Background: Marine sponges provided a great source of natural products with promising biological activity. This study was aimed to investigate the chemical constituents of methanol extracts of selected Indonesian marine sponges (Callyspongia sp., Clathria sp., Melophlus sarasinorum, and Xestospongia sp.), collected from the Saponda Islands, Southeast Sulawesi, Indonesia as well as to evaluate their antimicrobial and antioxidant activities. Methods: LCMS/MS analysis used to identify the compounds. Agar well diffusion and DPPH assays were used to evaluate the antimicrobial and antioxidant activities. Results: Chemical screening reported alkaloids, terpenoids, steroids, and saponins from all investigated sponges. The LC-MS/MS analysis identified various compounds which mainly contained steroids. Antimicrobial activity (against Bacillus subtilis, Escherichia coli, Salmonella enterica, and Candida albicans) was only shown by the Xestospongia sp. extract. Meanwhile, extracts of M. sarasinorum, Xestospongia sp., and Callyspongia sp. exhibited potent radical scavenging activity. Conclusion: The study concluded that the selected sponges could provide various groups of compounds. Methanol extracts of these sponges could be used as sources of antimicrobial and antioxidant agents.

The present study was carried out to investigate the antioxidant activity of the water and methanol extracts of leaves and fruits extracts of Chamaerops humilis L. by using ABTS cation radicals and cupric reducing antioxidant capacity (CUPRAC). Anticholinesterase effect of the extracts was tested against both AChE and BChE using a microplate-reader assay based on the Ellman method. The methanol extracts of C. humilis leaves contained relatively higher content of flavonoids and total phenolics than those of fruits. All the extracts were found to have different levels of antioxidant activity in the systems tested. The leave extract showed the highest value of antioxidant activity, based on ABTS radical-scavenging activity, while the fruit water extract showed the highest value (0.53±0.50 µg/mL) of cupric reducing antioxidant activity.  Our data indicates that both methanol and water fruit extract were active for BChE inhibition (31.65 ± 0.37 and 30.19 ± 0.56%) respectively, whereas, all leave extracts did not show any activity against BChE. The present study demonstrated that the methanol and water extracts fractions of C. humilis have different responses with different antioxidant methods. Our results suggest that the C. humilis could be used as a source of antioxidant agents and may be beneficial in the AD treatment.

Background: The role of small molecules as antioxidants to prevent the oxidation of other molecules and inhibit them from radical formation is the area of much interest to cure disease especially cancer. Moreover, the antioxidants play important role as stabilizers to prevent oxidation of fuels and lubricants. </P><P> Methods: In the present study, fifteen tetrakis-Schiff’s bases derivatives (1-15) were synthesized and screened for their antioxidant activities. Compounds 1-15 were synthesized by continuous stirring of reaction mixture of 1,4-bis (hydrazonomethyl)benzene (1 mmol) with various substituted aromatic aldehydes (2 mmol) in distilled water using acetic acid as catalyst at room temperature for 2-10 min. </P><P> Results: Our present study showed that all compounds are better ferrous ion-chelating agents except compound 1,4-bis((E)-((E)-(3,4-dimethoxybenzylidene)hydrazono)methyl)benzene (6) (IC50 = 329.26 &#177; 4.75 &#181;M) which has slightly low activity than the standard EDTA (IC50 = 318.40 &#177; 5.53 μM). In addition, DPPH radical scavenging activity of eleven compounds showed higher activity than the standard. However, remaining four compounds showed comparable radical scavenging activity to the standard DPPH (IC50 = 257.77 &#177; 4.60 &#181;M). </P><P> Conclusion: The series of fifteen Schiff’s bases (1-15) were synthesized and evaluated as antioxidants. From both assays, it has been demonstrated that most of the tetrakis-Schiff’s bases have potential to serve as leads for the development of antioxidant agents for future research.

In this research work Semicarbazide, thiosemicarbazide derivatives 3 to 25 were synthesized by conventional methods with high percentage yield and reaction rate. 1H-NMR and EIMS spectroscopic techniques were used to elucidate the structure of the synthesized compounds. The effect of thiosemicarbazide and semicarbazide derivatives as an antioxidant agents were studied by DPPH free radical scavenging, ferric ion reducing, ferrous ion chelating assays. Higher DDPH radical scavenging activity exhibited by most of the compounds as compared to standard vitamin C. Excellent ferric ion reducing activity was indicated by compounds of theseriesas compared to standard vitamin C. However most of the compounds generally showed average ferrous ion chelating activity than standard EDTA.

