Healthy and AY-affected plants of <em>L.sinuatum</em> have been propagated in vitro for 12 months on the media with and without cytokinins. In the contrary to the healthy plants the phytoplasma affected statice showed abnormal proliferation of the axillary shoots, shortening of the internodes, smaller leaves and severe chlorosis. On the medium without cytokinins, diseased plants proliferated and formed 7.0 axillary shoots per explant but the healthy ones only formed 2.3 shoots; however, the fresh weight of them was similar. On the media with cytokinins, the multiplication rate and fresh weight of healthy shoots greatly in-creased, but of the diseased plants were on the same laevel or decreased. During tissue culture phytoplasma could be detected in symptomatic plants by PCR as well as electron microscope however, the phytoplasmas showed the symptoms of degeneration.

The effect of white, blue, green, red and UV + white light on the growth and development of shoots and roots of Gerbera jamesonii cv. Queen Rebecca in relation to the presence of kinetin or IAA were investigated. The highest number of axillary shoots was obtained in red and green light on the medium with 5 mg l<sup>-1</sup> kinetin. Also, green and red light markedly increased the number of leaves developed on the plantlets on the medium supplemented with kinetin. Light quality and IAA added to culture medium variously affected the development of root system: roots were regenerated under all light treatments, higher root number was recorded under red light when 5 mg l<sup>-1</sup> IAA was added to the media, the shortest roots were found in red light on the medium supplemented with IAA. The greatest fresh weight of shoots was found under white light on the medium with kinetin. Red light markedly decreased shoot fresh weight on hormone-free medium. Blue and white light caused increase in fresh weight of roots.

<em>Helleborus niger</em> L. is a rhizomatous, herbaceous perennial with overwintering, divided, basal leaves. The objective of the study was to investigate the influence of different levels of sucrose (10, 20, 30, 40, 50, 60, 70, and 80 g l<sup>−1</sup>) and nitrogen salts (25%, 50%, and 100% according to MS medium) as well as temperature (15°C, 20°C) on in vitro multiplication and rooting and ex vitro acclimatization of <em>H. niger</em>. The growth and multiplication of axillary shoots were performed on modified MS medium supplemented with various growth regulators (2iP, BAP and kinetin – each at a concentration of 1.0 mg l<sup>−1</sup>, GA<span><sub>3</sub></span> 2.5 mg l<sup>−1</sup>). For the induction of roots, the medium was supplemented with IBA 1 mg l<sup>−1</sup> and NAA 0.1 mg l<sup>−1</sup>. Rooted plants were transplanted in a peat–perlite substrate (4:1) in a heated greenhouse for ex vitro acclimatization. The multiplication rate of <em>H. niger</em> shoots, in vitro rooting, and ex vitro acclimatization were strongly dependent on the sucrose/nitrogen salt relationship in the medium. The highest multiplication rate of axillary shoots (3.7) was found at a temperature of 15°C or 20°C, on the medium with cytokinins and GA<span><sub>3</sub></span> supplemented with sucrose 20–30 g l<sup>−1</sup> and nitrogen salts at 50%. Sucrose at a concentration of 50 g l<sup>−1</sup> strongly stimulated the number of roots per microplant (5.8–6.0) on the media with a reduced level of nitrogen salts (25% and 50%) when the temperatures were 20°C and 15°C, respectively. The plants rooted on the media with a high sucrose/nitrogen salt ratio showed acclimatization rates which ranged from 82% to 100%. Morphological observation of plantlets revealed obvious differences in leaf shape and size and the architecture of the root system as well as differences in the developmental stages of shoots grown on media with different sucrose and nitrogen salt concentrations.

The aim of work was to evaluate the influence of cytokinins on dahlia propagated in vitro and their consequent effect on acclimatization. Plant material were shoot tips and nodes. From three cytokinins, benzyladenine, kinetin and 2-isopentenyl-adenine, only BA effectively stimulated shoot multiplication from axiliary buds. The highest multiplication rate was obtained from nodes in presence of 0.25–0.5 mg·dm–3 BA. Higher concentrations shortened internodes and decreased leaf blades and growth of callus. 1 mg·dm–3 of KIN and 2iP positively influenced shoots growth and size of leaves. Gibberellic acid (GA3) used with BA increased the number of axillary shoots. The best quality shoots and the highest multiplication rate were obtained when 2 mg·dm–3 BA was used with 5 mg·dm–3 GA3. Cytokinins affected rooting and acclimatization ex vitro. Dahlias shoots multiplicated in presence of 1 mg·dm–3 KIN or 2iP rooted faster in the soil and 100% survived in field, while those from 1 mg·dm–3 BA media rooted slowly, had shorter shoots and only 60% survived. Plants bloomed after 11–12 weeks in the field. Dahlia plants that had been multiplicated in presence of KIN had a bigger diameter and fresh weight in the field. BA and 2iP positively influenced flowers diameter, length of flower stalk and a number of first order shoots.

