AbstractKiwifruit (Actinidia spp.) is a significant plantation crop belonging to family Actinidiaceae, having deciduous, dioecious, and scrambling vines with chromosome number 2n=58. Dioecy in kiwifruit forms the basis for several breeding programs. The present study was carried out for diversity analysis in kiwifruit genotypes using RAPD markers. 7 kiwifruit genotypes (2 males viz. Allision & Tomuri and 5 females viz. Hayward, Bruno, Allision, Monty & Abbott) were analysed for molecular polymorphism using 94 RAPD primers, out of which 23 primers amplified the genomic DNA in all the genotypes. RAPD data was analysed using NTSYS-pc software and dendrogram construction was done using UPGMA method. Two separate clusters of male and female genotypes were formed. Similarity matrix indices showed maximum similarity between Tomuri (M) and Allision (M) with a similarity coefficient of 0.719 while Abbott (F) and Allision (M) were found to have least similarity having a similarity coefficient of 0.521. Four RAPD primers amplified unique amplicons in Monty, Hayward, Bruno, Allision (M) and Abbott and two primers amplified unique amplicons in Allision (M) & Tomuri (M) along with the male and female plants of Allision genotype respectively. Therefore, these primers can help in distinguishing the genotypes of kiwifruit and can also be validated as putative markers for the sex identification in kiwifruit.

Genetic diversity among 24 genotypes of sorghum was studied by using 20 random primers. The total number of amplified PCR bands was 591 of which 495 (83.75%) were polymorphic displaying PIC values ranging from 0.27 to 0.69 with an average of 0.52. Similarity coefficients among the genotypes were ranged from 0.33 to 0.89. The similarity coefficient was used to construct a UPGMA based dendrogram using average linkage between groups. The analysis illustrates considerable potential of RAPD markers for estimating genetic diversity among 24 genotypes of sorghum.

Program pemuliaan ikan mas varietas Punten dilakukan dengan seleksi individu terhadap karakter bobot ikan. Pembentukan populasi dasar untuk kegiatan seleksi dilakukan dengan memijahkan secara massal induk ikan mas yang terdiri atas 20 induk betina dan 21 induk jantan yang dikoleksi dari daerah Punten, Kepanjen (delapan betina dan enam jantan), Kediri (tujuh betina dan 12 jantan), Sragen (27 betina dan 10 jantan), dan Blitar (15 betina dan 11 jantan). Larva umur 10 hari dipelihara selama empat bulan. Selanjutnya dilakukan penjarangan sebesar 50% dan benih dipelihara selama 14 bulan untuk dilakukan seleksi dengan panduan hasil sampling 250 ekor individu setiap populasi. Seleksi terhadap calon induk dilakukan saat umur 18 bulan pada populasi jantan dan betina secara terpisah dengan memilih berdasarkan 10% bobot ikan yang terbaik. Calon induk yang terseleksi kemudian dipelihara hingga matang gonad, kemudian dipilih sebanyak 150 pasang dan dipijahkan secara massal. Didapatkan respons positif dari hasil seleksi berdasarkan bobot ikan, yaitu 49,89 g atau 3,66% (populasi ikan jantan) dan 168,47 g atau 11,43% (populasi ikan betina). Nilai heritabilitas untuk bobot ikan adalah 0,238 (jantan) dan 0,505 (betina).Punten carp breeding programs were carried out by individual selection for body weight trait. The base population for selection activities were conducted by mass breeding of parent consisted of 20 female and 21 male collected from area Punten, eight female and six male (Kepanjen), seven female and 12 male (Kediri), 27 female and 10 male (Sragen), 15 female and 11 male (Blitar). Larvae 10 days old reared for four moths. Then after spacing out 50% of total harvest, the offspring reared for 14 months for selection activity based on the sampling of 250 individual each population. Selection of broodstock candidates performed since 18 months age on male and female populations separately by selecting based on 10% of fish with best body weight. Candidates selected broodstocks were then maintained until mature. In oder to produce the next generation 150 pairs were sets and held for mass spawning. The results revealed that selection response were positive, 49.89 g (3.66%) for male and 168.47 (11.43%) for female. Heritability for body weight is 0.238 (male) and 0.505 (female).

The experiment was conducted to assess the available genetic variability amongst Indian Basmati rice and identify co-dominant and reproducible robust simple sequence repeat markers for drought resistance and their utilization in marker assisted selection for developing drought resistant / tolerant aromatic rice cultivars in various parts of India as well as in world. DNA was isolated from fresh and young leaf tissues of 35 cultivars of basmati rice using the CTAB procedure of Doyle and Doyle with slightly modifications. The DNA was further quantified by spectrophotometer at 260 nm and 280 nm. The quality and quantity of DNA were checked by agarose gel electrophoresis. Out of 60 SSR markers, 18 were found to be polymorphic and the rest 42 primers were monomorphic. One of the main purposes of SSR markers in genomic study is the characterization of genetic resources to help gene bank management. The informative (18) SSR markers were capable to discriminate the entire cultivars of basmati rice used in this research work. A total of 52 alleles at 18 loci could be scored. The allelic richness per locus diversified from 2 to 5 with an average of 2.89 alleles per locus. The amplitude of Polymorphism Information Content (PIC) value is 0.066 (RM 1068) to 0.730 (RM 1059) with an average of 0.505. The Jaccard’s similarity coefficient ranged from 0.79 to 0.95. Polymorphic finding content showed a positive correlation (r =0.69) with number of alleles at the SSR locus. However it is recommended that SSR markers can be efficiently utilized for this purpose. The maximum similarity coefficient was observed between Jeerakasab and Kalanamak with a coefficient value of 95% and the minimum similarity was found between Jeerakasab and Pusa basmati-1 with a coefficient value of 79%.

