The content of astaxanthin, including its mono- and diesters, and photosynthetic pigments, has been analyzed in cells of H. pluvialis strain IBCE-H17 under the combined prolonged action of several inducers of astaxanthin accumulation. The effective induction of the astaxanthin accumulation, mainly in the form of monoesters of fatty acids, was shown after a 20-day cultivation under high-intensity light and with the addition of 1-2 g/l of sodium acetate to the culture medium. A simultaneous decrease in the content of chlorophylls and lutein in the H. pluvialis cells was observed under these conditions. The use of sodium acetate in combination with sodium chloride did not lead to noticeable changes in the content of astaxanthin compared with the use of sodium acetate alone. The obtained data can be helpful in the biotechnological production of Haematococcus biomass enriched with economically valuable compounds. astaxanthin, Haematococcus pluvialis, pigments, sodium acetate, sodium chloride, high-intensity light. To elucidate the mechanism underlying increased fatty acid and astaxanthin accumulation in Haematococcus pluvialis, transcriptome analysis was performed to gain insights into the multiple defensive systems elicited by salicylic acid combined with sodium acetate (SAHS) stresses with a time course. Totally, 112,886 unigenes and 61,323 non-repeat genes were identified, and genes involved in carbon metabolism, primary and secondary metabolism, and immune system responses were identified. The results revealed that SA and NaAC provide both energy and precursors to improve cell growth of H. pluvialis and enhance carbon assimilation, astaxanthin, and fatty acids production in this microalga with an effective mechanism. Interestingly, SA was considered to play an important role in lowering transcriptional activity of the fatty acid and astaxanthin biosynthesis genes through self-protection metabolism in H. pluvialis, leading to its adaption to HS stress and finally avoiding massive cell death. Moreover, positive correlations between 15 key genes involved in astaxanthin and fatty acid biosynthesis pathways were found, revealing cooperative relation between these pathways at the transcription level. These results not only enriched our knowledge of the astaxanthin accumulation mechanism in H. pluvialis but also provided a new view on increasing astaxanthin production in H. pluvialis by a moderate and sustainable way in the future.

Phytoene and astaxanthin are 2 important carotenoids in the green alga Haematococcus pluvialis. Under environmental stress, the synthesis of phytoene in H. pluvialis increases significantly, and phytoene is converted to astaxanthin through enzymatic catalysis. This paper analyzes the relationship between astaxanthin and phytoene accumulation in carotenoid synthesis pathways under different concentrations of sodium acetate (NaAc) by high-performance liquid chromatography. The highest concentrations of phytoene and astaxanthin were observed at the NaAc concentration of 6 g l-1 on the 12th day of induction. The highest astaxanthin concentration achieved was 2.26 ± 0.28%. Therefore, we concluded that 6 g l-1 NaAc and induction for 12 d provided the optimal inducing conditions for astaxanthin accumulation in H. pluvialis. psy, pds, lcyB, β-carotene ketolase crtw, and crtz, which are genes related to phytoene and astaxanthin synthesis, were cloned and studied at the transcriptional level. crtw and crtz were continuously up-regulated since the first day of induction, while psy, pds, and lcyB were continuously up-regulated starting on the 3rd day of induction. These findings are important for enhancing our understanding of the mechanism of accumulation of phytoene and astaxanthin in H. pluvialis and provide a foundation for identifying the induction conditions necessary for optimizing astaxanthin production and increasing astaxanthin yields.

Silicon (Si) is distributed throughout plant tissues, but its role in forages has not been clarified. Although Si has been suggested as an antiquality factor which limits the digestibility of structural carbohydrates, other research indicates that its presence in plants does not affect digestibility. We employed x-ray microanalysis to evaluate Si as an antiquality factor at specific sites of two cultivars of bermuda grass (Cynodon dactvlon (L.) Pers.). “Coastal” and “Tifton-78” were chosen for this study because previous work in our lab has shown that, although these two grasses are similar ultrastructurally, they differ in in vitro dry matter digestibility and in percent composition of Si.Two millimeter leaf sections of Tifton-7 8 (Tift-7 8) and Coastal (CBG) were incubated for 72 hr in 2.5% (w/v) cellulase in 0.05 M sodium acetate buffer, pH 5.0. For controls, sections were incubated in the sodium acetate buffer or were not treated.

Summary1. Separated leukocytes or total blood from normal subjects, untreated leukaemic patients and from leukaemic patients treated with cytostatic agents were incubated with CH3COONa-l-C14. Radioactivity of mixed lipids was measured at standard time intervals.2. The time incorporation curve observed with leukocytes from treated leukaemic patients showed after an initial linear part, a more rapid levelling off than the curves observed with leukocytes from untreated and normal subjects.3. Therefore, an indirect effect of treatment on leukocyte lipid synthesis seems to be present.4. Phospholipid and neutral lipid synthesis by leukaemic leukocytes was also studied. The results give no evidence that these fractions as a whole have any precursor-product relation.

Our objective was to develop a process for recovering acetyl groups from industrial northeast hardwood chips. Extraction experiments showed that the white liquor charge of 6% effective alkali at 50°C and liquor-to-wood ratio of 4:1 leads to the complete extraction of acetyl groups from industrial hardwood chips. Electrodialysis was assessed as a technique for separating and concentrating sodium acetate from synthetic hardwood extract. Preliminary separation experiments using dilute sodium acetate showed that the sodium acetate can be concentrated up to approximately 24% by weight from an initial concentration of about 2% by weight. The effects of current density, feed concentration, electro-osmosis, and osmosis on the separation of sodium acetate were evaluated. Finally, selectivity experiments were performed to study the effect of various components of the white liquor on the separation of sodium acetate. Fully oxidized synthetic white liquor was used in selectivity experiments to avoid deleterious effects of Na2S. Selectivity experiments using synthetic oxidized white liquor showed a significant decrease in the separation efficiency due to the presence of sodium hydroxide, sodium carbonate, and sodium sulfate in the synthetic extract.
