<p>ABSTRAK</p><p>Sekitar 75 juta bibit kakao asal embrio somatik (ES) telah ditanam <br /> di lapangan.  Evaluasi keragaan tanaman tersebut perlu terus dilakukan. <br /> Penelitian yang bertujuan untuk mengevaluasi keragaan tanaman kakao <br /> asal ES di lapangan telah dilaksanakan di delapan lokasi di Kabupaten <br /> Soppeng, Sulawesi Selatan dan kabupaten Mamuju (Sulawesi Barat) pada <br /> bulan Juni 2012 sampai  Desember 2013. Pengamatan dilakukan terhadap <br /> tanaman umur 1,5-4 tahun meliputi tinggi jorget, lingkar batang, jumlah <br /> cabang primer, persentase tanaman berbunga, persentase tanaman berbuah, jumlah buah per pohon, jumlah biji per buah, bobot kering biji serta kejadian serangan hama dan penyakit. Tanaman kakao asal ES memiliki arsitektur   seperti   tanaman   yang   berasal   dari   biji,   yaitu memiliki pertumbuhan  dimorfik  dan  membentuk  jorget.  Rataan  tinggi  jorget bervariasi antar lokasi, sebagian besar berkisar 100 -150 cm. Di lapangan ditemukan tanaman asal ES dengan tinggi jorget &gt; 2 m. Perlu dievaluasi lebih lanjut apakah tinggi jorget &gt; 2 m merupakan bentuk penyimpangan atau hanya pengaruh faktor lingkungan. Pada saat dilakukan pengamatan, tanaman kakao asal ES yang dievaluasi sudah berbunga dan berbuah dengan persentase berbunga 34-100%, berbuah 16-100%, dan dengan hasil buah per pohon 6-37 buah. Bobot buah yang dipanen berkisar 277-418 g dengan rataan jumlah biji per buah 43 biji dan bobot kering per biji 0.5 -1.4 g. Kejadian serangan penggerek buah di 3 lokasi pengamatan mencapai 82, 34.8 dan 49.6%, sedangkan busuk buah 2, 4.3 dan 18%. Studi lebih lanjut  diperlukan  untuk  memastikan  bahwa  tanaman  kakao  asal  ES mempunyai karakteristik tanaman dan hasil yang tidak berbeda dengan tanaman kakao  asal bibit tradisional yang telah biasa digunakan petani.</p><p>Kata kunci:  Theobroma   cacao   L.,   bibit   ES,   perbanyakan   masal, pertumbuhan dan komponen hasil, variasi somaklonal</p><p> </p><p>Field Performance of Cacao Somatic Embryos Derived Plants</p><p>ABSTRACT</p><p>Approximately 75 millions of cacao seedlings propagated through <br /> somatic embryogenesis (SE) have been planted in the field. Evaluation of <br /> the performance of those SE derived plants needs to be continued. The <br /> research aimed to evaluate the performance of the cacao SE derived plants <br /> in the field have been implemented in 8 locations in Soppeng district, <br /> South Sulawesi and the Mamuju district, West Sulawesi in June 2012 and <br /> December 2013.   Collected   data   include:   jorquette   height,   trunk <br /> circumference,  jorquette branch  numbers,  flowering  and  fruiting tree <br /> percentages, fruit numbers per tree, pod weight, bean number per pod, <br /> bean dry weight and the disease and pest infection rates. Cacao plants <br />derived from SE showed similar plant architectures to those of seed derived ones, such as having dimorphic growth and forming jorquette. The jorquette height of the majority of cacao trees in the evaluated regionsranged from 100-150 cm. A few SE derived cacao trees show jorquette height &gt; 2 m. However, it needs further studies to determine whether they indicate either abnormality or environment effects. The SE derived cacao trees in the studied locations has bear flowers and fruits. The percentages of cacao trees with flowers or fruits at the age of 2.5 years ranged from 34-100% or 16-100%, respectively. The observed pods number per tree was 6-37 pods, the pod weight was 277-418 g, the beans number per pod was 42-43  and the bean dry weight was 0.5 -1.4 g. The incidences of fruit borer infection at the three evaluated locations were 82, 34.8 dan 49.6%, while the incidences of black pod infection were 2, 4.3 dan 18%. To <br /> conclude whether the SE derived cacao trees are comparable to those of <br /> the traditional cacao planting materials  requires  more comprehensive <br /> studies.</p><p>Keywords:  Theobroma cacao L., SE derived seedling, mass propagation, growth and yield components, somaclonal variation</p>

