Comparative Assessment of Antibacterial Activity of Chromolaena odorata Leaf Extracts against Selected Clinical Bacterial Isolates

Aims: The study was carried out to determine the phytochemical constituents and antibacterial activity of aqueous and ethanolic extracts of fresh leaves of Moringa oleifera Lam., Pterocarpus santalinoides L’Herit DC and Ceiba pentandra L. on bacterial isolates; Salmonella typhi, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. Methodology: The plant leaves were dried, pulverized and phytochemical tests were done according to standard laboratory procedure. Aqueous and ethanolic extracts were obtained from 20 g of the of the ground leaves. Antibacterial assay was carried out with Disc diffusion method on seven concentrations of the extracts ;100,50,25,12.5, 6.25,3.125,1.5625 mg/ml and compared with standard antibiotics. Isolated bacterial pathogens; Salmonella typhi, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa (1.0 x 105 cfu /ml) were used as test organisms. Results: Alkaloids, steroidal aglycones, glycosides, proteins, carbohydrates, reducing sugars, tannins, saponins, vitamins A and E were present in all the plant samples. Flavonoids and cardiac glycosides were not detected in Pterocarpus santalinoides and Ceiba pentandra, respectively. Anthracene glycoside was absent in all samples. Aqueous and ethanolic extracts of M. oleifera showed antibacterial activities against all the bacterial isolates at minimum inhibitory concentration (MIC) of 3.125 mg/ml and 1.5625 mg/ml respectively. Pterocarpus santalinoides showed inhibitory activity only on Salmonella typhi at 3.125 mg/ml and Escherichia coli 1.5625 mg/ml MIC. Ceiba pentandra showed spectrum of antibacterial activity against all the bacterial isolates at 1.56 mg/ml MIC with exception of Salmonella typhi. E. coli was the most susceptible to the leaf extracts. Salmonella typhi was not sensitive to the leaf extracts of Ceiba pentandra, while Staphylococcus aureus and Pseudomonas aeruginosa were not sensitive to the leaf extracts of Pterocarpus santalinoides. Conclusion: It can be concluded that both aqueous and ethanolic leaf extracts had antibacterial activity against the test organism, thus justifying their use in folklore medicine.

Several studies have demonstrated the biological efficacy of leaf, stem and root powders or extracts of Chromolaena odorata (L.) King and Robinson against insect pests but those that are focused on the biological efficacy of aqueous leaf extracts against Macrotermes species are scanty. Current management of termites with synthetic insecticides is being discouraged due to human and environmental hazards. Therefore, the insecticidal effectiveness of aqueous leaf extract C. odorata against Macrotermes species was investigated. Five concentrations (0, 2.5, 5.0, 7.5 and 10.0% (w/v)) of the aqueous extract of C. odorata plant were evaluated for repellency and toxicity on the worker caste of Macrotermes species following standard procedures. The filter paper impregnation technique was used for the bioassay. Percentage repellency was monitored for 30 minutes and mortality recorded at 12, 24 and 36 hours post exposure. The leaf extract of C. odorata significantly repelled 95% of Macrotermes species at the highest concentration of 10% (w/v) after 30 minutes post treatment exposure. Mortality of Macrotermes species was independent of treatment concentration, but dependent on duration of exposure. All treatment concentrations of aqueous leaf extract of C. odorata caused significant mortality against Macrotermes species ranging between 94% and 98% compared to the control; indicating very great potential for adoption and use in the management of Macrotermes species.

Studies were conducted to identify Pseudomonas fluorescens isolates from a collection of bacteria isolated from bacterial haemorrhagic septicaemia infected carp and catfish, evaluate their antibiotic sensitivity pattern and screen the antibacterial activity of some medicinal plant extracts against the isolates.. A total of 10 isolates were identified as P. fluorescens by morphological, physiological and biochemical tests. In vitro antibiotic sensitivity test of the P. fluorescens isolates were conducted by disc diffusion method for seven antibiotics where, all of the isolates were found to be sensitive only against streptomycin and gentamycin but, most of the isolates (80%) were found resistant to chloramphenicol (C). Moreover, eighty percent of the isolates showed resistance to multiple antibiotics. A total of 118 plant extracts were screened for their antibacterial activity against the P. fluorescens isolates where the isolates exhibited sensitivity to 30 samples. Leaf extracts of Tamarindus indicus, Terminalia chebula, Citrus aurantifolia, Eugenia caryophyllata and Spondias pinnata were found to inhibit the growth of all of the P. fluorescens isolates.