Dimethylbenz(a)anthracene (DMBA)-induced and transplanted rat mammary tumours (2 lines) were examined for oestrogen receptor activity, and for sensitivity to hormones in vivo (by ovariectomy) and in vitro (by tissue culture). In vivo, the growth of all tumours induced by the administration of DMBA in random-bred Sprague–Dawley rats was found to be dependent on the ovary, whilst in all transplanted tumours (12 TG-3 and six TG-5 lines), maintained in an inbred strain of Sprague–Dawley rats, growth was found to be independent of the ovary. In vitro, the capacity for DNA synthesis in DMBA-induced tumours was better maintained after 24 h when insulin (10 μg/ml) and corticosterone (5 μg/ml) or insulin, corticosterone and prolactin (each 5 μg/ml) were present in the medium (five out of 12 and eight out of 11 tumours respectively); no effect of hormones in the media was detected after 48 h. In the transplanted tumours, no effect of hormones on DNA synthesis was detected after either 24 or 48 h of culture. Synthesis of lecithin was not detectably influenced by the presence of hormones in either DMBA-induced or transplanted tumours. Oestrogen receptor concentrations were, on average, significantly higher in the DMBA-induced tumours than in either line of transplanted tumour. For 22 DMBA-induced tumours and 15 transplanted tumours, the effect of hormones in vitro (`response') was directly correlated with receptor concentration at time 0 (Spearman's ρ = + 0·59) and inversely correlated with the rate of DNA synthesis (`basal') at time 0 (Spearman's ρ = −0·62). No single parameter or pair of parameters permitted accurate distinction between the tumour types.

The search for the occurrence of somaclonal variation of in vitro shoots and acclimatized plants of a hybrid larch (Larix × urolepis Henry) clone was performed by the analysis of eight isozyme systems. Cultures were established from short shoot buds of mature material. The effects of growth regulators in the media, subculture intervals, and periods in culture were analyzed for in vitro shoots. Variability was found in in vitro shoots but appeared to be related to a physiological response to culture conditions. Once acclimatized, most tissuecultured plants expressed the same enzymatic patterns as those of control plants (stecklings and the ortet). The variations observed for some acclimatized plants were also observed in control plants and were not related to ontogenic stage. Results from the isoenzymatic systems studied showed that hybrid larch plants regenerated from tissue culture were not significantly different from stecklings and the ortet.

In vitro tuberization was induced on explants with different number of nodes layered on a medium with high sucrose (8%) content: 30, 15, 10, 7 and 6 explants per jar were cultured containing 1, 2, 3, 4 or 5 nodes, respectively. Microtubers developed were graded by their smallest diameter, and the number of tubers per jar, their size distribution, their fresh weight and the multiplication rate were recorded. The highest multiplication rate (1.98) was obtained for explants with 5 nodes. The size distribution of tubers was markedly affected by treatments. The majority of microtubers (49.4%) were 6-8 mm in the case of the smallest explants (with I node). When explants with 2 to 5 nodes were used, the most microtubers were 8-10 mm but with an increase of explant size, more and more microtubers were produced with larger diameter up to 16 mm and average fresh weight of tubers also increased with the increase of explant size. For the microtuber production of Desiree the use of explants with two nodes can be suggested because in this treatment the average fresh weight of microtubers was high enough (250 mg) and the number of large sized microtubers was very high (79% was larger than 6 mm and 53% was larger than 8 mm).