Research is needed to better understand the influence of cell volume and fertility on watermelon transplant size and field performance in order to determine the most economic production practices. `Jubilee' watermelon transplants were grown using a 4 x 4 factorial experimental design consisting of 4 cell volumes (30.7, 65.5, 147.5, and 349.6 cm3) and 4 fertility rates (0, 1/4, 1/2, and full-strength Hoagland's solution). Transplant shoot dry weight significantly increased as cell volume and fertility increased. Increasing cell volume linearly increased watermelon number/ha and tons/ha for early and total harvest in 1995. The average weight per watermelon significantly increased for early-harvested fruit but not for total harvest as cell volume increased in 1995. Soluble solids concentration linearly increased with increasing cell volume for early and total harvests in 1995. Cell volume had no significant influence on the harvest parameters measured in 1997. In 1995, increasing fertility linearly increased watermelon number/ha and tons/ha for early harvests. Increasing fertility increased the soluble solids concentration linearly for early-harvested watermelons in 1997 but not in 1995. Fertility rate had no significant influence on any of the other harvest parameters measured in 1995 and 1997. The growing conditions and disease pressure in 1997 reduced melons/ha, yield, and soluble solids content when compared to 1995 values. The half-strength Hoagland's solution produced the greatest number of watermelons/ha, tons/ha, and the highest soluble solids concentration in 1995 and 1997. Pretransplant nutritional conditioning had no significant effect on total `Jubilee' watermelon production in Louisiana for 1995 and 1997.

A total of 60 farmers' cacao accessions (Theobroma cacao L.) from Nicaragua were investigated using microsatellite markers to reveal their genetic composition and to identify potentially resistant genotypes against the black pod disease caused by Phytophthora palmivora. These accessions were compared with 21 breeders' accessions maintained locally, two Criollo accessions from Costa Rica and two accessions from Ecuador. The analyses showed a low level of differentiation among groups of farmers' accessions (FST = 0.06) and that six Nicaraguan accessions were genetically closely related to the two Criollo accessions used as a reference. In addition, seven distinct genotypes were found to have allelic composition that may indicate linkage to resistance alleles, thus being potential parental lines in future breeding programmes.

Clonal plants that produced by somatic embryogenesis technique is one of the best choice to produce supperior clonal cacao (Theobroma cacao L.) planting materials. The somatic embryogenesis technique is a possible way for massive propagation, the outcome is true to type plants, the architecture similarity that the seedlings but there is not segregation like seedlings plants. At present mass production started of plantlets production until post-acclimatized plantlets of somatic embryogenesis cocoa was done at Indonesian Coffee and Cocoa Research Institute. Distribution system of the planting materials to whole areas in form of as up-rooted post-acclimatized plantlet. Some problems identified to reduce probability of decreasing viability of up-rooted post-acclimatized plantlets and one of them is extreme internal water deficit. This research investigate of the influece storage condition (air tight and non-air tight) and box storage (mica plastic and cardboardbox). The first experiment result show, there is no significant different between mica plastic and cardboard box usage for storage of post-acclimatized cocoa pantlet. Viability of up-rooted post acclimatized cocoa plantlet influenced exactly by air tight and non-air tight storage condition. Air tight storage condition have better viability of up-rooted post acclimatised (81,58%) than non-air tight storage condition (65,00%). Leaf sanasence on air tight storage condition (10,33%) lower than non-air tight storage (32,58%). There is not significantly on volume storage per plantlet between 4.416 cm3 and 12.600 cm3. Relationship between fallen leaves and cocoa planlet viability follow negative linear correlation y = -1,4719x + 104,88 (R2 = 0,9772). The second experiment treatment showed that maximal storage periode of post cclimatized cocoa plantlet just until 6 days stored (97%) and not significant with 3 days one. Viability of post acclimatized cocoa plantlet decreased after 6 days storage period.Key words : Somatic embryogenesis, post acclimatized cacao plantlet, storage condition, box storage, volume storage, storage period and viability.

