Boehmeria glomerulifera Miq. is an indigenous plant of Bangladesh. Traditional healers use this plant to treat various diseases. The crude methanol extract of whole plant of B. glomerulifera and its Kupchan fractions were screened for in vitro membrane stabilizing and only the crude extract at doses of 200- and 400- mg/kg body weight were screened for analgesic activity in mice model in order to establish the medicinal values of this plant. The membrane stabilizing activity of the extractives was evaluated by the ability of test materials to inhibit hypotonic solution- and heat-induced haemolysis of human erythrocytes. Moreover, the analgesic activity of methanol extract was evaluated by acetic acid induced writhing method and tail immersion method in mice. In hypotonic solution- and heat-induced conditions, the crude methanol extract showed inhibition of haemolysis by 64.80±0.34% and 21.63±0.76%, respectively as compared to 77.74±0.67% and 40.41±0.69% demonstrated by the standard, acetyl salicylic acid. On the other hand, the analgesic activity was determined for its peripheral and central pharmacological responses using acetic acid-induced writhing test and tail immersion method at doses of 200- and 400- mg/kg body weight. The extract reduced the acetic acid-induced writhings significantly (p < 0.05) in a dose dependent manner with the highest activity observed at 400 mg/kg (46.0%) body weight when compared to that of Diclofenac sodium (64.0%), as the standard drug. A significant increase (p < 0.01) of latency period was also found in tail immersion method. Therefore, the present study established that the methanol extract of B. glomerulifera possesses moderate membrane stabilizing and significant analgesic activities.Bangladesh Pharmaceutical Journal 19(2): 185-189, 2016

The hexane (HE), dichloromethane (DCME), ethyl acetate (EAE) and methanol (ME) extracts of seed and stem of Anethum sowa were subjected to screenings for analgesic, anti-diarrheal and anti-depressant activities. The peripheral and central analgesic actions were determined by using formalin-induced writhing test and tail immersion method. The DCME extract of stem, at a dose of 200 mg/kg body weight, significantly reduced the number of writhing movements whereas the methanol extract of seed at the same dose exhibited remarkable analgesic activity in tail immersion method. In castor oil induced anti-diarrheal assay, the ethyl acetate extract of stem, at 400 mg/kg body weight, exhibited significant anti-diarrheal effect. Lasia spinosa Linn. (Family: Araceae) is an important medicinal plant, which is traditionally used for treatment of different human ailments. The present study was undertaken to evaluate the in vitro thrombolytic, antiinflammatory and in vivo analgesic and hypoglycemic potentials of n-hexane, chloroform and aqueous soluble fractions of methanol extract of L. spinosa whole plant. Additionally, phytochemical screening was carried out by qualitative tests, which confirmed the presence of alkaloids, glycosides, steroids, tannins, saponin in this plant. During in vitro thrombolytic assay, the aqueous fraction at a dose of 500 μg/100 μl showed the maximum 33.15% lysis of the blood clot, as compared to the standard streptokinase (80.10%). The in vitro anti-inflammatory test was performed by inhibition of egg albumin denaturation assay and RBC membrane stabilization method. The chloroform fraction exhibited maximum anti-inflammatory potential by inhibiting 51.53% denaturation of albumin and by inhibiting 54.8% hemolysis of RBC membrane against hypotonic solution. Analgesic activity was evaluated by tail immersion method for central mechanism and by formalin-induced lick test for peripheral mechanism in mice. In tail immersion method, all the solvent fractions of L. spinosa at a dose of 500 mg/kg body weight exhibited a significant (p<0.05) elongation in pain reaction time. In peripheral analgesic activity test, the chloroform fraction at a dose of 500 mg/kg body weight inhibited a maximum of 35.44% licking response induced by formalin, as compared to the standard aspirin (53.22%). In the hypoglycemic activity test, all the fractions showed a moderate effect in reducing the blood glucose level in mice treated with 10% glucose. As there is a resurgence