This study aims to investigate the chemical constituents of the stem of Retama raetam growing in Saudi Arabia. The organic and inorganic composition of ethanol extract of R. raetam stem has been explored using direct analysis in real time-mass spectrometry (DART-MS), gas chromatography-mass spectrometry (GC-MS), and inductively coupled plasma-mass spectrometry (ICP-MS). Analysis conducted by DART-MS and GC-MS reveals the presence of several interesting organic constituents identified as 2,4-di-tert-butylphenol, sparteine, benzenepropanoic acid, 3,5-bis(1,1-dimethylethyl)-4-hydroxy-, methyl ester, phthalic acid, 1-octadecanol, squalene, argentamin, 2,4-di-tert-butylphenol, sparteine, benzene propanoic acid, 3,5-bis(1,1-dimethylethyl)-4-hydroxy-, methyl ester, phthalic acid, 1-octadecanol, squalene, argentamin, and hentriacontane in the ethanol extract of the R. raetam stem. The ICP-MS analysis of stem extract showed the presence of a significant amount of important inorganic elements including aluminum, chlorine, calcium, bromine magnesium, phosphorus, scandium, and chromium. The current study complements other R. raetam extract investigations carried out in the past and provides the additional data for the future research studies.

Objective: The objective of the study was to evaluate brine shrimp lethality of Blighia sapida stem-bark extract and its fractions and identify the bioactive constituents in the ethyl acetate fraction (EAF) using gas chromatography-mass spectrometry (GC-MS) technique. Methods: The ethanol extract (EE) and its fractions were subjected to lethality assay, and GC-MS analysis of EAF was carried out. Results: The lethality test showed a concentration-dependent mortality rate in the brine shrimp nauplii for the EE and its fractions. GC-MS analysis of EAF of the extract revealed the existence of 13 peaks of the GC-MS chromatogram with only one prominent compound, n-hexadecanoic acid (peak area of 10.13%). Conclusion: The result revealed the presence of 13 bioactive components in the EAF of the extract, the majority of which have been reported for different biological activities, hence, justifies the use of the plant in the treatment and management of different diseases ethnomedicinally.

Gas Chromatography-Mass Spectrometry (GC-MS) is normally used for direct analysis of chemical components existing in herbal medicines. The medicinal plants are having numerous bioactive components which are identified even at less than 1ng by using GC-MS or LC-MS analysis. The aim of this study is to identify the secondary metabolites present in the leaves of B. tomentosa using gas chromatography-mass spectrometry (GC-MS) analysis. In the present study the ethanol extract of the leaves of Bauhinia tomentosa has been subjected to GC-MS analysis, while the mass spectra of the compounds found in the extract was matched with the National Institute of Standards and Technology (NIST) library. GC-MS analysis revealed the presence of 14 secondary metabolites. These compounds were identified by comparing their retention times and peak areas with those from the literature and by interpretation of the mass spectra. The major secondary metabolites were DL-.alpha.-tocopherol (14.84%), 2-[(trimethylsilyl oxy]-, methyl ester, 1-alpha,2-alpha.-epoxy-1-beta-methylcholesta-4,6-dien-3-one (12.93%), pentacosenoic acid (12.71%), phytol (10.28%), Ethyl Isoallocholate (8.197%), Spirost-8-en-11-one-3-hydroxy-,(3-beta,5 alpha,14 beta,20 beta, 22 beta,25R)-(8.162%), Urs-12-en-28-ol (6.675%), 1-Octadecyne (5.702%) and Cholest-8-en-3-beta-ol,Acetate (5.426%). The compounds having area less that 5% were considered of no significance. These findings suggest that the presence of these secondary metabolites may be the cause for the properties exhibited by Bauhinia tomentosa. Thus, presence of various bioactive compounds justifies the use of the leaf for various ailments by traditional practitioners.

Gas Chromatography-Mass Spectrometry (GC-MS) the best technique to identified the compounds of essential oils by comparison of mass spectra data obtained from the sample with that taken from pure commercially available standards injected under the same conditions. To characterize the chemical constituents of Hibiscus Flower using GC-MS, the shade dried flower powder was extracted with methanol by using Microwave-assisted Hydrodistillation (MAHD). The GC-MS analysis provided different peaks determining the presence of ten compounds. These compounds havebiological activity namely 2-Phenylthiolane (57.31%), Cyclohexene, 3-ethenyl- (25.91%), Acetaldehyde (12.70%), N-Methylallylamine (9.99%),  ropanamide (6.79%) and Phthalic acid, bis (7-methyloctyl) ester (5.21%). From the results, it can be concluded that Jasmine flower extract shows the presence of 10 phytocompounds. The presence of various bioactive compounds justifies the use of the jasmine flower for various ailments by traditional practitioners.

