study the use ofBrassicacrops in biofumigation for the control of multiple diseases.