of interest in plants as promising sources of new active pharmaceutical constituents, the present study has been designed to establish the preliminary biological activities of Blumea clarkei Hook.f. leaves. Here, the crude methanol extract of the leaves along with its organic and aqueous soluble fractions were subjected to different in vitro and in vivo assays. The ethyl acetate soluble fraction exhibited the highest total phenolic content (71.59 mg of GAE/gm of extract) and DPPH free radical scavenging (IC50 value 19.25 μg/ml) whereas another polar fraction; the aqueous soluble extractive showed the maximum cytotoxicity (LC50 value of 18.60 μg/ml) in the brine shrimp lethality assay. In membrane stabilizing activity evaluation, the inhibition of hypotonic solution and heat induced hemolysis were revealed maximum by the DCM (30.64%) and aqueous soluble fraction (39.15%), respectively while the crude methanol extract exhibited the highest thrombolytic potential (30.47% clot lysis). The central analgesic activity of the crude extract significantly increased the latent response time in tail-immersion method (p<0.001) at doses 200 and 400 mg/kg of body weight after 90 minutes of administration when compared to the control group. In acetic acid-induced writhing method, the plant extract showed prominent peripheral analgesia (p<0.001) with 47.54% and 65.57% inhibition at 200- and 400-mg/kg of body weight, respectively. The crude methanol extract of Gouania tiliaefolia Lam. was partitioned by the modified Kupchan method and the fractions were evaluated for total phenolic content, antioxidant, cytotoxic, thrombolytic, hypotonic and heat-induced membrane stabilizing activities. The petroleum ether soluble fraction (PESF) and methanol extract (ME) showed the highest phenolic content of 78.30 ±1.60 mg and 70.37 ± 0.84 mg, respectively, which were expressed in gallic acid equivalent (GAE). Similar trends were observed in case of anti-oxidant and cytotoxic activities, where the PESF possessed the highest free radical scavenging activity and brine shrimp lethality (IC50 = 2.88 ± 0.02 μg/ml, LC50 = 2.59 ± 0.14 μg/ml), followed by ME (IC50 = 4.79 ± 0.17 μg/ml, LC50 = 3.38 ± 0.08 μg/ml) and CSF (IC50 = 37.51 ± 0.96 μg/ml, LC50 = 73.55 ± 0.26 μg/ml). In case of assays for thrombolytic and membrane stabilizing activities, all extractives showed insignificant results compared to the respective standards. The crude methanol extract of G. tiliaefolia was used to examine the in-vivo analgesic (central and peripheral), antidiarrheal and antidiabetic activities in Swiss albino mice. In case of castor oil induced diarrhea, the ME gave better reduction of diarrhea by 71.43% (at 400 mg/kg-body weight) compared to loperamide (64.29%). Antidiabetic activity was evaluated by oral glucose tolerance test and the ME showed 71.42% and 75.39% reduction of blood glucose at doses 200 and 400 mg/kg-body weight, respectively when compared with the standard glibenclamide that reduced blood glucose by 66.17%. The central- and peripheral-analgesic activity was evaluated by the tail-flick test and acetic acid induced writhing test, respectively. Background: Polyalthia simiarum (Hook. F. &Thomson) exhibits different effects in human body. Objective: As a part of ongoing research on medicinal plants of Bangladesh, the present study is focused to investigate the analgesic and anti-inflammatory activities of stem bark of Polyalthia simiarum (Annonaceae). Methodology: The ethyl acetate (EA) and petroleum ether (PE) extracts were subjected to qualitative chemical investigation for the identification of different phytoconstituents. The analgesic activity was determined for its central and peripheral pharmacological actions using tail immersion method and acetic acid-induced writhing test. The anti-inflammatory activity was evaluated by carrageenan induced paw edema in rats. Analgesic and anti-inflammatory data were evaluated statistically analysed by Dunnett’s-T test. Result: Both extracts at the dose of 50- and 100 mg/kg b.w., produced significant increase in pain threshold in tail immersion method whereas significantly reduced the writhing caused by acetic acid in a dose dependent manner. The EA and PE extracts showed anti-inflammatory activities at 50- and 100 