Abstract BackgroundAtopic dermatitis (AD) is a chronic, inflammatory, and severely Pruritus cutaneous disease that has an immunologic base. The most common treatments are topical steroids, which bring many adverse side effects. Vitamin D can adjust the immune system; therefore, in this study, we investigated the effect of vitamin D on the abundance of Treg cells in AD patients. MethodsIn this interventional study, 40 subjects who completed the intervention were enrolled, including 20 AD patients (as the experimental group) and 20 healthy subjects (as the control group). Accordingly, the subjects whose vitamin D levels was less than 30 ng/ml were assigned to receive vitamin D (1000 IU) daily for a 2-month duration. The severity of AD was evaluated based on SCORAD (Scoring Atopic Dermatitis) and the amount of CD4+CD25+Foxp3+Treg cells was also assessed using Flow cytometry. ResultsAverage serum level of vitamin D before performing the treatment in the experimental (14/90±4/5) and control groups (15/95±5/0) was lower than that of after the treatment (experimental group 24/60±5/2 and control group 23/60±7/3) (P<0.001). As well, there was a significant difference between the average scores of SCORAD after the vitamin D intervention (P<0.001). The amount of Treg cells significantly increased in the experimental group (P=0.002, Diff=0.35) after performing the intervention.Conclusionit was found that besides conventional medicines, Vitamin D can be adjusted as an effective complementary therapy by modulating the immune system.Trial registration: IRCT20150716023235N13, Registered 11 Feb 2018, https://www.irct.ir

Background: CATSPER 1 (Cation Channel Sperm Associated 1) and CATSPER2 channels have an important role in sperm motility. In this study, the effects of hyperthyroidism on Catsper1 and 2 genes of seminiferous tubules in mice testes were investigated. Objective: The present study was conducted to investigate the effect of hyperthyroidism on the expression of CATSPER1 and CATSPER2 genes in the seminiferous tubules of mice. Materials and Methods: This study was conducted on 20 BALB/C male mice divided into two groups - experimental and control. The experimental group was administered with 500 mg/l levothyroxine (L-thyroxine) liquid solution for two months for inducing hyperthyroidism, which was confirmed by radioimmunoassay. On the other hand, the control group was kept in animal houses under a normal condition. The implementation of real-time polymerase chain reaction and immunohistochemical studies was accomplished after the removal of the testes of the mice under anesthesia induced by chloroform. Results: Results showed that there was no significant difference in CATSPER1 (p = 0.45) and CATSPER2 (p = 0.34) gene expression between groups. At the same time, the color intensity showed no significant enhancement in the hyperthyroidism group (CATSPER1 p = 0.17 and CATSPER2 p = 0.22) as compared to the control group. Conclusion: Considering the key role of CATSPER in the molecular structure of the sperm, our findings showed that the hyperactivity of the thyroid gland has no significant effects on the function of these components. Therefore, it might be concluded that hyperthyroidism has no considerable effects on the seminiferous tubules. Key words: CATSPER1, CATSPER2, Hyperthyroidism, Mice, Sperm.

The results of the use of probiotic strains of microorganisms of the Bacillus family for the correction and formation of the microflora of the gastrointestinal tract and the impact on metabolism in calves are presented. The aim of the study. To analyze the effect of probiotics on the microflora of the gastrointestinal tract in calves and biochemical parameters of blood in calves up to one month. Materials and methods. The research was conducted during 2020 in the conditions of Ukrainian farms for cattle breeding. Five experimental groups of five one-week-old calves were formed in each and one control group. Calves were kept separately in the same conditions on the same diet, but with feeding together with colostrum substitute probiotics of five grams per animal: Bacillus amyloliquefaciense, Bacillus mucilaginosus, Bacillus coagulans, Bacillus megaterium, Bacillus pumilus. The strains are deposited and produced by “Kronos Agro” Ukraine. Results. It was found that as a result of studies when feeding calves B. coagulans, B. pumilus and B. mucilaginosus the number of Lactobacillus sp. was 80 % higher than in the control group. The level of opportunistic pathogens in the experimental group with B. coagulans had minimal values. Animals in the group where B. mucilaginosus was given had a higher amount of Candida - up to 300 CFU/g and Enterobacteriaceae – 200 CFU/g; which is 50 % less compared to control groups, but more than in the experiment with B. coagulans. According to the results of biochemical examination of blood serum in calves, the absence of toxic effects of probiotic strains: Bacillus amyloliquefaciense, Bacillus mucilaginosus, Bacillus coagulans, Bacillus megaterium, Bacillus pumilus on the internal organs of animals was established. Conclusions. It was found that the maximum positive effect on the microflora of the gastrointestinal tract of calves up to 30 days of age had B. coagulans (1×109) when fed at a dose of 5 g per animal. The amount of Lactobacillus sp. was the maximum and reached 800 CFU/g, which is 80 % more than in the control group. At the same time, the level of opportunistic pathogens in the experimental group with B. coagulans had minimal indicators and was: Clostridium by 20 %, Escherichia coli – by 70 %, Enterobacteriaceae, Staphylococcus and Candida – 100 % less than the control. In the study of biochemical parameters, it was found that the activity of enzymes, protein and glucose levels in the serum of experimental animals fed with B. coagulans were within the physiological norm, indicating a normal metabolic process and no toxic effects.