In vitro tissue culture is a growth optimization technique of Dendrobium orchid with according to media composition. Nutritions in the media are important for dendrobium orchid. Dendrobium orchid  include plant from orchidaceae family its spread throughout the world like indonesia. Its features are easily planted, intersest is continuous and varied, easily assembled, the flower crown is not easy to fall and wither. Research aimed at obtaining media compositions that are easily available and able to fulfill the needs of orchid plants. The research was conducted using descriptive methods to compare different trearment; Vacin & Went and VW media with adding organic matter; extract bean sprouts, potato extrac, and water coconut; wich is conducted for eight weeks after planting. The result of addition organic matter on VW media was different toward organogenesis of orchid. The average number of shoots is 1.8; the number of leaves average of 6.8 and the number of roots average of 3.6 formed from two until eight weeks after culture.Keywords: tissue culture, growing media, Dendrobium orchid, organogenesis.ABSTRAKKultur jaringan in vitro adalah salah satu teknik optimalisasi pada pertumbuhan tanaman angrek Dendrobium dengan menyesuaikan komposisi medianya. Anggrek Dendrobium termasuk tanaman dari keluarga Orchidaceae yang penyebarannya sampai ke pelosok dunia seperti Indonesia. Keistimewaanya mudah ditanam, bunganya terus-menerus dan bermacam-macam, mudah disusun, serta mahkota bunga tidak mudah jatuh dan layu. Penelitian yang bertujuan untuk mendapatkan komposisi media yang mudah didapat dan mampu memenuhi kebutuhan tanaman anggrek. Penelitian dilakukan menggunakan metode deskriptif untuk membandingkan perlakuan media yang berbeda yaitu media Vacin & Went, dan VW dengan penambahan bahan organik; ekstrak tauge kacang hijau, ekstrak kentang, dan air kelapa muda; yang dilakukan selama delapan minggu setelah tanam. Hasil penambahan bahan organik pada media VW berbeda terhadap organogenesis eksplan anggrek. Jumlah tunas rata-rata 1,8; Jumlah daun rata-rata 6,8 dan jumlah akar rata-rata 3,6 yang terbentuk dari dua minggu setelah kultur (MSK) sampai minggu terakhir pengamatan delapan MSK.Kata kunci: kultur jaringan, media tanam, angrek Dendrobium,organogenesis.

Initiation of tissue culture of many plant species is a very difficult stage due to appearance of many contaminations. The other problem might be a choice of media for regeneration. Initiation of grass species tissue cultures are thought to be very difficult. Therefore, a research was undertaken to evaluate the use of nano-silver particles for plant material disinfection and to estimate a medium Pennisetum alopecuroides. The plant material were buds and nodal explants that were disinfected in 2% NaOCl for 30 min or 0.1% HgCl2 for 1 min. Half of the explants disinfected with NaOCl were soaked in 50, 100 or 250 mg·dm Ag–3 NPs for 1 hour. Explants not soaked in nano-silver were placed on media with Ag NPs at concentrations of 4, 8 or 16 mg·dm–3. An influence of growth regulators on Pennisetum alopecuroides was evaluated in vitro. Regenerated shoots were placed on MS media with: 3 mg·dm–3 BA + 0.3 mg·dm–3 IBA, 3 mg·dm–3 KIN + 0.3 mg·dm–3 IAA, 1 mg·dm–3 BA + 0.1 mg·dm–3 IBA. It was observed that the use of nano-silver particles lowered the level of contamination. The best results were obtained when Ag NPs was used at concentration of 100–250 mg·dm–3 alone or as a supplementation of the media, at concentration of 4 mg·dm–3 for nodes and 16 mg·dm–3 for adventitious buds. The use of nodal explants allowed to obtain less contamination. Regeneration depended on a media content. The most regenerated shoots were obtained on the MS media supplemented with 1 mg·dm–3 BA and 0.1 mg·dm–3 IBA.

