In a previous research dealing with the structure of semi-permeable membranes, it has been shown that the copper ferrocyanide membrane is built up from colloidal particles having diameters ranging from 50 to 400 μμ ; also that the pores enclosed by these particles have an average diameter which, although large enough to allow passage to any non-colloidal molecule, is small enough to be entirely within the range of capillary effects. On the basis of these facts the hypothesis was advanced that the selective properties of colloidal membranes are due to selective adsorption; a copper ferrocyanide membrane immersed in cane-sugar solution, for instance, being permeable to the water and not to the cane sugar, because the water, rather than the sugar, is selectively adsorbed or “condensed” on to the membrane surface and into its capillaries. The present research is an experimental investigation of this view. The measurements made herein consist in a determination of the change in concentration brought about by immersing more or less dry colloidal copper ferrocyanide in cane-sugar solutions of various strengths. The results obtained tend to confirm the adsorption hypothesis for this case at least.

Abstract The autosomal recessive transmission of Glanzmann's thrombasthenia (GT) and Bernard-Soulier syndrome (BSS), together with requests of families who already had children with these diseases, prompted us to investigate the feasibility of their antenatal diagnosis. The preliminary step leading to the early detection of GT or BSS was to characterize, in the normal human fetus, the platelet antigens and glycoproteins (GPs) and to define their normal amounts on the membrane surface. Blood samples from 32 fetuses between 18 to 26 weeks of gestation were collected by direct puncture of the umbilical vein using an ultrasound-guided needle. Polyclonal antibodies from human origin directed against PLA1, Leka antigens, and the GPIIb IIIa complex (IgGL), or murine monoclonal antibodies specific for GPIb (AN51, 6D1), GPIIIa (AP-3), or GPIIb IIIa (AP-2) were studied using platelet suspension immunofluorescence tests. The binding of each antibody was quantified using a cytofluorograph (Ortho 50H). PLA1 and Leka antigens were expressed in normal amounts on fetal platelets as early as 16 weeks of intrauterine life. The GPIIb IIIa complex quantified by polyclonal or monoclonal antibodies was in the same range in fetuses (IgGL = 427 +/- 23 AUF, AP-2 = 459.5 +/- 8.5; AP-3 = 536 +/- 14) and in adults (IgGL = 420 +/- 30; AP-2 = 498 +/- 11; AP-3 = 515 +/- 13). The platelet binding of antibodies that recognized GPIb was higher in fetuses (AN51 = 491.5 +/- 14; 6D1 = 479 +/- 15) than in adults (AN51 = 426.5 +/- 9; 6D1 = 449 +/- 8.7). These results suggest that immunological techniques can be applied as early as 18 weeks of gestation for the antenatal diagnosis of GT and BSS.

Murine bone marrow-derived macrophages, which measure 13.8 +/− 2.3 microns diameter in suspension, can ingest IgG-opsonized latex beads greater than 20 microns diameter. A precise assay has allowed the determination of the phagocytic capacity, and of physiological parameters that limit that capacity. Ingestion of beads larger than 15 microns diameter required IgG-opsonization, and took 30 minutes to reach completion. Despite the dependence on Fc-receptors for phagocytosis of larger beads, cells reached their limit before all cell surface Fc-receptors were occupied. The maximal membrane surface area after frustrated phagocytosis of opsonized coverslips was similar to the membrane surface area required to engulf particles at the limiting diameter, indicating that the capacity was independent of particle shape. Vacuolation of the lysosomal compartment with sucrose, which expanded endocytic compartments, lowered the phagocytic capacity. This decrease was reversed when sucrose vacuoles were collapsed by incubation of cells with invertase. These experiments indicate that the phagocytic capacity is limited by the amount of available membrane, rather than by the availability of Fc-receptors. The capacity was also reduced by depolymerization of cytoplasmic microtubules with nocodazole. Nocodazole did not affect the area of maximal cell spreading during frustrated phagocytosis, but did alter the shape of the spread cells. Thus, microtubules may coordinate cytoplasm for engulfment of the largest particles.

