Mosquitoes are the most important vectors for arthropod-borne viral diseases. Mixed viral infections of mosquitoes allow genetic recombination or reassortment of diverse viruses, turning mosquitoes into potential virologic mixing bowls. In this study, we field-collected mosquitoes of different species (Aedes aegypti and Culex pipiens complex), from different geographic locations and environments (central Europe and the Caribbean) for highly sensitive next-generation sequencing-based virome characterization. We found a rich virus community associated with a great diversity of host species. Among those, we detected a large diversity of novel virus sequences that we could predominately assign to circular Rep-encoding single-stranded (CRESS) DNA viruses, including the full-length genome of a yet undescribed Gemykrogvirus species. Moreover, we report for the first time the detection of a potentially zoonotic CRESS-DNA virus (Cyclovirus VN) in mosquito vectors. This study expands the knowledge on virus diversity in medically important mosquito vectors, especially for CRESS-DNA viruses that have previously been shown to easily recombine and jump the species barrier.

ABSTRACT Gynandromorphic insects bear both male and female characteristics. In mosquitoes, gynandromorphy has been documented in multiple species and it can include many different combinations of sex-specific traits such as plumose or filiform antennae, short or elongated palpi, and male or female terminalia. Here we report 4 gynandromorphic specimens of Culex erythrothorax, Cx. pipiens complex, Cx. tarsalis, and Culiseta incidens collected in the Northern San Joaquin Valley of California. Each of the specimens exhibited heads with female characteristics of short palpi and filiform antennae while displaying fully developed external male terminalia including basistyles and dististyles. The potential implication of gynandromorphism in genetic control of mosquito vectors is discussed.

Mosquitoes collected in Germany in 2016, including Culex pipiens pipiens biotype pipiens, Culex torrentium and Aedes albopictus, as well as Culex pipiens pipiens biotype molestus (in colony since 2011) were experimentally infected with Zika virus (ZIKV) at 18 °C or 27 °C. None of the Culex taxa showed vector competence for ZIKV. In contrast, Aedes albopictus were susceptible for ZIKV but only at 27 °C, with transmission rates similar to an Aedes aegypti laboratory colony tested in parallel.

Aedes aegypti y Culex pipiens se encuentran entre las principales especies de mosquitos vectores de enfermedades a nivel mundial. Ambas, son de gran importancia sanitaria en la Argentina. Aedes aegypti, es el principal vector del dengue y la fiebre amarilla. Está presente en la Argentina desde 1983, cuando fue confirmada su reemergencia en nuestro país. A partir de entonces comenzaron a detectarse casos de dengue hasta que el último brote de esta enfermedad en 2009 produjo 24.720 casos autóctonos confirmados por la OPS a nivel nacional. Culex pipiens es vector de algunas filariasis, y de importantes arbovirosis como la ocasionada por el virus del Nilo Occidental. La actividad vectorial de este mosquito se ve incrementada en zonas donde la densidad de zanjas de desagüe domiciliario es elevada, ya que este tipo de ambiente constituye un lugar propicio para su desarrollo. De su importancia sanitaria nace la necesidad de controlar su actividad vectorial, manteniendo las poblaciones en bajas densidades. El objetivo de este trabajo fue desarrollar estrategias para el control de A. aegypti y C. pipiens, basadas en métodos biológicos, eficaces, permanentes y seguros para el ambiente, mediante el empleo de enemigos naturales presentes en las poblaciones de ambas especies. En este trabajo, fueron evaluados copépodos, turbelarios y peces como depredadores; y bacterias entomopatógenas (α-protobacterias y bacterias esporulantes). El estudio de los copépodos, tuvo como objetivo identificar a la comunidad de copépodos larvívoros que se desarrolla en los criaderos de mosquitos, ubicados en los alrededores de la ciudad de La Plata, para identificar nuevas especies capaces de depredar mosquitos que habitan recipientes artificiales (A. aegypti y C. pipiens). La diversidad de copépodos larvívoros fue mayor en los cuerpos de agua permanentes. Acanthocyclops robustus, Diacyclops uruguayensis, Macrocyclops albidus y Mesocyclops longisetus fueron seleccionados por su capacidad depredadora. En el laboratorio fueron evaluadas: la capacidad de depredación de ambos sexos y diferentes estadios, preferencia de especie presa, y la tasa de depredación diaria. Las hembras de estas especies de copépodos presentaron mayor capacidad depredadora. No se detectó preferencia de los copépodos hacia ninguna de las especies de mosquitos. También se evaluó la tolerancia a la desecación y la capacidad de vivir en el agua que se acumula en los recipientes artificiales. Diacyclops uruguayensis y A. robustus sobrevivieron en las condiciones de sequía ensayadas en este estudio, pero D. uruguayensis mostró una menor supervivencia en el agua de recipientes artificiales. Macrocyclops albidus no sobrevivió en condiciones de sequía ni toleró el agua extraída de los recipientes artificiales. La supervivencia de M. longisetus ante estas condiciones, fue reducida. Se concluyó que las especies D. uruguayensis y A. robustus podrían ser buenos candidatos para el control de especies de mosquitos que crían en recipientes artificiales.

Ligusticum sinense Oliv. cv. is a species of Umbelliferae (Apiaceae), a large plant family in the order Apiales. In this study, L. sinense hexane extract nanoemulsion gel (LHE-NEG) was investigated for mosquito repellency and compared to the standard chemical, N,N-diethyl-3-methylbenzamide (DEET), with the goal of developing a natural alternative to synthetic repellents in protecting against mosquito vectors. The results demonstrated that LHE-NEG afforded remarkable repellency against Aedes aegypti, Anopheles minimus, and Culex quinquefasciatus, with median protection times (MPTs) of 5.5 (4.5–6.0), 11.5 (8.5–12.5), and 11.25 (8.5–12.5) h, respectively, which was comparable to those of DEET-nanoemulsion gel (DEET-NEG: 8.5 (7.0–9.0), 12.0 (10.0–12.5), and 12.5 (10.0–13.5) h, respectively). Evaluation of skin irritation in 30 human volunteers revealed no potential irritant from LHE-NEG. The physical and biological stability of LHE-NEG were determined after being kept under heating/cooling cycle conditions. The stored samples of LHE-NEG exhibited some changes in appearance and differing degrees of repellency between those kept for 3 and 6 heating/cooling cycles, thus providing slightly shorter MPTs of 4.25 (4.0–4.5) and 3.25 (2.5–3.5) h, respectively, when compared to those of 5.0 (4.5–6.0) h in fresh preparation. These findings encourage commercially developed LHE-based products as an alternative to conventional synthetic repellents in preventing mosquito bites and helping to interrupt mosquito-borne disease transmission.

In the period from 2015 to 2020, an entomological survey for the presence of West Nile virus (WNV) and Usutu virus (USUV) in mosquitoes was performed in northwestern Croatia. A total of 20,363 mosquitoes were sampled in the City of Zagreb and Međimurje county, grouped in 899 pools and tested by real-time RT-PCR for WNV and USUV RNA. All pools were negative for WNV while one pool each from 2016 (Aedes albopictus), 2017 (Culex pipiens complex), 2018 (Cx. pipiens complex), and 2019 (Cx. pipiens complex), respectively, was positive for USUV. The 2018 and 2019 positive pools shared 99.31% nucleotide homology within the USUV NS5 gene and both clustered within USUV Europe 2 lineage. The next-generation sequencing of one mosquito pool (Cx. pipiens complex) collected in 2018 in Zagreb confirmed the presence of USUV and revealed several dsDNA and ssRNA viruses of insect, bacterial and mammalian origin.
