Australia has one of the world’s largest marine park estates. At 3.3 million km2, it spans an area three times larger than Germany, France, and the UK combined. Managing and monitoring such a vast and often remote area is logistically challenging and expensive. Current monitoring of Australian parks is decentralised and depends on traditional survey methods. As a result, real-time data on the state of Australia’s marine parks is incomplete, hampering effective management. Environmental DNA has been suggested as a potential solution to some of these challenges, but practical large-scale applications remain largely lacking in Australia. To overcome this, we are developing a roadmap towards integrating eDNA methods in marine park monitoring. We present an overview of the current state of marine monitoring in Australia marine, identify the aspects of bio-monitoring that eDNA can best contribute to, and suggest pathways towards best practice use of eDNA for resource managers in Australia and globally.

Global efforts to conserve declining amphibian populations have necessitated the development of rapid, reliable, and targeted survey methods. Environmental DNA (eDNA) surveys offer alternative or complementary methods to traditional amphibian survey techniques. The California Tiger Salamander Ambystoma californiense (CTS) is endemic to California where it breeds in vernal pools. In the past 25 years, CTS has faced a 20.7 percent loss of known occurrences, largely through habitat loss, and is threatened by hybridization with an introduced congener. Protecting and managing remaining CTS populations relies on accurately monitoring changes in their spatial distribution. Current monitoring practices typically employ dip-net surveys, which are time-consuming and prone to false negative errors. To provide a new resource for monitoring and surveying larval CTS, we designed an assay and tested it on environmental samples collected from 29 vernal pools at two locations in California. We compared eDNA results to contemporaneous dip-net surveying results and found 100% agreement on the presence/absence surveys of larval CTS. In several instances we also detected the presence of CTS genetic material in the early spring months before larvae hatched, potentially offering a new, earlier detection option for this imperiled species. This assay provides a valuable, non-invasive molecular tool for monitoring the spatial distribution of CTS in vernal pools.

AbstractIn recent decades, consensual approaches to poverty measurement have been widely adopted in large-scale survey research both in the UK and internationally. However, while ascertaining the extent of public agreement on the ‘necessities of life’ has been central to this approach, long-standing critiques have questioned the nature of public consensus on poverty derived using survey methods. By drawing on new primary research preparatory to the 2012 UK Poverty and Social Exclusion Survey, we consider the contribution of qualitative methods in understanding public views on necessities and discuss their implications for survey-based poverty measurement. Our findings raise some important conceptual and measurement issues for consensual poverty measures within large-scale social surveys. Firstly, our research suggests that public understandings of the term ‘necessity’ are diverse and may not always be consistent with researchers’ interpretations or with wider usage of this term within consensual poverty measurement. Secondly, a better understanding of the considerations which inform survey respondents’ deliberations is needed. Thirdly, our findings have important implications for how we should interpret the concept of ‘consensus’ within the context of consensual poverty surveys, and emphasise the need for the application of more deliberative methods in determining public views on the ‘necessities of life’.

There are various opportunities and challenges in the delivery of care to those diagnosed with chronic oedema/lymphoedema. Service provision is not consistent within the UK, and non-specialist nurses and other health professionals may be called on to fill the gaps in this area. The latest best practice guidance on chronic oedema is directed at community services that care for people within their own homes in primary care. This guide was developed in order to increase awareness, knowledge and access to an evidence base. Those involved in its creation cross specialist fields (lymphoedema and tissue viability), resulting in the document covering a number of areas, including an explanation of chronic oedema, its assessment and management and the association between chronic oedema and wet legs. The document complements existing frameworks on the condition and its management and also increases the available tools within chronic oedema management in the community. The present article provides an overview of the guidance document and discusses its salient features.

ABSTRACT Dolomite is a common and volumetrically important mineral in many siliciclastic sandstones, including Permian Rotliegend sandstones (the Slochteren Formation). These sandstones form extensive gas reservoirs in the Southern Permian Basin in the Netherlands, Germany, Poland, and the UK. The reservoir quality of these sandstones is negatively influenced by the content and distribution of dolomite. The origin and the stratigraphic distribution of the dolomite is not yet fully understood. The aim of this study is to identify the origin of carbonate. The main methods used to achieve those aims are a combination of thin-section petrography, scanning electron microscopy (SEM and EDX), and XRD analyses. The present study shows that the typical dispersed occurrence of the dolomite is a consequence of dispersed detrital carbonate grains that served both as nuclei and source for authigenic dolomite cement. The dolomite cement formed syntaxial outgrowths and overgrowths around detrital carbonate grains. The study also shows that dolomite cement, often in combination with ankerite and siderite, precipitated during burial after mechanical compaction. Most of the carbonate grains consisted of dolomite before deposition. The carbonate grains were affected by compaction and pressure dissolution, and commonly have no well-defined outlines anymore. The distribution of dolomite cement in the Rotliegend sandstones was controlled by the presence of stable carbonate grains. Due to the restricted and variable content of carbonate grains and their dispersed occurrence, the cement is also dispersed and the degree of cementation heterogeneous. Our findings have important implications on diagenesis modeling. The presence of detrital carbonate excludes the need for external supply by any large-scale advective flow of diagenetic fluids. By knowing that the carbonate source is local and related to detrital grains instead of being externally derived from an unknown source, the presence of carbonate cement can be linked to a paleogeographic and sedimentological model.