DNA fingerprinting and genetic diversity of four Bangladesh Rice Research Institute (BRRI) hybrid varieties and their parental lines were carried out. A total of 73 microsatellite markers were tested for screening the genotypes. Among the 73 amplified products, 37% had polymorphic bands showing 81 alleles. The number of alleles per locus ranged from two (RM10) to eight (RM327), where average allele number was 4.333. The Polymorphism Information Contents (PIC) lied between 0.337 (RM10) and 0.852 (RM327). RM327 was the most robust marker providing the highest PIC value (0.852). Pair-wise genetic dissimilarity coefficient interaction showed that BRRI hybrids two was the most genetically distant from each other whereas BRRI hybrids one, three, four and their respective parents were very close. Cluster analysis based on Dice’s similarity coefficient UPGMA system grouped BRRI hybrid and their parental lines into four major clusters at 0.41 cut off similarity coefficient. Four BRRI hybrid varieties grouped into four distinct clusters along with their component lines indicating their genetic closeness. SUMMARY. Diversity analysis of moroccan carob (Ceratonia siliqua L.) accessions using phenotypic traits and RAPD markers. The carob (Ceratonia siliqua L.) is a perennial leguminous (Caesalpinioideae) that grows as an evergreen shrub or tree. It’s an important component of the Mediterranean vegetation and its adaptation in marginal soils of the Mediterranean regions is important environmentally and economically. Phenotypic and genetic diversity among 10 Ceratonia siliqua accessions coming from different areas of Morocco were studied with morphometric and RAPD approaches. The analysis of discriminative fruit characters showed highly significant differences among these accessions, the closely related ones have a similarity level less than 65%. No molecular studies have been carried out so far on Ceratonia siliqua. To provide markers useful for molecular diversity study of the carob tree, genomic DNA extraction and amplification conditions were finalized and 67 arbitrary primers were screened. 52 of them showed clearly reproducible banding patterns. The analysis of RAPD profiles revealed a high degree of genetic diversity within these carob accessions and enabled each of them to be uniquely fingerprinted. Overall, in our study we note that morphological relationship between carob accessions is deeply different to their molecular relationship. Concerning the distribution of the accessions according to their geographical origin, clustering based on RAPD data revealed a rough distribution of theses accessions. Indeed, Sidi Bou Ottman, Demnate and Essaouira accessions coming from the south of Morocco seem to be linked in the PCA plot. However, two geographically distant accessions, Aïn Sfa and Ouazzane coming from the north (about 700 Km) were clustered with Essaouira and Demanate - Sidi Bou Ottman, respectively.Key words. Analyse de la diversite des accessions du caroubier marocain (Ceratonia siliqua L.) utilisant des traits phenotypiques et des marqueurs RAPD. Le caroubier (Ceratonia siliqua L.) est une légumineuse pérenne (Caesalpinioideae), à feuillage persistant et pouvant croître entant qu’arbrisseau ou arbre. C’est un composant important de la végétation méditerranéenne et son adaptation aux sols marginaux des régions méditerranéennes est d’une grande importance écologique et économique. La diversité phénotypique et génétique au sein de 10 accessions de caroubier provenant de différentes régions du Maroc a été étudiée par des approches morphométriques et moléculaires (RAPD). L’analyse des caractères morphologiques discriminants du fruit a montré des différences hautement significatives au sein des accessions, puisque les apparentées d’entre elles n’ont approximativement que 65% de similarité. Jusqu’à nos jours, aucune étude moléculaire n’a été entreprise sur Ceratonia siliqua. Pour mettre à disposition des marqueurs utiles pour l’étude de la diversité moléculaire du caroubier, l’extraction de l’ADN génomique et les conditions de l’amplification ont été mises au point et 67 amorces arbitraires ont été criblées. 52 d’entre elles ont montré clairement des profils de bandes reproductibles. L’analyse des profils RAPD a révélé une diversité génétique de haut niveau entre les accessions de caroubier ce qui a permis à chacune d’elles d’être distinguée génétiquement de façon singulière. De manière générale, nous avons noté que la relation morphologique entre les accessions du caroubier est profondément différente de leur relation moléculaire. Concernant la distribution des accessions selon leur origine géographique, le regroupement basé sur les données de la RAPD a révélé une distribution grossière. En effet, les provenances de Sidi Bou Ottman, Demnate et Essaouira, originaires du sud de Maroc, semblent être liés par l’analyse des composantes principales (PCA). Cependant, deux accessions géographiquement distantes, Aïn Sfa et Ouazzane, originaires