Honey is composed of macromolecules arranged into multicomponent colloidal particles dispersed in a supersaturated sugar solution. The core part of colloidal particles in honey is made up of high-molecular weight protein-polyphenol complexes. We designed a multi-step extraction process to gain better insight into the phenolic compounds strongly bound to proteins in honey. Honeys were sequentially extracted by solvents of reduced polarities and the extraction process was monitored by LC-ESI-MS/MS. Unexpectedly, the results revealed ubiquinone-like compounds that partitioned to both, soluble supernatants and protein-bound insoluble residues from which they were released after the pronase-digestion of proteins. The accurate mass measurement and MS/MS fragmentation patterns using UPHLC-MS/MS coupled to quadrupole orbitrap confirmed their identification as ubiquinones. Distribution of ubiquinone-bound proteins was further investigated by the fractionation of honey protein-polyphenol complexes by size-exclusion chromatography followed by LC-ESI-MS analysis. Mass spectra revealed the presence of ubiquinones (UQs) in fractions of high polyphenol to protein ratio. The dominant mass peaks observed in these fractions were identified as UQ-3, UQ-5, and UQ-7. Since the quinone group of UQs is involved in redox reaction, we discuss the possibility that UQs may contribute to the antioxidant/proxidant activity of these complexes.


The author concludes, from his microscopic examinations of the structure of muscular fibres, that those subservient to the functions of animal life have, in man, an average diameter of one 400dth of an inch, and are surrounded by transverse circular striae varying in thickness, and in the number contained in a given space. He describes these striae as constituted by actual elevations on the surface of the fibre, with intermediate depressions, considerably narrower than the diameter of a globule of the blood. Each of these muscular fibres, of which the diameter is one 400dth of an inch, is divisible into bands or fibrillae, each of which is again subdivisible into about one hundred tubular filaments, arranged parallel to one another, in a longitudinal direction, around the axis of the tubular fibre which they compose, and which contains in its centre a soluble gluten. The partial separation of the fibrillae gives rise to the appearance of broken or interrupted circular striae, which are occasionally seen. The diameter of each filament is one 16,000dth of an inch, or about a third part of that of a globule of the blood. On the other hand, the muscles of organic life are composed, not of fibres similar to those above described, but of filaments only ; these filaments being interwoven with each other in irregularly disposed lines of various thickness; having for the most part a longitudinal direction, but forming a kind of untraceable network. They are readily distinguishable from tendinous fibres, by the filaments of the latter being uniform in their size, and pursuing individually one unvarying course, in lines parallel to each other. The fibres of the heart appear to possess a somewhat compound character of texture. The muscles of the pharynx exhibit the character of animal life; while those of the oesophagus, the stomach, the intestines, and the arterial system, possess that of inorganic life. The determination of the exact nature of the muscular fibres of the iris presented considerable difficulties, which the author has not yet been able satisfactorily to overcome.

Abstract. The article is dedicated to the determination of the wash depth of mountain rivers in floods and overflows. As well-known, hydrotechnical equipment is exposed to destructive damages of the floods and overflows. The undersurface of bridge basement and coast guards are washed away, the surface of drainage devices in water supply and dams (Düker) at river crossings are opened up. Therefore, the protection of the equipment against the destructive damage of the floods and overflows should be ensured. To this aim, first of all, the depth of the fortification of the equipment basement in riverbeds and the width through which the river can flow should be determined and the works of installing coast guards should be accordingly implemented. Since the flow regime of rivers due to floods in the course of mountain riverbeds dramatically changes, the width, depth, roughness and cross-sectional area of the riverbed also changes. To that reason, in the smallest case, calculation formulas includes the average width and wash depth of the riverbed in the course of the floods and overflows, average diameter of undersurface soils, the slope of the location of hydrotechnical structures, velocity due to the average diameter of the riverbed soils and so on. The article contains the analyses of theoretical and practical materials about the floods and overflows in mountain rivers. For the rivers flowing through the southern hills of Great Caucasus Mountains, the expressions for determining the riverbed parameters and hydrological parameters of rivers are used. According to the expressions, average width due to non-washing of the riverbed in floods in accordance with flood flow and slope of the studied part of the riverbed, the average depth of the riverbed crossing the flood, the average velocity of the flow and the wash depth in accordance with them are determined. basing on all these, the determining method of the wash depth of mountain riverbeds in floods have been worked out and determining the wash depth have been recorded. The studied methodology can be used in determining the riverbed parameters in floods and overflows.