In this study, dichloromethane, acetone, and methanol extracts of the aerial parts of the Salvia marashica plant which is an endemic species to Anatolia, were investigated. The total phenolic amounts of these extracts were determined as pyrocatechol equivalent and total flavonoids as quercetin equivalent. Antioxidant activity was determined by four complementary methods including inhibition of lipid peroxidation (by β-carotene color expression), DPPH free radical scavenging activity, ABTS cation radical scavenging activity and CUPRAC methods. Anticholinesterase activity of the extracts was investigated by the Ellman method against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes. Viability and cytotoxic activity tests were carried out on the fibroblast L929 cells and cytotoxic A549 lung cancer cells, respectively. The triterpenoids and diterpenoids constitute the major secondary metabolites of the S. marashica acetone and methanol extracts isolated by chromatographic methods. Their structures were determined based on spectroscopic methods, namely NMR and mass analyses. Ten terpenoids were obtained from either acetone or methanol extracts of the S. marashica. Seven of them were triterpenoids, elucidated as lupeol, lupeol-3-acetate, lup-12, 20(29)-diene, lup-20(29)-ene, α-amyrin-tetracosanoate, oleanolic acid and ursolic acid besides a steroid β-sitosterol. Two abietane diterpenes, abieta-8,11,13-triene (1) and 18-acetoxymethylene-abieta-8,11,13-triene (2), were obtained from the acetone extract which were isolated from a Salvia species for the first time in the present study. The methanol extract was found to be very rich in rosmarinic acid determined by LC-MS/MS analysis.

Microalgae are attracting considerable interest worldwide. In the present study, two native microalgae strains isolated from Tunisian cost were identified as Chlamydomonas sp. and Navicula sp. We characterized their pigment and protein contents, as well as their carbohydrate and lipid productivity. The predominant fatty acids were found to be α-linolenic acid (C18:3n-3) and palmitoleic acid (C16:1n-7) for Chlamydomonas sp. and Navicula sp. strains respectively. Microalgae methanol extracts showed important in vitro antibacterial activity against all tested gram negative bacteria. Antioxidant activities of methanol extracts were investigated by determining radical scavenging activity according to DPPH (2,2-diphenyl-1-picrylhydrazyl) and ABTS (2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) methods. MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay showed that the newly isolated microalgae were not toxic and have potential application in the fish feeds domain as a supplement in fish meal. Also, the biosorption of cadmium from aqueous solutions by microalgae living cells was evaluated. Large removal capacities were obtained with values ranging from 89.8% to 99.38%. These results are very promising as a starting point for a potential application of these new isolates for in situ bioremediation of heavy metals contaminating aqueous systems.

Melon (Cucumis melo L.) fruits contain multiple health-promoting compounds, including phenolic compounds, which are antioxidants. Accurate measurement of antioxidant activities and total phenolic contents (TPCs) require an efficient solvent extraction. In this study, we evaluated free radical scavenging activity and TPC of melon extracts extracted with 22 different solvent combinations. The DPPH scavenging activities were high in 100% methanolic (39.48 ± 0.36 µg g−1) and 80% methanolic extracts (38.99 ± 0.44 µg g−1). Similarly, the ABTS scavenging activities were high in 100% methanolic (315.11 ± 10.38 µg g−1) and 80% methanol extracts (297.39 ± 14.98 µg g−1). The Folin–Ciocalteu (F–C) assay is typically used to measure TPC but may be affected by interference from sugars and other compounds. Therefore, we optimized an assay for TPC using Fast Blue (FB) salt and developed a standard operating procedure for microplate analysis using FB. Our analysis of standard samples and comparisons with the F–C assay suggested that the optimized FB assay could be used to measure TPC in fruit and juice samples. Moreover, we successfully detected six phenolic compounds in methanol extracts of melon by LC-HR-QTOF/MS.

