Objective: To study and compare the effect of Poly herbal plant extract (PHPE) with Glibenclamide (GL) on various parameters in Streptozotocin (STZ) induced diabetic rats. Methods: Diabetes was induced by combining High Fat-diet and injecting low dose Streptozotocin (35 mg/kg body wt.) to Sprague-dawley rats. Diabetic rats were treated with chloroform leaf extract of Azadirachta indica, aqueous leaf extract of Bougainvillea spectabilis and ethanolic seed extract of Trigonella foenum graecum combined in ratio of 1:2:3 respectively at dose of 600 mg/kg body weight by oral gavaging for 28 days. The results were compared with standard anti diabetic drug Glibenclamide given in dose of 500 µg/kg body weight. Results: Increase in body weight of both PHPE and GL treated diabetic rats was found to be statistically significant (p<0.05) compared to diabetic control rats. Decline in FBG levels of both PHPE and GL treated diabetic rats were found to be highly significant statistically (p<0.001) when compared to diabetic control rats at the end of study. Total Cholesterol (TC) and Triglycerides level in diabetic rats treated with PHPE were found to be highly statistically significant (p<0.001) compared to diabetic control rats. Pancreas of PHPE treated diabetic rats revealed partial restoration in size and number of islet of langerhans. Reduction in widening between acinar and islet cells noted. Glibenclamide treated diabetic rats showed much more improvement in pancreatic cell architecture by returning to its normal structure and size. Conclusion: In present study PHPE has shown to decrease elevated FBG level and improve in body weight at the end of study in diabetic rats which can be suggested due to modification in carbohydrate metabolic pathways, stimulation of insulin production by the pancreas, increased peripheral utilization of glucose in the cells and regeneration of β-cells of the pancreas.

The study was designed to evaluate the glucose and cholesterol lowering effect of the aqueous extract of Centella asiatica leaf using the alloxan-induced diabetic rats and compared the activity with diabetic control and antidiabetic drug (Glibenclamide). Leaf extract (50 mg/kg) of C. asiatica and Glibenclamide were administered to normal and experimental diabetic rats for the duration of 10 days. In the alloxan-induced diabetic rat model, C. asiatica extract (50 mg/kg) significantly (p < 0.05) lowered the fasting blood glucose level as well as the total cholesterol level. Serum insulin levels were not stimulated in the animals treated with the extract. In addition, changes in body weight, serum lipid profiles and liver glycogen levels assessed in the extract treated diabetic rats were compared with diabetic control and normal animals. Significant results (p < 0.05) were observed in the estimated parameters. Surprisingly, body weight was increased significantly (p < 0.05) in the C. asiatica treated diabetic group. Diabetic complications occur as a result of increased reactive oxygen species (ROS) due to long term hyperglycaemia. Honey and ginger have been shown to exhibit antioxidant activity which can scavenge ROS. The main aim of this study was to evaluate the antioxidant and antidiabetic effects of gelam honey, ginger, and their combination. Sprague-Dawley rats were divided into 2 major groups which consisted of diabetic and nondiabetic rats. Diabetes was induced with streptozotocin intramuscularly (55 mg/kg body weight). Each group was further divided into 4 smaller groups according to the supplements administered: distilled water, honey (2 g/kg body weight), ginger (60 mg/kg body weight), and honey + ginger. Body weight and glucose levels were recorded weekly, while blood from the orbital sinus was obtained after 3 weeks of supplementation for the estimation of metabolic profile: glucose, triglyceride (TG), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), reduced glutathione (GSH): oxidized glutathione (GSSG), and malondialdehyde (MDA). The combination of gelam honey and ginger did not show hypoglycaemic potential; however, the combination treatment reduced significantly (P<0.05) SOD and CAT activities as well as MDA level, while GSH level and GSH/GSSG ratio were significantly elevated (P<0.05) in STZ-induced diabetic rats compared to diabetic control rats.

The present study was carried out to evaluate the antidiabetic activity ofVinca roseamethanolic whole plant extracts in alloxan induced diabetic rats for 14 days. The methanolic whole plant extract at high dose (500 mg/kg) exhibited significant anti-hyperglycemic activity than whole plant extract at low dose (300 mg/kg) in diabetic rats. The methanolic extracts also showed improvement in parameters like body weight and lipid profile as well as regeneration ofβ-cells of pancreas in diabetic rats. Histopathological studies reinforce the healing of pancreas, by methanolicVinca roseaextracts, as a possible mechanism of their antidiabetic activity.