Tanaman bawang merah (Allium ascolonicum L) termasuk dalam genus Allium sp yang diperbanyak secaravegetatif melalui umbi. Perbanyakan benih bawang sudah dilakukan secara in vitro (konvensional), untuk tujuanpeningkatan mutu atau hanya perbanyakan tanaman . Penelitian dilaksanakan di Laboratorium Kultur JaringanBalai Penelitian Tanaman Sayuran, pada bulan Februari sampai Agustus 2014. Untuk menghasilkan tanamanbebas penyakit terutama virus dapat digunakan teknik kultur jaringan yang dikombinasikan dengan perlakuanpemanasan. Penelitian bertujuan untuk melihat pengaruh pemanasan dan sumber eksplan terhadap pertumbuhandan perkembangan eksplan bawang merah.. Perlakuan pemanasan bahan eksplan bawang merah dilakukan secarabertahap selama 4 minggu, masing-masing 1 minggu untuk suhu (30, 33, 35 dan 37 oC). Media yang dipergunakanuntuk penumbuhan eksplan adalah MS + MS vits + sucrose 30 g/l + IAA 2 mg/l + Kinetin 2 mg/l + GA3 0.01mg/l agar gelgro 2 g/l pH 5.7. Penelitian terdiri dari 2 kegiatan yaitu Perlakuan pada cv. Pikatan, dan pada cv.Bima Brebes. Sebagai eksplan dipergunakan yaitu (1/3 bulb/ umbi) dan (shoot tip/ jaringan meristematik denganbeberapa daun primordia). Perlakuan eksplan yang digunakan yaitu tanpa pemanasan dan dengan pemanasan.Pertumbuhan dan perkembangan dari planlet diamati pada penelitian ini. Hasil dari penelitian menunjukkanbahwa perlakuan pemanasan bahan eksplan bawang merah cv. Bima Brebes, belum menurunkan persentase planletyang terinfeksi virus. Eksplan (1/3 bulb) dan (shoot tip), mempunyai pertumbuhan eksplan diatas 50%. Umumnyasemakin kecil eksplan persentase planlet abnormal semakin tinggi. Kontaminasi kultur umumnya disebabkanbakteri dan jamur yang terbawa dari eksplan (endogen). Perlakuan pemanasan bahan eksplan bawang merah secaravisual tidak berpengaruh pada persentase pertumbuhan dan persentase kultur terkontaminasi.Kata kunci: Bawang merah, Allium ascolonicum L, pemanasan, asal eksplanABSTRACTRed onion plants (Allium ascolonicum L) are included in the genus Allium sp which is propagatedvegetatively through tubers. Propagation of onion seeds has been done in vitro (conventionally), for the purposeof quality improvement or only plant propagation. The research was carried out at the Tissue Culture Laboratory,Balai Penelitian Tanaman Sayuran (Balitsa) from February to August 2014. To produce plants free of mainly viraldiseases can be used tissue culture techniques combined with heating treatment. Research aims were to look atthe effect of heating and explants on the growth and development of red onion explants. The treatment of the onionexplants was heated gradually for 4 weeks, 1 week each for temperatures (30, 33, 35 and 37 oC). The media usedfor the growth of explants is MS + MS vits + sucrose 30 g / l + IAA 2 mg / l + Kinetin 2 mg / l + GA3 0.01 mg / lso that gelgro 2 g / l pH 5.7. The study consisted of 2 activities, namely, treatment at cv. Pikatan, and at cv. BimaBrebes. As explants it is used (1/3 bulb / tuber) and (shoot tip / meristematic tissue with several leaves ofprimordia). The explants treatment used are without heating and by heating. Growth and development of plantletswas observed in this study. The results showed that the heating treatment of explants onions cv. Bima Brebes, hasnot reduced the percentage of plantlets infected with the virus. Explants (1/3 bulb) and (Shoot tip), have explantsgrowth above 50%. Generally the smaller explants the higher percentage of abnormal plantlets. Culturecontamination is generally caused by bacteria and fungi that are carried away from explants (endogenous). Thetreatment of heating the onion explants material visually had no effect on the percentage of growth and percentageof contaminated culture.Keywords: red onion, Allium ascolonicum L, heating treatment, source of explant

Poor growth of bulblet and ontogenic development are the major problems of adventitious bulb production of lilies in vitro through explant culture. Ontogenic development from the juvenile phase to the adult vegetative phase strongly correlates with bulblet growth of lily, and it is important to understand how lily bulblets grow in vitro. This study was at aimed to determining an effective in vitro culture process for lily to track bulblet growth, ontogenic development, and bulb production. In the results, bulblet performance was higher with the increase in the amount of medium; however, bulblet size was not satisfactory and was in the juvenile phase under an inducing condition. The threshold weight at which 100% ontogenic development was about 300 mg. Avoiding depletion of sucrose in the media is the key to achieving desired bulblet growth and ontogenic development in vitro and indicates growth could be sustained by subculture. The circumference of bulblets increases with its weight after subcultured in vitro. Therefore, a threshold circumference of about 3.2 cm in proportion to bulblet fresh weight of 300 - 350 mg may be an indicator of ontogenic development.
