Background: Diabetes Mellitus is a chronic disorder characterised by abnormally elevated glucose levels in the blood. Diabetes is caused by one of two mechanisms: insufficient insulin synthesis (which is produced by the pancreas and reduces blood glucose) or insufficient response of cells to insulin action. The current aim of this research project was to formulate and evaluate the Polyherbal preparation (PHP) of the plants constituted with Cinnamonum zeylanicium (CJ) bark, Eugenia jambolana (EJ) seeds, Vinca rosea (VR) whole plant, Gymnema sylvestre (GS) leaves and determination of the anti-diabetic potential of the formulation in the animal model induced by Streptozotocin. Methods: Plant components in the current study used were Cinnamonum zeylanicium (CJ) bark, Eugenia jambolana (EJ) seeds, Vinca rosea (VR) whole plant, Gymnema sylvestre (GS) leaves were collected.Using a hydroalcoholic solvent, physico-chemical parameters and active chemical constituents were evaluated. The active components present in the extracts were identified by Preliminary phytochemical screening. The PHP acute toxicity analysis was conducted in compliance with OECD Guideline 423, with 200 mg/kg and 4000 mg/kg administered orally to rats over 28 days. Results: Diabetes was induced by STZ and treated with PHF did not show any alterations in behavior and no mortality was observed up to the 2000 mg/kg dose level during the interventional period. By oral administration of PHP with a dosage of 200 and 400 mg/kg, OGTT resulted in a steady decrease in blood glucose levels of 68.74±4.63 mg/dl and 63.83±1.74 mg/dl at 180min after the trial which proves that PHP possess anti-diabetic activity. By mixing each extract in varying proportions, PHP was developed and evaluated. PHP (200 and 400mg/kg) antidiabetic activity wasdetermined for streptozotocin (STZ)-induced diabetes in rats and glibenclamide (5.0mg/kg body weight) was used as a standard drug.The investigational drug was administered for 28 days and the blood glucose level effect of the PHP was analysed on the 28th day after the intervention time. Conclusion: The experimental study showed that a persistent and substantial decrease in the average blood glucose level of diabetic rats was observed with repeated administration of PHP and glibenclamide for 28 days. PHP demonstrated substantial antidiabetic and antihyperlipidemic activity similar to the standard drug. The formulation will emerge as a possible mixture that may challenge the synthetic drug.

White bentul tuber is one of tuber plant species which have bioactive compound of Water Soluble Polysaccharide (WSP) and potentially healthy nutrition in the therapy of metabolic syndrome disease. The purpose of this research is to prove the ability of WSP isolate to reduce blood glucose level in white mice.  Research method include the yield of WSP isolates white bentul tuber using enzymatic method, WSP identification using HPLC with Aminex HPX-87C BIORAD5 columns, and antidiabetic activity test using white mices. Test activity was performed in six treatment groups (Normal, Induction STZ 20 mg/ kgBW, Induction STZ 20 mg / kgBW + metformin 195mg / KgBW, three treatment with STZ induction 20 mg/ kgBW and WSP isolate with concentration 200, 400, and 600mg / kgBW). Determination of blood glucose levels using glucometer and supported by observation of histologic improvement of beta pancreatic cells in white mice that have necrosis. The result research are WSP yield of 4.81% and WSP level of 94.45%. Results of blood glucose levels of mice induced STZ 20mg/kgBW decreased optimal blood glucose with a dose of WSP 400mg/kgBW in the first week and histologic improvement of beta pancreatic cells that experienced the most optimal necrosis at WSP dose of 200 mg/kgBW. The conclusion of this research is the provision WSP isolate of white bentul tubers at doses of 200, 400, 600 mg/kgBW can decrease the blood glucose level induced STZ 20mg/kg BW and histological improvement in pancreatic beta cells at the most optimal dose of 200 mg / kgKeywords: White bentul tuber, water soluble polysaccharides, diabetes mellitus, beta pancratic cells

