An efficient regeneration was developed using mature and immature embryos by using Maize (Zea mays L) variety MU 2092. Mature embryos are removed from surface sterilized seeds, slice them into halves and immature embryos are detached from seed endosperm. Both are used as explants to initiate callus on N6 medium supplemented with 2,4 D @ 4.0 mg.L-1. The induction frequency of primary calli i.e embryogenic callus was 90% in maize. The embryogenic calli on N6 medium supplemented with 6-benzylaminopurine (BAP) @ 0.5 mg.L-1 and kinetin @ 0.5 mg.L-1 was more effective in producing shoots. The culture expressed maximum plant regeneration potential with eight shoots per embryo on regeneration. Green shoots thus developed were successfully rooted within 20 days on MS media containing IBA (Indole-3-Butyric acid) 1mg L-1. Over 86 % of rooted plants grew well and produced seeds normally when transferred to green house. The important advantage of this improved method is shortening of regeneration time by providing an efficient and rapid regeneration tool for mature and immature embryos.

Callus induction from explants is a critical process in regeneration, micropropagation and transformation of maize (Zea mays L.) plants. Formation of callus from plant tissues on culture is affected by several factors. This study revealed to establish the effect of genotype, source of explants and auxin concentration on callus induction from five genotypes UMI 757 (G1), UMI 615 (G2), UMI 112 (G3), UMI 285 (G4) and CO 1 (G5) and one hybrid CO H (M) 5 (G6). Callus induction of the six maize varieties was investigated using immature embryos (E1), leaf bits (E2), root tips (E3), hypocotyls (E4) and seeds (E5) as explants with different concentrations of hormones. In this study, immature embryo was taken from 10 to 12 days after pollination (DAP) to get maximum response. The highest percentage of callus induction was observed (99.10) in immature embryo culture and seed culture gave the highest percentage of rhizogenic callus formation when compare to immature embryo. Among the genotypes tested, CO H (M) 5 recorded the highest callus induction percentage on (2D2K2) medium composition.

Penelitian tentang pengaruh pemberian produktivitas pupuk organik terhadap hasil Tanaman Jagung (Zea mays L) di tanah mineral penelitian ini di laksanakan pada bulan Februari sampai Mei, yang bertempat di Sungai Salak Kecsmstsn Tempuling Kabupaten Indragiri Hilir Provinsi Riau. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) yang disusun secara faktorial yang terdiri dari 2 faktor dan 3 ulangan. Faktor B adalah bokashi pupuk kandang yang terdiri dari 4 taraf yaitu B0 (tanpa pemberianpupuk kandang), B1 (5 ton/ha) dan B2 (10 tom/ ha), serta B3 (15 ton/ha) Parameter yang di amati adalah tinggi tanaman, panjang daun ke tujuh, berat brangkasan basah, berat berangkasan kering, berat tongkol pertanaman sampel, diameter tongkol , produksi per plot, dan berat 100 biji. Selanjutnya data yang di peroleh di olah secara statistik, apabila F hitung lebih besar dari F tabel di lanjutkan dengan uji lanjut Tukey HSD pada taraf 5%. Hasil penelitian menunjukan bahwa interaksi bokashi pupuk kandang dan varietas berpengaruh nyata terhadap berat tongkol dan produksi dan produksi per plot, akan tetapi tidak berpengaruh nyata terhadap tinggi tanaman, panjang daun ke tujuh, berat brangkasan basah, berat brangkasan kering , diameter tongkol dan berat 1000 biji. Untuk perlakuan bokashi pupuk kandang secara tunggal berpengaruh nyata terhadap terhadap diameter tongkol , akan tetapi tidak berpengaruh nyata terhadap tinggi tanaman, panjang daun ke tujuh, berat brangkasan basah, berat brangkasan kering, berat tongkol, produksi per plot, dan berat 1000 biji, perlakuan bokashi terbaik terdapat pada pemberian 15 ton/ha. Sedangkan perlakuan varietas secara tunggal berpengaruh nyata terhadap berat brangkasan basah, berat tongkol, dan produksi per plot seta berat 1000 biji, akan tetapi tidak berbeda nyata dengan tinggi tanaman, panjang daun ke tujuh, berat brangkasan kering, dan diameter tongkol. Varietass terbaik adalah NT 10