The hot water and ethanol extracts of Andrographis paniculata (local name Kalomegh) collected from Chittagong exhibited a significant hypoglycemic (blood glucose lowering) activity in both glucose-loaded and alloxan-induced diabetic rats. Oral administration of glucose (1.5 g/kg body weight) increased the blood sugar level while the intraperitonial (ip) administration of alloxan (40 mg/kg body weight) enhanced the blood sugar level much higher than that of the glucose-loaded rats. The hot water (0.8 g/kg b.w.) and ethanol extracts (2 g/kg b.w.) of A. paniculata reduced the elevated glucose level by 41.51 and 41.82%, respectively in glucose-loaded rats as compared to the respective diabetic control rats. On the other hand, administration of hot water and ethanol extracts of A. paniculata decreased the blood sugar level by 46.21 and 45.13%, respectively in alloxan-induced diabetic rats, when compared with that of diabetic control rats. AbstractThe aims of the present study were to assess whether the antidiabetic activity of mature coconut water (MCW) is mediated through L-arginine-nitric oxide pathway in diabetic rats, and to study the effects of MCW on blood coagulation.Diabetes was induced in male Sprague-Dawley rats by injecting them with alloxan (150 mg/kg body weight). MCW (4 mL/100 g body weight) and L-arginine (7.5 mg/100 g body weight) was given orally for 45 days. L-NAME was given at a dose of 0.5 mg/kg body weight. Concentrations of blood glucose, plasma insulin, glycosylated hemoglobin (HbA: Treatment with MCW and L-arginine reduced the concentration of blood glucose and HbAThe results clearly indicate that L-arginine is a major factor responsible for the antidiabetic and antithrombotic potential of coconut water, and is mediated through the L-arginine-nitric oxide pathway.

Aim: The present study was designed to investigate the antidiabetic & hypolipidemic activity of Calotropis gigantean (Family: Apocynaceae) in alloxan-induced diabetic rat model. Study Design: In vivo study was carried out by ethanolic leaf extract was administered in 250 mg/kg body weight concentration and then subjected to different rats models to authenticate the antidiabetic and hyperlipidimic properties of the plant. Place and Duration of Study: Department of Pharmacy, Southeast University, Banani, Dhaka-1213,Bangladesh within a period of July 2018 to December, 2018. Methodology: Diabetes was induced in rats by an intraperitoneal injection (i.p) of alloxan (100 mg/kg B.W). Ethanolic leaf extract of C. gigantean (250 mg/kg B.W) was administrated orally as a single dose per day to the diabetic rats for 7 days. The negative control group received 0.5 ml of sterile normal saline water orally & positive control group received metformin orally. Synergistic effect of plant was evaluated by combination with 100 mg/kg B.W & 50 mg/kg B.W oral administration of metformin. After 7 days study period, fasting blood glucose, total cholesterol, triglyceride, high-density lipoprotein cholesterol, liver weight & body weight were measured only for diabetic group to observe the effects of diabetes induction. Results: Individual plant extract (250 mg/Kg B.W) & Metformin (100 mg/kg B.W) reduced FBG significantly by 52% (P<0.001) & 55.3% (P<0.001) correspondingly. Metformin (100 mg/kg B.W) potentiated reduction (68%) (P<0.001) when combined to plant extract (250 mg/Kg B.W). Significant dose dependent manner was followed when metformin (50 mg/kg B.W) was combined to plant extract (250 mg/Kg B.W). Our results clearly suggests that C. gigantean exhibit hypoglycemic & hypolipidemic activity with an alteration in body-liver weight. The present study also suggested to develop a combination therapy of extract along with metfromin in different doses to minimize the intake of synthetic drug. Significant reduction of TG, TC were noted by extract (250 mg/kg B.W) with 32.42% (P<0.001) & 41.32% (P<0.001) respectively where standard shown the diminution 43.43% (P<0.05) & 47.21% (P<0.001) respectively as compare to Untreated diabetic rats. 50.21% (P<0.01) & 42.38% (P<0.001) reduction of TG & TC were estimated by C.gigantea extracts (250 mg/kg B.W) when combined with Metformin (100 mg/kg B.W). 34.53% (P<0.05) & 41.54% (P<0.001) reduction of TG & TC by C.gigantea extracts (250 mg/kg B.W) were confirmed when combined to Metformin (50 mg/kg B.W). Combination therapy also has shown synergistic effect in elevation of plasma HDL-cholesterol. Conclusion: The results of the study concluded that C. gigantean have potential antidiabetic and antioxidant properties.