AbstractThe characteristics of pod related to cocoa pod borer resistance (CPB, Conopomorpha cramerella Snell.) had been identified in a series study. This research has objective to evaluate performance of the characteristics using more diverse of genetic background to select criteria for selection. Genetic materials for this study were 25 cocoa clones which be planted in Central Sulawesi for resistant evaluation. Field evaluation of the resistance were assessed using the variable of the percentage of unextractable bean, number of entry and exit hole larvae by which the clones were grouped into 5 groups of resistance. A laboratory works were carried out to assess pod characteristics based on the number of trichome, granule of tannin and thickness the lignified-tissue of sclerotic layer using micro-technique method at the different level of pod maturity (3.0; 3.5; 4.0 months). Correlation between groups of those variables was analyzed using Canonical Correlation. The analysis performed a positive association between the thickness of sclerotic layer at the secondary furrow with the number of entry holes and the number of entry holes through sclerotic layer. The thickness performed a higher value of the coefficient in association with the variables of canonical for pod characteristics (0.59; 0.55; 0.43) and the variables of canonical for CPB resistance (0.54; 0.51; 0.39) that would presenting the characteristics of pod related to CPB resistance. Lignification at sclerotic layer was considered as genotypic expressions due to the thickness at the secondary furrow at 3.0, 3.5 and 4.0 months of pod maturity performed high value of broad-sense heritability i.e. 0.75, 0.89 and 0.92 respectively. A qualitative assessment of the lignification clearly differentiate the resistant clones (ARDACIAR 10) with the susceptible clones (ICCRI 04, KW 516 and KW 564).Key words : cocoa pod borer, Theobroma cacao L., pod characteristics, resistance

Cocoa grafting and budding is usually carried out on 4-5 month old seedling, thus it needs 9-12 months to be ready tranplanted to the field. Effort to shorten time in the nursery can be done by early propagation. The aim of this research was to study effect of clones, tying and foliar application of grafsticks on the percentage of graftake. This study was carried out in Kaliwining Experimental Station, 45 m a.s.l. and D climate type (Schmidt & Ferguson). First experiment was RCBD design and replicated 3 times, and factorial treatment 4 x 3. The first factor were clones, i.e. TSH 858, ICS 13, ICS 60 and DR 2, the second factor was grafstick wiring, i.e. without, wiring 2 and 4 weeks before using. The 2nd experiment using RCBD design replicated 3 times and factorial treatment 3 x 3 x 2. The first factor was clones, i.e. KW 162, KW 163 and KW 165; the second ones was wiring, i.e. without, wiring 2 and 4 weeks before using, and the 3rd factor was manuring, i.e. with and without foliar application. Rootstock was ICS 60 seedlings of 30 day old, and grafting method was cleft grafting above cotyledons. Variables observed include C and N total of the grafstick, percentage of graftake, shoot length, diameter, wet and dry weight. The result showed that because the grafstick flush periodically, graftstick wiring was not effective to increase total nutrient (C and N) on the grafstick. Furthermore, their effect on the graftake was not significant. Until 30 days first, percentage of graftake was 90-100%, but then decreased sharply to 30-60% depend on the clones. From the first experiment, DR 2 showed the highest graftake (62%), and KW 162 (39%) was the 2nd ones. Symptom on the death plants was started on the new leaves, that showed wilt, necrotic then fall. Isolation of those symptoms in the laboratory showed that Rhizoctonia solani, Phytophthora palmivora and Colletotrichum gloeosporioides. Anatomical analysis of the death graft union showed parenchymatous linked, weak and hollow when microtomised. Health union showed lignified accumulation that made tight union. It can be concluded that early cocoa grafting will success if the nursery is separated far from cocoa plantation, the medium is steril from soil borne disease, the graftstick is health, and tying of graft union must tight enough, then disease control must be done properly.Key Words : Theobroma cacao, clone, wiring, graft union, graftake.