Objective: The aim of this study was to evaluate the antibacterial and antioxidant properties of ethyl acetate extract of Streptomyces omiyaensis SCH2. The chemical constituents of the extract were investigated using gas chromatography-mass spectrometry (GC-MS).Methods: Secondary metabolites from S. omiyaensis were produced by submerge fermentation using ISP2 medium with 3% NaCl (w/v) for 21 days. The culture filtrate was extracted with ethyl acetate. The chemical constituents were detected in the GC-MS analysis. Antibacterial activity was performed using disc diffusion and broth microdilution methods. Antioxidant activity was evaluated by determining the reducing power capacity and free radical scavenging assays.Results: The GC-MS analysis of the SCH2 extract revealed the presence of four compounds. The main constituents were 2-phenylacetamide (79%). The extract exhibited the highest zone of inhibition against some pathogenic bacteria such as Enterobacter cloacae, Klebsiella pneumoniae, and Bacillus subtilis. In addition, the lowest minimum inhibitory concentration (IC) and minimum bactericidal concentration values of extract were obtained for E. cloacae (0.125 and 4 mg/ml). The extract showed antioxidant potential with IC50 values of 2.078.13±24.58 μg/ml and 475.74±4.56 μg/ml for 2,2-diphenyl-1-picryl-hydrazyl and 2,2′-azino-bis[3-ethylbenzothiazoline-6-sulphonic acid] assays, respectively. The ferric reducing antioxidant power value of extract was 110.36±1.75 mmol FeSO4.7H2O/g extract.Conclusion: This study indicated that S. omiyaensis extract possesses antibacterial and antioxidant activities. GC-MS analysis revealed the presence of major chemical constituents, acetamide, and pyrrolopyrazine which could be responsible for the biological activities. S. omiyaensis extract could be used as a potential of natural antibacterial and antioxidant agents for pharmaceutical and medical applications.

Carica papaya leaves are used as a remedy for the management of cancer. Freeze-dried C. papaya leaf juice was extracted using a supercritical fluid extraction system. Compound identification was carried out using analytical techniques including liquid chromatography coupled to high-resolution quadrupole time-of-flight mass spectrometry (LC–QToF-MS) and gas chromatography–mass spectrometry (GC–MS). The cytotoxic activities of the scCO2 extract and its chemical constituents were determined using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay on squamous cell carcinoma (SCC25) and human keratinocyte (HaCaT) cell lines. The chemical constituents were quantified by QToF-MS. The supercritical carbon dioxide (scCO2) extract of papaya freeze-dried leaf juice showed cytotoxic activity against SCC25. Three phytosterols, namely, β-sitosterol, campesterol, and stigmasterol, together with α-tocopherol, were confirmed to be present in the scCO2 extract. Quantitative analysis showed that β-sitosterol was the major phytosterol present followed by α-tocopherol, campesterol, and stigmasterol. β-Sitosterol and campesterol were active against SCC25 (half maximal inhibitory concentration (IC50) ≈ 1 µM), while stigmasterol was less active (~33 µM) but was biologically more selective against SCC25. Interestingly, an equimolar mixture of phytosterols was not more effective (no synergistic effect was observed) but was more selective than the individual compounds. The compounds identified are likely accountable for at least part of the cytotoxicity and selectivity effects of C. papaya.

In addition to the trichilianones A–D recently reported from Trichilia adolfi, a continuing investigation of the chemical constituents of the ethanol extract of the bark of this medicinal plant yielded the five new limonoids 1–5. They are characterized by having four fused rings and are new examples of prieurianin-type limonoids, having a ε-lactone which in 4 and 5 is α, β- unsaturated. The structures of the isolated metabolites were determined by high field NMR spectroscopy and HR mass spectrometry. The new metabolites were shown to have the ε-lactone fused with a tetrahydrofuran ring which is connected to an oxidized hexane ring joined with a cyclo-pentanone having a 3-furanyl substituent. As the crude extract possesses antileishmanial activity, the compounds were assayed for cytotoxic and antiparasitic activities in vitro in murine macrophage cells (raw 264.7 cells) and in Leishmania amazoniensis as well as L. braziliensis promastigotes. Metabolites 1–3 and 5 showed moderate cytotoxicity (between 30–94 µg/mL) but are not responsible for the antileishmanial effect of the extract.