Background: The phytochemical screening and antidiabetic activities of the methanol leaf extract and fractions of Sansevieria liberica Ger. and Labr. were investigated.Objectives: To identify the phytoconstituents present and evaluate the anti-diabetic activity of the leaf of S. liberica Ger. and Labr.Material and Methods: A 1.2 g of the pulverized leaf was cold macerated in methanol to obtain 240 g of the crude extract (CE), fractionation was done using n-hexane, ethylacetate and butanol. The CE and the fractions were screened for phytochemical and antidiabetic analysis using standard procedures. Diabetes was induced by intraperitonial injection of alloxan. The blood glucose levels were analyzed as indices of diabetes.Results: After 24 hours, alloxan increased the blood glucose level of rats indicating hyperglycaemia. Treatment of the rats with the extract and fractions reduced the blood glucose level within 3 to 7 days of treatment. The standard antidiabetic, metformin (500 mg/kg) also showed similar effect. Extracts (250 and 500 mg/kg) of the crude extract decreased significantly (p<0.05) the blood glucose level. The antidiabetic potency of the extract and fractions were in the order of CE ˃ ethylacetate fraction > n-Hexane fraction > butanol fraction. The phytochemical screening of the extract and fractions revealed the presence of flavonoids, steroids, alkaloids, glycosides etc.Conclusions: From the findings, S. liberica leaf posses antidiabetic activity which may be due to the phytochemical constituents present, and this justifies its use by local users as traditional remedy for diabetes mellitus.

Objective: As per the ethnopharmacological information has Butea monosperma been used to treat diabetes mellitus by the tribal people of tropical and subtropical areas. However, there is no much more scientific report available about the antidiabetic property of the leaves of the plant. Hence, the study was undertaken to evaluate the antidiabetic effect of ethanolic extract of B. monosperma on blood levels of adrenaline-induced and glucose feed diabetic rabbits.Methods: The three different doses of the extracts (100, 200, and 400 mg/kg) were administered orally to an experimental animal. The animals were induced diabetics by adrenaline and high glucose diet. Blood glucose level was measured accordingly. For antidiabetic activity, photocolorimeter was used to monitoring the blood glucose level with crest kit box (GOP-POD method).Results: The extracts showed considerable dose-dependent activity. However, the dose 400 mg/kg showed considerable lower of blood glucose level. p<0.01 indicates the significance result. 8 hrs reading 182.5±3.83 for 400 mg does is most effective for reducing blood sugar.Conclusion: The study indicates that the ethanolic extract of B. monosperma leaves possesses antidiabetic properties which suggest the presence of biologically active components.

Abstract Background Blood glucose levels that are too high or too low after traumatic brain injury (TBI) negatively affect patient prognosis. This study aimed to demonstrate the relationship between blood glucose levels and the Glasgow Outcome Score (GOS) in TBI patients. Methods This study was based on a randomized, dual-center, open-label clinical trial. A total of 208 patients who participated in the randomized controlled trial were followed up for 5 years. Information on the disease, laboratory examination, insulin therapy, and surgery for patients with TBI was collected as candidate variables according to clinical importance. Additionally, data on 5-year and 6-month GOS were collected as primary and secondary outcomes, respectively. For multivariate analysis, a generalized additive model (GAM) was used to investigate relationships between blood glucose levels and GOS. The results are presented as odds ratios (ORs) with 95% confidence intervals (95% CIs). We further applied a two- piecewise linear regression model to examine the threshold effect of blood glucose level and GOS. Results A total of 182 patients were included in the final analysis. Multivariate GAM analysis revealed that a bell-shaped relationship existed between average blood glucose level and 5-year GOS score or 6-month GOS score. The inflection points of the average blood glucose level were 8.81 (95% CI: 7.43–9.48) mmol/L considering 5-year GOS as the outcome and were 8.88 (95% CI 7.43−9.74) mmol/L considering 6-month GOS score as the outcome. The same analysis revealed that there was also a bell relationship between average blood glucose levels and the favorable outcome group (GOS score ≥ 4) at 5 years or 6 months. Conclusion In a population of patients with traumatic brain injury, blood glucose levels were associated with the GOS. There was also a threshold effect between blood glucose levels and the GOS. A blood glucose level that is either too high or too low conveys a poor prognosis. Trial registration ClinicalTrials.gov NCT02161055. Registered on 11 June 2014.