du Nord (approximativement 700 Kms) ont été liées avec Essaouira et Demanate - Sidi Bou Ottman, respectivement.Mots clés. Random Amplified Polymorphic DNA (RAPD) markers were used to study the molecular genetic diversity analysis among six BARI released lentil varieties viz. BARI masur-1, BARI masur-2, BARI masur-3, BARI masur-4, BARI masur-5 and BARI masur-6. PCR amplified products were visualized on 1.0% agarose gel and the band for each primer were scored. Ten RAPD markers were used in this study. Out of them 7 primers showed amplification of 53 DNA fragments with 60.37% of them being polymorphic. The highest number of polymorphic loci was noticed in the variety BARI masur-3. The same variety also showed maximum Nei’s gene diversity value (0.0552). The highest Nei’s genetic distance (0.5002) was observed in BARI masur-1 vs. BARI masur-5 whereas, the lowest genetic distance (0.0692) was found in BARI masur-1 vs. BARI masur-2. The unweighted pair group method of arithmetic mean (UPGMA) dendrogram based on Nei’s genetic distance grouped the six cultivars into two main clusters. BARI masur-1, BARI masur-2 and BARI masur-3 were in cluster I and BARI masur-4, BARI masur-5 and BARI masur-6 were in cluster II. The cultivar BARI masur-4 was closest to the cultivar BARI masur-6 with the lowest genetic distance (0.0972) and the highest genetic distance (0.5002) was found between BARI masur-1 and BARI masur-5. The RAPD markers were found to be useful in molecular characterization of lentil varieties which could be utilized by the breeders for the improvement of lentil cultivars. Randomly amplified polymorphic DNA (RAPD) markers were used for the genetic variation and relationship analysis among 12 Mango (Mangifera indica L.) germplasm. Five oligonucleotide primers were employed to amplify DNA from 12 cultivars. PCR amplification with five primers generated 45 reproducible, clear and distinct bands, out of which 41 bands are considered polymorphic and the remaining four fragments (8.88%)  monomorphic. The size of amplified product ranged from 200 (RPI-5) to 3000 base pairs (RPI-1) with an average of nine bands per primer. The average polymorphism in all the 12 cultivars using the five primers was found to be 91.91%. Among all the primers RPI-2 and RPI-4 have shown 100% polymorphism while RPI-5 was found to be least polymorphism (81.81%). One specific band, namely was found with RPI-5, in a particular variety, Chiratpuri. The UPGMA (Unweighted Pair Group Method of Arithmetic Mean) dendrogram based on Jaccard’s similarity coefficient segregated the 12 mango germplasm into two clusters. Langra, Chiratpuri, Pravasankar, Alphanso, Sindhu and Kesar formed one cluster and rest six mango germplasm grouped together into another cluster. Sindhu and Alphanso cultivar pair was very close to each other with highest similarity coefficient (0.78), which was comparatively higher than all other cultivar pairs. On the other hand, Pravasankar and Neelam cultivar pair was more distinct to each other with the lowest intervarietal similarity coefficient 0.38. Bak-ri-hyang (Thymus quinquecostatus Celak.) is an important medicinal and aromatic plant in Korea. T. quinquecostatus population and is always mixed with other thyme cultivars during cultivation and marketing. Hence, this study aimed to determine the genetic variability and the essential oil composition of three Korean native thyme, T. quinquecostatus cultivars collected from the Wolchul, Jiri, and Odae mountains, in comparison with six commercial thyme cultivars (T. vulgaris), to distinguish Bak-ri-hyang from other thyme cultivars. The composition of essential oils obtained from nine individuals was analyzed by gas chromatography–mass spectrometry (GC–MS). The random amplified polymorphic DNA (RAPD) analysis was accomplished using 16 different primers. The GC–MS analysis revealed that Wolchul, creeping, golden, and orange cultivars belong to the geraniol chemotype. Whereas the Odae, lemon, and silver cultivars belong to the thymol chemotype. Further, linalool was the most abundant component in carpet and Jiri cultivars. The RAPD analysis demonstrated that all thyme cultivars showed characteristic RAPD patterns that allowed their identification. In total, 133 bands were obtained using 16 primers, and 124 bands were polymorphic, corresponding to 93.2% polymorphism. Cluster analysis of RAPD markers established the presence of clear separation from nine thyme cultivars. The highest dissimilarity and similarity coefficient of the RAPD markers were 0.58 and 0.98, respectively. According to the RAPD patterns, the nine thyme cultivars could be divided into two major clusters. Among three Korean cultivars, the Wolchul and Odae cultivars were placed into the same cluster, but they did not show identical clustering with their essential oil compositions. The findings of the present study suggest that RAPD analysis can be a useful tool for marker-assisted identification of T. quinquecostatus from other Thymus species.
