Role of Effort in Performance of Observing Responses

The role of effort in performing observing responses in an E-Maze was studied in 32 rats and 4 pigeons. In Phase I of Exp. 1, rats' performance of observing responses was tested in a regular E-Maze condition and in an extra-length condition in which the predictable arm of the E-Maze was 22 in. or 44 in. longer than the unpredictable arm of the E-Maze. The rats in the regular E-Maze condition preferred the predictable arm on free-choice trials, while those in the extra-length condition preferred the unpredictable, shorter side. In Phase 2 of Exp. 1, the rats in the regular E-Maze condition were switched to an extra-length condition, and rats in the extra-length condition were switched to a regular E-Maze condition. When these experimental conditions were reversed, the choice behavior of the subjects in the two groups also reversed. In Exp. 2, pigeons' performance of the observing response was tested in an E-Maze. Four pigeons learned to perform the observing response in a regular E-Maze condition. They were then changed to an extra-length condition. Preference for the predictable side declined after the change. The results of these experiments show that, when extra effort is required to gain access to the discriminative stimuli, subjects do not perform an observing response.

Neurons in the orbitofrontal cortex (OFC) have been shown to encode subjective values, suggesting a role in preference-based decision-making, although the precise relation to choice behavior is unclear. In a repeated two-choice task, subjective values of each choice can account for aggregate choice behavior, which is the overall likelihood of choosing one option over the other. Individual choices, however, are impossible to predict with knowledge of relative subjective values alone. In this study we investigated the role of internal factors in choice behavior with a simple but novel free-choice task and simultaneous recording from individual neurons in nonhuman primate OFC. We found that, first, the observed sequences of choice behavior included periods of exceptionally long runs of each of two available options and periods of frequent switching. Neither a satiety-based mechanism nor a random selection process could explain the observed choice behavior. Second, OFC neurons encode important features of the choice behavior. These features include activity selective for exceptionally long runs of a given choice (stay selectivity) as well as activity selective for switches between choices (switch selectivity). These results suggest that OFC neural activity, in addition to encoding subjective values on a long timescale that is sensitive to satiety, also encodes a signal that fluctuates on a shorter timescale and thereby reflects some of the statistically improbable aspects of free-choice behavior.

<p>Slot machines are a remarkably popular mode of gambling even though they are programmed to make a profit by paying out less money than is put in. One common feature of slot machines, which may increase the likelihood of persistent gambling in the face of this monetary loss, is the near win. This study’s aim was to investigate the conditioned reinforcing properties of near wins using an observing response procedure in the context of a simulated slot machine. In an observing response procedure, participants can use an observing button to produce a stimulus correlated with the availability of reinforcement (S+) or a stimulus correlated with no reinforcement or less reinforcement (S-). The percentage of observing responses made for each stimulus is thought to reflect the reinforcing efficacy of the reinforcer correlated with each stimulus. Experiment 1 successfully tested the procedure with an obvious reinforcer - wins - and found consistently more observing for the S+. In Experiment 2 and 3 the S+ was correlated with near wins, and in Experiment 2 only those with slot-machine experience had consistently more observing for the S+. Experiment 3 increased the probability of wins to enhance the reinforcing efficacy of near wins, but failed to find consistently more observing for the S+, regardless of slot machine or scratchie card experience. These results indicated that near wins are not conditioned reinforcers. However, participants tended to bet more following near wins than losses, which suggested that near wins may instead function as discriminative stimuli.</p>

The increased number of lysosomes as well as the close approximation of lysosomes to the Golgi apparatus in tissue under variety of experimental conditions is commonly observed. These observations suggest Golgi involvement in lysosomal production. The role of the Golgi apparatus in the production of lysosomes in mouse liver was studied by electron microscopy of liver following toxic injury by CCI4.

Empathy theoretically serves an affiliative interpersonal function by satisfying motives for intimacy and union with others. Accordingly, empathy is expected to vary depending on the situation. Inconsistent empirical support for empathy’s affiliative role may be due to methodology focused on individual differences in empathy or differences between controlled experimental conditions, which fail to capture its dynamic and interpersonal nature. To address these shortcomings, we used ecological momentary assessment to establish typical patterns of empathy across everyday interactions. Associations among empathy, affect, and interpersonal behavior of self and interaction partner were examined in a student sample (N=330), then replicated in a pre-registered community sample (N=279). Multi-level structural equation modeling was used to distinguish individual differences in empathy from interaction-level effects. Results show people are more empathetic during positively-valanced interactions with others perceived as warm and when expressing warmth. By confirming the typically affiliative role of empathy, existing research to the contrary can be best understood as exceptions to the norm.

HIV-1 can infect non-dividing cells. The nuclear envelope therefore represents a barrier that HIV-1 must traverse in order to gain access to the host cell chromatin for integration. Hence, nuclear entry is a critical step in the early stages of HIV-1 replication. Following membrane fusion, the viral capsid (CA) lattice, which forms the outer face of the retroviral core, makes numerous interactions with cellular proteins that orchestrate the progress of HIV-1 through the replication cycle. The ability of CA to interact with nuclear pore proteins and other host factors around the nuclear pore determines whether nuclear entry occurs. Uncoating, the process by which the CA lattice opens and/or disassembles, is another critical step that must occur prior to integration. Both early and delayed uncoating have detrimental effects on viral infectivity. How uncoating relates to nuclear entry is currently hotly debated. Recent technological advances have led to intense discussions about the timing, location, and requirements for uncoating and have prompted the field to consider alternative uncoating scenarios that presently focus on uncoating at the nuclear pore and within the nuclear compartment. This review describes recent advances in the study of HIV-1 nuclear entry, outlines the interactions of the retroviral CA protein, and discusses the challenges of investigating HIV-1 uncoating.

AbstractAccording to the principle of energy-conservation principle, effort investment is usually reduced in situations that are perceived as uncontrollable. This is because when success is recognized as impossible, any effortful actions are no longer justified. However, we predicted that individual differences in uncertainty tolerance, i.e., the need for closure (NFC), may moderate effort investment in uncontrollable situations. We tested this prediction in two experimental studies in which we exposed participants with differing levels of NFC to uncontrollable events, and indexed effort through the assessment of systolic blood pressure (SBP) responses. As predicted, in the uncontrollability (vs. controllability) condition, effort investment decreased significantly among low- but not high-NFC participants. Since gaining certainty and achieving closure is not a critical epistemic goal for low-NFC individuals, exerting extra effort to gain certainty is therefore no longer justified. On the other hand, high-NFC participants do not withhold their efforts, as they are highly motivated to obtain certainty. These results may help to account for contradictory findings in effort-investment behaviour and add substantively to the literature concerning motivation toward closure.

AbstractWe present detailed experimental results with cryogenic Yb:YLF gain media in rod-geometry. We have comparatively investigated continuous-wave (cw) lasing and regenerative amplification performance under different experimental conditions. In the cw lasing experiments effect of crystal doping, cw laser cavity geometry and pump wavelength on lasing performance were explored. Regenerative amplification behavior was analyzed and the role of depolarization losses on performance was investigated. A recently developed temperature estimation method was also employed for the first time in estimating average crystal temperature under lasing conditions. It is shown that the thermal lens induced by transverse temperature gradients is the main limiting factor and strategies for future improvements are discussed. To the best of our knowledge, the achieved results in this study (375 W in cw, and 90 W in regenerative amplification) are the highest average powers ever obtained from this system via employing the broadband E//a axis.
