Influence of NaCl on Productivity and Fluorescence Parameters of Nasturtium officinale R. Br. and Its Relevance to Artificial Closed Ecosystems

The study demonstrated the effects of precursor feeding on the production of glucosinolates (GSLs), flavonoids, polyphenols, saccharides, and photosynthetic pigments in Nasturtium officinale microshoot cultures grown in Plantform bioreactors. It also evaluated the antioxidant and antimicrobial activities of extracts. L-phenylalanine (Phe) and L-tryptophan (Trp) as precursors were tested at 0.05, 0.1, 0.5, 1.0, and 3.0 mM. They were added at the beginning (day 0) or on day 10 of the culture. Microshoots were harvested after 20 days. Microshoots treated with 3.0 mM Phe (day 0) had the highest total GSL content (269.20 mg/100 g DW). The qualitative and quantitative profiles of the GSLs (UHPLC-DAD-MS/MS) were influenced by precursor feeding. Phe at 3.0 mM stimulated the best production of 4-methoxyglucobrassicin (149.99 mg/100 g DW) and gluconasturtiin (36.17 mg/100 g DW). Total flavonoids increased to a maximum of 1364.38 mg/100 g DW with 3.0 mM Phe (day 0), and polyphenols to a maximum of 1062.76 mg/100 g DW with 3.0 mM Trp (day 0). The precursors also increased the amounts of p-coumaric and ferulic acids, and rutoside, and generally increased the production of active photosynthetic pigments. Antioxidant potential increased the most with 0.1 mM Phe (day 0) (CUPRAC, FRAP), and with 0.5 mM Trp (day 10) (DPPH). The extracts of microshoots treated with 3.0 mM Phe (day 0) showed the most promising bacteriostatic activity against microaerobic Gram-positive acne strains (MIC 250–500 µg/mL, 20–21 mm inhibition zones). No extract was cytotoxic to normal human fibroblasts over the tested concentration range (up to 250 μg/mL).

In Northern Europe, an increase in heterogeneity of summer precipitation regime will subject forests to water deficit and drought. This is particularly topical for Norway spruce (Picea abies Karst.), which is a drought sensitive, yet economically important species. Nevertheless, local populations still might be highly plastic and tolerant, supporting their commercial application. Accordingly, the tolerance of Norway spruce seedlings from an Eastern Baltic provenance (western part of Latvia) to artificial drought according to soil type was assessed in a shelter experiment. To simulate drought, seedlings were subjected to reduced amounts (0%, 25%, 50%, 75%, and 100%) of naturally occurring precipitation (irrigation intensity). Three soil types (oligotrophic mineral, mesotrophic mineral, and peat) were tested. Seedling height, chlorophyll a concentration, and fluorescence parameters were measured. Both growth and photochemical reactions were affected by the irrigation intensity, the effect of which experienced an interacted with soil type, implying complex controls of drought sensitivity. Seedlings were more sensitive to irrigation intensity on mesotrophic mineral soil, as suggested by growth and photosynthetic activity. However, the responses were nonlinear, as the highest performance (growth and fluorescence parameters) of seedlings occurred in response to intermediate drought. On peat soil, which had the highest water-bearing capacity, an inverse response to irrigation intensity was observed. In general, fluorescence parameters were more sensitive and showed more immediate reaction to soil water deficit than concentration of chlorophyll on mesotrophic mineral and peat soils, while the latter was a better indicator of seedling performance on oligotrophic soil. This indicated considerable plastic acclimation and hence tolerance of seedlings from the local Norway spruce population to drought, though drought sensitivity is age-dependent.

Drought is one of the major factors limiting wheat yield in many developing countries worldwide. Parameters of chlorophyll a fluorescence kinetics under drought stress conditions have been used to characterize dehydration tolerance in wheat. In the present study, a set of 94 doubled haploid lines obtained from Chinese Spring × SQ1 (CSDH), mapped with 450 markers, was evaluated for yield (grain dry weight/main stem ear), number of grains/main stem ear (NG) and chlorophyll a fluorescence parameters (FC) under moderate and severe drought stress, and compared with results for well-watered plants. quantitative trait loci (QTLs) were identified using Windows QTLCartographer version 2.5 software and the results were analysed using single-marker analysis (SMA) and composite interval mapping (CIM). Analysis using SMA and CIM showed mostly similar QTLs for all traits, though more QTLs were identified by SMA than by CIM. The genetic control of yield, NG and FC varied considerably between drought-stressed and non-stressed plants. Although no major QTL co-locations were found for yield and FC using CIM, the co-location of QTLs for NG, yield and Fv/Fm in drought-stressed plants was observed on chromosome 5A using SMA.

The effect of increasing concentrations (0·01, 0·1, 1 and 5 mg l−1) of intact (FLT) and photo-modified (phFLT) fluoranthene and the duration of exposure (1, 2, 3, 5 and 7 days) on the chlorophyll fluorescence parameters (F0, FV/FM, and ΦII) of symbiotic algae in the thalli of two foliose lichens Lasallia pustulata and Umbilicaria hirsuta was investigated. In addition the FLT concentration in thalli of both lichen species was determined and a bioconcentration factor (BCF) was calculated. The results obtained demonstrated that the concentrations of FLT and especially phFLT (1 and 5 mg l−1) applied affected primary photochemical processes of photosynthesis in the algae of both lichen species. The F0 value increased and the FV/FM and ΦII values decreased. The fluoranthene content in the thallus of both lichen species increased with increasing FLT concentration in the environment.
