A method for the quantitative determination of streptomycin sulfate in medicines by the turbidimetric method has been developedand validated. Based on the results of the experiments, it was found that the metrological characteristics of such validation parameters of the method as linearity, precision, and correctness do not exceed the validation criteria. Linearity was noted in the range of streptomycin concentrations from 3.75 to 8.43 μg/ml. The results of validation tests of the method for the quantitative determination of streptomycin indicate the prospects and feasibility of introducing the turbidimetric method into the domestic system for standardization and quality assessment of aminoglycoside antibiotics.

The quality of research antibodies is an issue for decades. Although several papers have been published to improve the situation, their impact seems to be limited. This publication makes the effort to simplify the description of validation criteria in a way that the occasional antibody user is able to assess the validation level of an immunochemical reagent. A simple, 1-page checklist is supplied for the practical application of these criteria.

On validation of practice-based qualitative research Erica Baarends & Frits Simon An analysis of the most used literature on (practice-based) qualitative research in the Netherlands shows that the described criteria for validity are mainly derived from a quantitative perspective. As a consequence, the voice of the researcher dominates in the research instead of the voice of the research respondents. It is argued that in the age of Facebook and considering the current legitimization crisis of science, practice-based qualitative researchers need validation criteria that respect the polyvocality of the respondents.

AbstractBackground:Disturbance of the circadian rhythm has been associated with disease states, such as metabolic disorders, depression and cancer. Quantification of the circadian markers such as melatonin and cortisol critically depend on reliable and reproducible analytical methods. Previously, melatonin and cortisol were primarily analyzed separately, mainly using immunoassays.Methods:Here we describe the validation and application of a high-throughput liquid chromatography in combination with mass spectrometry (LC-MS/MS) method for the combined analysis of melatonin and cortisol in plasma and saliva. The LC-MS/MS method was validated according to international validation guidelines. We used this method to analyze total plasma, free plasma (as obtained by equilibrium dialysis) and saliva melatonin and cortisol in healthy adults.Results:Validation results for plasma and saliva melatonin and cortisol were well within the international validation criteria. We observed no difference between saliva collected by passive drooling or Salivette. Moreover, we noted a significant difference in saliva vs. free plasma melatonin. We observed on average 36% (95% CI: 4%–60%) higher salivary melatonin levels in comparison to free plasma melatonin, suggestive of local production of melatonin in the salivary glands.Conclusions:The novel outcome of this study is probably due to the high precision of our LC-MS/MS assay. These outcomes illustrate the added value of accurate and sensitive mass spectrometry based methods for the quantification of neuroendocrine biomarkers.

Abstract The recent episodes of forest fire in Brazil and Australia of 2019 are tragic reminders of the hazards of the forest fire. Globally incidents of forest fire events are in the rise due to human encroachment into wilderness and climate change. Sikkim with a forest cover of more than 47%, suffers seasonal instances of frequent forest fire during the dry winter months. To address this issue, a GIS-aided and MaxEnt machine learning-based forest fire prediction map has been prepared using forest fire inventory database and maps of environmental features. The study indicates that amongst the environmental features, climatic conditions and proximity to roads are the major determinants of the forest fire. Model validation criteria like ROC curve, correlation coefficient and Cohen’s Kappa show a good predictive capability (AUC = 0.95, COR = 0.78, κ = 0.78). The outcomes of this study in the form of a forest fire prediction map can aid the stakeholders of the forest in taking informed mitigation measures.

The success of a teaching process cannot be separated from the learning tools used, one of the requirements is that the learning tools must be valid. The success of a teaching process is measured by the extent to which students can master the subject matter presented by the teacher. A learning model is needed that can make it easier to develop critical thinking skills so that a truly valid learning tool is obtained to measure the potential and skills that exist in students. Among the learning models that emphasize the process of seeking and finding is the inquiry model. This study aims to prepare in measuring critical thinking skills of Madrasah Tsanawiyah students. The Learning Toolkit has been validated by 3 experts and will then be averaged to find the final score. The values obtained will be entered into the validation criteria table based on the validation criteria table and the results are that the tools made are very valid and can be used in the teaching and learning process because previously the tools made have been revised several times. Diperlukan suatu model pembelajaran yang dapat memudahkan dalam mengembangkan perangkat keterampilan berpikir kritis sehingga didapatkan suatu perangkat pembelajaran yang benar-benar valid untuk mengukur potensi dan keterampilan yang ada pada diri siswa. Diantara model pembelajaran yang menekankan kepada proses mencari dan menemukan adalah model inkuiri. Penelitian ini bertujuan untuk mempersiapkan dalam mengukur keterampilan berpikir kritis siswa Madrasasah Tsanawiyah. Perangkat Pembelajaran sudah divalidasi oleh 3 orang ahli kemudian akan dirata-ratakan untuk mencari nilai akhir. Nilai yang didapat akan dimasukkan ke dalam tabel kriteria validasi diberdasarkan tabel kriteria validasi dan hasilnya perangkat yang dibuat termasuk sangat valid dan dapat digunakan dalam proses belajar mengajar karena sebelumnya perangkat yang dibuat sudah dilakukan beberapa kali revisi.  Kata kunci: kevalidan, perangkat pembelajaran, keterampilan berpikir kritis