Challenges in lifestyle, such as increasesin energy intake and decreasesin physical activity are causing overweight and obesity leading to epidemic increases in type II Diabetes Mellitus. The research approach used for this study was evaluative approach and the research design was true experimental design. 60 patients with type II diabetes, 30 in experimental group and 30 in control group were selected for this study by using purposive sampling technique. Data was collected with the help of self-structured interview schedule. Descriptive statistics (frequency, percentage, mean and standard deviation) and inferential statistics (chi-square, paired ‘t’ test) were used to analyse the data and to test the hypotheses. In the experimental group,the pre-test mean score was 2.966, mean percentage was 59% and standard deviation was 1.129 and in post-testmean score was 2.533, mean percentage was 50.66% and standard deviation was 1.074 with effectiveness of 8.34% and paired‘t’ test value of t=3.971,which was statistically significant (p<0.05) which is an evidence ofthe effectiveness of Amla juice in reducing blood glucose level. Comparison of blood glucose levels in experimental and control groups, shows that the value is statistically highly significant, as was observed from the unpaired ‘t’ test value of 13.39 with P value of <0.05, which is an evidence indicatingthe effect of Amla juice in reducing postprandial blood glucose levels. The resultsfound that the administration of Amla juice did have aneffect in reducing blood glucose level in the experimental group. By comparing the findings of pre-test and post test between the experimental group and the control group,the effect was identified (assessed). The study concluded that the Amlajuice is effective in reducing blood glucose level.

The calibration method is an alternative method that can be used to analyze the relationship between invasive and non-invasive blood glucose levels. Calibration modeling generally has a large dimension and contains multicolinearities because usually in functional data the number of independent variables (p) is greater than the number of observations (p>n). Both problems can be overcome using Functional Regression (FR) and Functional Principal Component Regression (FPCR). FPCR is based on Principal Component Analysis (PCA). In FPCR, the data is transformed using a polynomial basis before data reduction. This research tried to model the equations of spectral calibration of voltage value excreted by non-invasive blood glucose level monitoring devices to predict blood glucose using FR and FPCR. This study aimed to determine the best calibration model for measuring non-invasive blood glucose levels with the FR and FPCR. The results of this research showed that the FR model had a bigger coefficient determination (R2) value and lower Root Mean Square Error (RMSE) and Root Mean Square Error Prediction (RMSEP) value than the FPCR model, which was 12.9%, 5.417, and 5.727 respectively. Overall, the calibration modeling with the FR model is the best model for estimate blood glucose level compared to the FPCR model.

The aim: To compared blood glucose concentrations after intravenous injection of dexamethasone in the pregnant and non-pregnant women under general anesthesia. Materials and methods: Eighty patients aged 18-50 years took part in the study (ASA class 1 and 2). Forty of patients were undergoing elective cesarean section under G/A and the other forty undergoing elective laparoscopic Cholecystectomy under G/A. Anesthesia was induced using IV anesthetic drugs (0.5mg/kg ketamine, sleeping dose of propofol up to 2mg/kg, muscle relaxant was 0.6 mg/kg rocuronium and maintained with isoflurane). All of patients have been injected with 0,1mg/kg dexamethasone intravenously, at induction of anesthesia, Blood glucose concentrations were measured at induction and then in 60min, 180min and in 360 min after injection of dexamethasone and results were compared between the groups; IV fluid added was normal saline (0.9%) during the study. Results: Regarding to blood glucose levels, we noticed that its level significantly increased over time and peaked in 180min after dexamethasone injection in both groups. The difference percentage between the lower reading (pre injection) and the upper reading (in 180min after) was 33.5% in pregnant woman and 46.2%for non-pregnant women, this difference was statistically significant relative to the pre injection, as this difference was lower in the pregnant women. In 360min after blood glucose level began to drop in both groups. After giving 0.1 mg/kg of dexamethasone, blood glucose level increased in both groups, but it was lower in pregnant women.

Diabetes is a chronic disease which characterized by hyperglycemia (elevated or abnormally high blood sugar levels) and other metabolic disturbances, including metabolism of lipids and haemostasis. Caesalpinia pulcherrima has previously showed strong anti-diabetic and hepatoprotective potential. The present research work was to investigate the anti-diabetic activity and hepatoprotective activity Caesalpinia pulcherrima in streptozotocin-induced (STZ) diabetic rats. The dose-dependent effects of 45days oral treatment with methanol extract of plant (200 and 300mg/kg) of CPAE on body weight, blood glucose level, total protein, albumin, liver marker enzymes and carbohydrate metabolizing enzymes were evaluated in STZ-induced diabetic rats. Oral administration methanolic extract of Caesalpinia pulcherrima of showed significant restoration of the body weight and decrease in the blood glucose level, liver marker enzymes (ALT, AST ALP) and carbohydrate metabolizing enzymes were observed in diabetic rats. These results suggest that fruit extract of Caesalpinia pulcherrima has valuable anti-diabetic activity in STZ-induced diabetic rats which is comparable to the standard drug metformin and hence might be of use in the management of diabetes.