mg/kg body weight. Among all the extracts, the EA extract showed a dose dependent and comparable analgesic activity in all the tested methods and also reduced the paw edema considerably (27.5% and 39.1% inhibition after 4h), in dose dependent manner when compared to carrageenan induced control rat. Conclusion: Therefore, the EA and PE extracts of Polyalthia simiarum were capable to exhibit moderate analgesic and anti-inflammatory activities. This is the first report of analgesic and anti-inflammatory potential of Polyalthia simiarum and can be further investigated to isolate the active compounds responsible for the biological activities. The methanol extract of leaf of Podocarpus neriifolius D. Don exhibited in vivo peripheral analgesic and antidiarrheal activities in Swiss Albino mice. In the peripheral analgesic activity assay, the methanolic extract showed 50.00 ± 8.57% and 70.25 ± 1.18% inhibition of acetic acid-induced writhing at 200 and 400 mg/kg body weight, respectively. The methanol extract of leaves of Picrasma javanica Blume and its n-hexane, carbon tetrachloride, chloroform and aqueous soluble partitionates were evaluated for membrane stabilizing and preliminary hypoglycemic activities. In hypotonic solution and heat-induced conditions, the crude methanol extract inhibited 65.30±0.52% and 36.79±0.38% haemolysis of RBC as compared to 72.79% and 42.12% produced by standard drug, acetyl salicylic acid, respectively (0.10 mg/ml). The crude extract also showed blood sugar lowering efficacy at 200 and 400 mg/ kg body weight, with 48.63% reduction of blood sugar level at 400 mg/kg body weight while the standard drug, Glibenclamide (10 mg/kg body weight) demonstrated 53.7% reduction of blood sugar level at 3rd hour of administration. The sunflower seed is the seed of the sunflower (Helianthus annuus). The methanol extract of seeds of Helianthus annuus were screened for analgesic activity in mice model to systematically explore the medicinal values of the plant. Acetic acid induced writhing and hot plate methods were used to confirm the central and peripheral analgesic action. In case of acetic acid-induced writhing test the extract showed significant (P <0.05) analgesic potential at doses 100 and 200 mg/kg body weight (50.35 and 57.85% inhibition, respectively). In the hot plate method, increase (p < 0.05) of latency period was also observed in comparison to standard aspirin. At 60 minutes, the latency period of two different doses (100 and 200 mg/kg body weight) was found at 13 ± 0.91 and 16.5 ± 1.55 second. Brassica rapa subsp. chinensis (L.) Hanelt is a popular herb in Bangladesh. In the present studies, the plant has been assessed to evaluate its in vitro thrombolytic and membrane stabilizing properties. Initially, the plant was collected, processed and extracted with methanol and the concentrated crude methanol extract was fractionated into petroleum ether, carbon tetrachloride, chloroform and aqueous soluble materials. All extractives, including the parent extract were subjected to thrombolytic and membrane stabilizing bioassays. In case of thrombolytic study, the carbon tetrachloride soluble fraction exhibited highest clot lysis. Here, streptokinase and distilled water were used as positive and negative control, respectively. However, in the assay for membrane stabilizing activity, the crude methanol extract along with other extractives were capable to inhibit hemolysis of erythrocyte membrane in hypotonic solution- and heat- induced conditions, which indicated anti-inflammatory property of the samples. Acetyl salicylic acid was used as standard drug in this assay. This represents the first report of thrombolytic and membrane stabilizing activities of B. rapa subsp. chinensis. In order to systematically explore the medicinal values of this plant, the crude methanol extract of leaves of A. dichotoma were screened for analgesic and antidiarrheal activities in mice model. The peripheral and central analgesic actions were determined by using acetic acid-induced writhing and tail immersion methods. The extract significantly (p < 0.05) attenuated the acetic acid-induced writhing in a dose dependent manner. A noticeable dose-dependent increase (p < 0.05) of latency period was also observed in the tail immersion method.
