Early transcriptional response of seedling roots to Ralstonia solanacearum in tobacco (Nicotiana tabacum L.)

En el municipio de Arriaga Chiapas, desde el año 2009 se ha observado la muerte de plantas de piñón (Jatropha curcas L.) de 1 y 2 años de edad, con síntomas de marchitez y pudrición basal del tallo. La incidencia registrada ha sido de hasta 85%; la virulencia del patógeno es muy alta ya que ocasiona la muerte de la planta en un lapso de 20 y 30 días. El presente trabajo se realizó en los años 2011 a 2013, para la determinación del agente causal presuntivo. Se realizaron en laboratorio aislamientos en medio B de King (BK) y cloruro de trifenil tetrazolio (TTC), posteriormente los aislamientos fueron sometidas a pruebas bioquímicas y pruebas de hipersensibilidad en tabaco (Nicotiana tabacum L.) cv. Xanthi. Los resultados obtenidos dieron positivos en cinco cepas a Ralstonia solanacearum (E. F. Smith 1896) Yabuuchi et al., 1993.

AbstractGenetic mapping of seed germination traits has been performed with many plant species. In tobacco, however, investigations are rare. In the present study, a bi-parental mapping population consisting of 118 doubled haploid lines and derived from a cross between ‘Beinhart-1000’ and ‘Hicks’ was investigated. Four germination-related traits, total germination (TG), normal germination (NG), time to reach 50% of total germination (T50), and the area under the curve after 200 h of germination (AUC) were considered by examining seeds either untreated or after a moderate controlled deterioration (CD). Quantitative trait loci were found for all traits distributed on 11 out of the 24 linkage groups. It was demonstrated that, as in many other species, germination-related traits are very complex and under polygenic control.