Torreya grandis cv. Merrillii is an endemic tree species in China, seeds of which are used as a popular snack, possessing beneficial effects on preventing angiosclerosis and coronary heart diseases. In this study, antioxidant activity and chemical constituents of T. grandis cv. Merrillii seed (TGMS) were investigated. The antioxidant activity of different fractions and the ethanol extract was evaluated using 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity, hydroxyl radical scavenging activity and lipid peroxidation assays. The oil, CH2Cl2 and n-BuOH fractions, and ethanol extract of TGMS all showed antioxidant activities in these models, especially the DPPH one. By GC-MS analysis, twenty-seven constituents were identified from the oil fraction of TGMS. The total content of phenolic compounds in the CH2Cl2 and n-BuOH fractions and ethanol extract was also determined by the Folin-Ciocalteau method as 17.6, 21.6 and 12.9 μg/mg, respectively. In addition, analysis of the CH2Cl2 fraction yielded four phenolic compounds: 4-hydroxybenzaldehyde, 4-methoxy pyrocatechol, coniferyl aldehyde, 4-hydroxy cinnamaldehyde, and two steroids, β-sitosterol and daucosterol. These results provide scientific support for the empirical use of TGMS as a medicinal food for cardiovascular diseases.

Anacyclus pyrethrum (L.) Lag has a long tradition of medical use in Algeria. We report for the first time the antioxidant, functional and neuroprotective properties of this plant from Algeria. Differences in biochemical and physiological properties for the aqueous and methanol extracts of A. pyrethrum are reported and related to the main chemical constituents. The A. pyrethrum extracts contained amino acids, caffeoylquinic acids, N-feruloyltyramine and alkaloids, including including N-alkylamides, common in this plant species. Aqueous extracts (0.1 mg/ml) are neuroprotective against H2O2-induced oxidative stress, while methanol extracts (high phenolic content), despite significant antioxidant and free-radical scavenging activity, caused dysregulation of calcium signalling in neurons and was not apparently neuroprotective. Such knowledge can provide incentive for the sustainable cultivation of Algerian A. pyrethrum, with the aim of supporting conservation strategies and preserving biodiversity.

The objective of this the study was investigating the chemical constituents and to assess the potential antioxidant of Fixed Oil from Peganum harmala Seeds. Using cold maceration method to extract the Fixed Oil from Peganum harmala Seeds. The chemical constituents of Peganum harmala Oil were identified and quantified by GC-MS, where Antioxidant activities were evaluated using 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity. Twenty-three components have been identified. The most abundant fatty acid methyl ester were 9,12, -Octadecadienoic acid (Z,Z) methyl ester with concentration 54.16 % followed by 9- octadecenoic acid (Z) methyl ester with concentration 21.75 %. , Hexadecenoic acid, methyl ester 9.33 %, 11-Octadecenoic acid, methyl ester 2.24 % , Methyl stearate 4.86 %and Stigmasterol 2.76 % .The DPPH assay, showed strong antioxidant potential (94, 0.01 compared with standard 91± 0.01. is a good source of antioxidants due to the presence of phenolic compounds, also is a potential source of natural antibacterial, and justify its uses in folkloric medicine.

The fruit of Melothria scabra Naudin is traditionally used by natives of South East Sulawesi and has economic values in the local markets. Nonetheless, little scientific information was gained from this plant to support its development for nutraceutical and pharmaceutical aspects. This study aimed to investigate the phytochemicals contained in the ethanol extract and organic fractions (methanol, ethyl acetate, and hexane) of the fruits using specific reagents and an LC-MS/MS analysis, as well as to evaluate their total phenolics, total flavonoids, and DPPH radical scavenging activity using a dot-blot staining and spectrophotometric assays. Results showed that the fruits of M. scabra contained alkaloids, tannins, flavonoids, terpenoids, and saponins. Six compounds were successfully identified from the ethanol extract of the fruits for the first time that is D-1-[(3-carboxypropyl)amino]-1-deoxyfructose (1), fructose-C3H5NO (2), valine (3), 1β, 3α, 9β-trihydroxyeudesma-5,11(13)-dien-12-oic acid (4), Cucurbitacin B-2-O-α-L-rhamnopyranosyl-β-D-glucopyranoside (5), and 2-heptyl-3-hydroxy-4(H)-quinolone (6). Total phenolics in the extract and organic fractions were in the range of 54.2 ± 2.4 to 259.1 ± 8.4 mg GAE/g, while total flavonoids were in the range of 1.6 ± 0.2 to 22.4 ± 0.2 mg QE/g. The ethanol extract and its organic fractions (methanol and ethyl acetate) were potent radical scavengers with SC50 values ranging from 20.7 to 37.5 µg/mL when compared with ascorbic acid, gallic acid, and quercetin (SC50 of 2.8 to 9.4 µg/mL). This study concludes that M. scabra fruits could be developed as a source of natural antioxidant agents for nutraceuticals and pharmaceuticals purposes.