BACKGROUND Recent advances in mobile sensor technology have led to increased popularity of wrist-worn fitness trackers. The possibility to use a smartwatch as a rehabilitation tool to monitor patients’ heart rate during exercise has won the attention of many researchers. OBJECTIVE The aim of the study was to evaluate the accuracy and precision of HR measurement performed by two wrist monitors: Fitbit Charge 4 (Fitbit) and Xiaomi Mi Band 5 (Xiaomi). METHODS 31 healthy volunteers were asked to perform a stress test on a treadmill. During the test their heart rate was recorded simultaneously by both wristbands and ECG at 1minute intervals. The mean absolute error percentage (MAPE), Lin’s concordance correlation coefficient (LCCC) and Bland-Altman were calculated to compare precision and accuracy of heart rate measurements. The estimated validation criteria were MAPE < 10% and LCCC < .8 RESULTS The overall MAPE of the Fitbit device was 10.19% (±11.79%) and the MAPE of Xiaomi was (6.89 % ± 9.75). LCCC of Fitbit HR measurements was .753 (95% CI:0.717-0.785) and of Xiaomi – .903 (0.886-0.917). In both devices the precision and accuracy were decreasing with the increasing exercise intensity. Age, sex, height, weight, BMI did not influence the accuracy of both devices. CONCLUSIONS The accuracy of a wearable wrist-worn heart rate monitor varies and depends on the intensity of training. The decision concerning the application of such a device as a monitor during in-home rehabilitation should be taken with caution, as it may prove not reliable enough.

Abstract Study question Can we quantitatively determine concentrations of endocrine disruptors namely bisphenol A and S in seminal fluid? Summary answer We developed selective analytical method to simultaneously screen for the presence of bisphenol A (BPA) and S (BPS). What is known already The male reproductive system involves processes, which may be influenced by the disruption of the endocrine system by chemicals called endocrine disruptors (EDs). There is a growing evidence that EDs such as bisphenol A and S may be responsible for the decline in male reproductive health. To date, the claimed adverse effects on male fertility are largely based on the results from studies assessing the relationship between urinary BPA and BPS concentration and semen parameters. The best evidence of an adverse effect of BPA and BPS directly on spermatozoa could be provided by measuring bisphenols concentration directly in seminal fluid. Study design, size, duration To selectively and quantitatively analyzed bisphenols in any biological matrix advanced analytical tools and selective sample preparation protocols must be employed. In this study we developed targeted analytical method based on liquid chromatography tandem mass (LC-MS/MS) detection to measure bisphenol A and S in seminal fluid samples obtained from IVF clinic. A total of 140 samples were analysed. Participants/materials, setting, methods BPA and BPS was extracted from 140 seminal fluid samples using solvent extraction followed by preconcentration step. Samples were analyzed on Agilent 6495 Triple Quadrupole (Agilent Technologies, Santa Clara, CA) operating in the ESI-negative mode. Two MS/MS transitions were used for quantitative LC-MS/MS analyses. Chromatographic separation was achieved on Waters™ ACQUITY™ UPLC™BEH C18 (100 × 2.1 mm, 1.7 µm) column using gradient elution with a mixture of 0.1mM ammonium fluoride and methanol as mobile phases. Main results and the role of chance We developed selective sample preparation method for detection of BPA and BPS in seminal fluid followed by LC-MS/MS detection. The method validation was performed based on FDA guidelines. Validation criteria included limit of detection (LOD), limit of quantitation (LOQ), accuracy and precision. Due to the lack of the certified reference material the validation criteria of the method were assessed in pool of spiked seminal samples. The accuracy of the LC-MS/MS method was evaluated as a percent recovery of the amount of target analyte added into the sample. Recovery rates were above 80% for both analytes. LOD was 0.04 ng/mL for BPA and 0.01 ng/mL for BPS. LOQ was 0.14 ng/mL and 0.02 ng/mL for BPS. Measured BPA concentration ranged from 0.04 ng/mL to 1.62 ng/mL. For BPS, the concentration ranged from 0.01 ng/mL to 0.47 ng/mL. BPA and BPS were detected in 64% and 81% of samples, respectively. Interestingly, BPA showed lower detection frequency compared to BPS. These results are consistent with other studies performed on urine samples. Limitations, reasons for caution The limitation of the developed method is the time-consuming sample preparation and analysis cost. Wider implications of the findings: These results document for the first time the presence of BPS in seminal fluid. Knowing the concentration of BPA and BPS in seminal fluid is crucial for mitigating the associated health risks and initiating intervention and prevention strategies.