The UK’s COVID-19 epidemic during early 2020 was one of world’s largest and unusually well represented by virus genomic sampling. Here we reveal the fine-scale genetic lineage structure of this epidemic through analysis of 50,887 SARS-CoV-2 genomes, including 26,181 from the UK sampled throughout the country’s first wave of infection. Using large-scale phylogenetic analyses, combined with epidemiological and travel data, we quantify the size, spatio-temporal origins and persistence of genetically-distinct UK transmission lineages. Rapid fluctuations in virus importation rates resulted in >1000 lineages; those introduced prior to national lockdown tended to be larger and more dispersed. Lineage importation and regional lineage diversity declined after lockdown, while lineage elimination was size-dependent. We discuss the implications of our genetic perspective on transmission dynamics for COVID-19 epidemiology and control.

AbstractThe COVID-19 pandemic caused by SARS-COV-2 virus fortunately resulted in few children suffering from severe disease. However, the collateral effects on the COVID-19 pandemic appear to have had significant detrimental effects on children affected and young people. There are also some positive impacts in the form of reduced prevalence of viral bronchiolitis. The new strain of SARS-COV-2 identified recently in the UK appears to have increased transmissibility to children. However, there are no large vaccine trials set up in children to evaluate safety and efficacy. In this short communication, we review the collateral effects of COVID-19 pandemic in children and young people. We highlight the need for urgent strategies to mitigate the risks to children due to the COVID-19 pandemic. What is Known:• Children and young people account for <2% of all COVID-19 hospital admissions• The collateral impact of COVID-19 pandemic on children and young people is devastating• Significant reduction in influenza and respiratory syncytial virus (RSV) infection in the southern hemisphere What is New:• The public health measures to reduce COVID-19 infection may have also resulted in near elimination of influenza and RSV infections across the globe• A COVID-19 vaccine has been licensed for adults. However, large scale vaccine studies are yet to be initiated although there is emerging evidence of the new SARS-COV-2 strain spreading more rapidly though young people.• Children and young people continue to bear the collateral effects of COVID-19 pandemic

The purpose of the present study was to evaluate the performance of a low-cost commercial smartwatch, the Xiaomi Mi Band (MB), in extracting physical activity and sleep-related measures and show its potential use in addressing questions that require large-scale real-time data and/or intercultural data including low-income countries. We evaluated physical activity and sleep-related measures and discussed the potential application of such devices for large-scale step and sleep data acquisition. To that end, we conducted two separate studies. In Study 1, we evaluated the performance of MB by comparing it to the GT3X (ActiGraph, wGT3X-BT), a scientific actigraph used in research, as well as subjective sleep reports. In Study 2, we distributed the MB across four countries (Austria, Germany, Cuba, and Ukraine) and investigated physical activity and sleep among these countries. The results of Study 1 indicated that MB step counts correlated highly with the scientific GT3X device, but did display biases. In addition, the MB-derived wake-up and total-sleep-times showed high agreement with subjective reports, but partly deviated from GT3X predictions. Study 2 revealed similar MB step counts across countries, but significant later wake-up and bedtimes for Ukraine than the other countries. We hope that our studies will stimulate future large-scale sensor-based physical activity and sleep research studies, including various cultures.

All good quality trials of psychological interventions need to check formally that therapists have used the techniques prescribed in the published therapy manuals, and that the therapy has been carried out competently. This paper reviews methods of assessing adherence and competence used in recent large-scale trials of Cognitive Behaviour Therapy (CBT) for psychosis in the UK carried out by our research groups. A combination of the Cognitive Therapy Rating Scale and specific versions of the Cognitive Therapy for Psychosis Adherence Scales provides an optimal assessment of adherence and competence. Careful assessment of the competence and adherence can help identify the procedures actually carried out with individuals within trials. The basic use of such assessments is to provide an external check on treatment fidelity on a sample of sessions. Such assessment can also provide the first step towards moving research towards making sense of CBT for psychosis as a complex intervention and identifying which techniques work for which problems of people with psychosis, at which stages of disorder?