The present study was carried out to evaluate antidiabetic and antihyperlipidemic effects of Dillenia indica methanolic leaves extracts in streptozotocin induced diabetic Wistar rats by administering graded oral doses (250 and 500 mg/kg body weight) for 21 days. The extract showed significant antidiabetic activity (p<0.001). Furthermore, the decreased body weight of rats was significantly improved after extract treatments. Daily oral treatment with the extract for 21 days to diabetic rats, also resulted in significant reduction in serum cholesterol, triglycerides and serum transaminase levels but HDL-cholesterol level was found to be improved (p<0.001) as compared to the diabetic control group. The extract treatment also showed to enhance serum insulin level in diabetic rats as compared to the diabetic control group. In conclusion, D. indica leaf extract might be useful for diabetes mellitus management and other abnormalities associated with this metabolic disorder.

Study was conducted to evaluate the hepatoprotective effects of Seabuckthorn leaf extract (SLE) supplementation on serum enzymatic levels in streptozotocin (STZ) induced diabetes mellitus in Wistar rats. Thirty-two adult male Wistar rats were divided into four groups namely CON (negative control), SCO (Seabuckthorn control), DCO (Diabetic control), and DSL (Diabetic seabuckthorn treatment group). Diabetes mellitus was induced by single intra peritoneal injection of STZ @ 50 mg/kg body weight in DCO and DSL group of rats. SLE was administered orally @ 100mg/kg body weight for 40 days to SCO and DSL groups. CON served as the negative control. Blood samples were collected from experimental animals on zero, 20th, and 40th days of trial to study liver specific serum enzyme profile viz aspartate amino transaminase (AST), alanine amino transaminase (ALT), alkaline phosphatase (ALP) and acid phosphatase (ACP). Significantly (P less than 0.01) higher levels of all the enzymes studied were observed in experimentally induced diabetic rats in comparison to normal rats. However, in SLE treated diabetic rats (DSL group), significant (P less than 0.01) improvement was observed in all the above enzymes. It may be concluded that SLE exerts hepatoprotective effect in STZ induce Diabetes mellitus in Wistar rats.

Background: Ficus glomerata (F. glomerata) Linn. Family Moraceace is a large tree found all over India including outer Himalayan ranges, Punjab, Chota Nagpur, Bihar, Orissa, West Bengal, Rajasthan, Deccan and also as a common plant in South India. It is planted around the home and temples. It is cultivated throughout the year, distributed in evergreen forests and moist localities. Objective: The Ethanolic Extract of roots of F. Glomerata (EEFG) belonging to the family Moraceace, was investigated for its antidiabetic activity using alloxan induced diabetic rats. Methods: Thirty rats were divided into 5 groups having 6 rats in each group. The alloxan was administered to the rats of all groups except normal control group through intraperitoneal route at a concentration of 140mg/kg body weight. A dose of 100mg/kg and 200 mg/kg body weight of EEFG was administered to alloxan induced diabetic rats. The administration of the extract was lasted for 11 days. Effectiveness of the extract on glucose, cholesterol, triglycerides, and high density lipoprotein and protein concentrations was analyzed. Results: Significant (p<0.05) reduction in the levels of glucose, cholesterol, triglyceride of the diabetic rats was observed after treatment with ethanolic extract. After subjecting to oral glucose tolerance test EEFG also showed significant improvement in glucose tolerance. Conclusion: F. glomerata root ethanolic extract showed that it possesses antidiabetic effect and can be found useful for the management of diabetes mellitus.

Previous studies found that Kelulut Honey produced by Trigona spp. bees is able to prevent oxidative damage in various pathological conditions.  Thus, the present study aimed to determine whether Kelulut Honey could prevent the sperm and testicular damage in streptozotocin-induced diabetic rats. Male Adult male Sprague-Dawley rats were divided into four groups: Non-Diabetic (NDM), Non-Diabetic with Kelulut Honey supplementation (NDMKH), Diabetic without supplementation (DM) and Diabetic with Kelulut Honey supplementation (DMKH).  Kelulut honey was given at the dose of 2.0 g/kg weight daily via gavage for 28 consecutive days. Results showed that sperm quality produced by diabetic rats supplemented with Kelulut honey significantly improved compared to the diabetic control groups (p<0.05). SOD activity and GSH level increased significantly (p<0.05) whereas PC and MDA levels significantly decreased in sperm and testis of DMKH rats when compared to DM rats (p<0.05). Histological observation showed obvious increase in spermatozoa in the lumen of epididymis and increased spermatogenic cells density in the testis of DMKH group.  In conclusion, Kelulut Honey has a potential in preventing the damage of sperm and testis in diabetic rats.
