Kepariwisataan merupakan suatu kegiatan bersifat multidimensional yang dapat menunjukan identitas tempat wisata, salah satu nya objek Wisata Colo Muria yang memiliki daya tarik dan kearifan lokal yang dapat dikunjungi oleh para wisatawan. Penelitian ini bertujuan untuk membuat perencanaan sistem digitalisasi pada objek wisata Colo Muria Kabupaten Kudus dengan menggunakan teknologi Augmented Reality. Metode penelitian yang digunakan adalah egile development. Langkah-langkah penelitian ini adalah Planning, Analysis, Design, Implementation, dan testing. Pada pengujian aplikasi, dilakukan dengan pengujian alpha berupa functional testing (uji black box), portablity testing dan pengujian beta berupa usablity testing (uji ahli media dan uji pengamat pengguna aplikasi). Pengujian alpha yang menguji aspek functionality menggunakan uji black box menunjukkan bahwa aplikasi berjalan dengan baik dan sesuai dengan yang direncanakan. Pengujian beta dilakukan oleh ahli media diperoleh hasil persentase 80%. Dari hasil ini dapat diketahui bahwa sistem digitalisasi pada objek wisata colo muria memiliki kemampuan yang baik, sehingga dapat dinyatakan layak digunakan sebagai sarana promosi wisata. Untuk pengujian beta dengan menggunakan uji responden, mendapatkan skor rata-rata akhir sebesar 79%, sehingga masuk dalam kriteria acceptable atau layak untuk digunakan. Saran yang diajukan untuk penelitian selanjutnya yaitu menambahkan video animasi perjalanan wisata dan animasi objek 3D yang lebih baik lagi agar sistem digitalisasi objek wisata colo muria lebih lengkap.

ABSTRACTBackgroundPathogenic variants in the LRRK2 gene are a common monogenic cause of Parkinson’s disease. However, only seven variants have been confirmed to be pathogenic.ObjectivesWe identified two novel LRRK2 variants (H230R and A1440P) and performed functional testing.MethodsWe transiently expressed wildtype, the two new variants, or two known pathogenic mutants (G2019S and R1441G), in HEK-293T cells, with or without LRRK2 kinase inhibitor treatment. We characterized the phosphorylation and kinase activity of the mutants by western blotting. Thermal shift assays were performed to determine the folding and stability of the LRRK2 proteins.ResultsThe two variants were found in two large families and segregate with the disease. They display altered LRRK2 phosphorylation and kinase activity.ConclusionsWe identified two novel LRRK2 variants which segregate with the disease. The results of functional testing lead us to propose these two variants as novel causative mutations for familial Parkinson’s disease.

Kegiatan akreditasi bertujuan untuk mengevaluasi mutu dan kelayakan suatu institusi berdasarkan data yang dimilikinya. Proses akreditasi memerlukan data pendukung yang bersumber dari berbagai dokumen dan database yang berbeda-beda, Sehingga diperlukan sistem untuk proses pengumpulan dan visualisasi data yang cepat dan efisien. Salah satu sistem yang dapat digunakan adalah penggunaan sistem dashboard dan data warehouse. Dalam penelitian ini, dilakukan pengembangan dan implementasi data warehouse dan sistem dashboard yang berdasar pada pedoman Instrumen Akreditasi Program Studi 4.0 (IAPS 4.0) dari BAN-PT Dari hasil implementasi, didapatkan 38 indikator bersifat kuantitatif yang dapat divisualisasikan ke dalam sistem dashboard. Dari proses pengujian menggunakan requirement traceability matrix dan functional testing, didapatkan hasil semua item pengujian berhasil lolos uji dan sistem ini dapat digunakan untuk proses akreditasi berdasar pada IAPS 4.0 BAN-PT. Dengan adanya sistem ini diharapkan dapat digunakan program studi untuk dilakukan proses evaluasi mandiri pada sebelum atau saat dilakukan proses akreditasi program studi.

Abstract Introduction The CHaDOx1 nCov-19 AstraZeneca (AZ) vaccination has been associated with an antibody-mediated prothrombotic syndrome, termed "Thrombosis with Thrombocytopenia Syndrome" (TTS)[1-3]. The current diagnostic criteria for TTS are thrombosis (venous or arterial) within 4-42 days of AZ vaccine, thrombocytopenia and presence of an antibody to platelet factor 4 (PF4)[4, 5]. TTS commonly presents with cerebral venous sinus thrombosis (CVST) or splanchnic vessel thrombosis (SVT), but outside of TTS, CVST and SVT are uncommon, with an overall incidence of less than 0.5 per 100,000 [5-7]. Deep vein thrombosis (DVT) and pulmonary embolism (PE) are also associated with TTS, however the background incidence of venous thromboembolism (VTE) is much higher, with 1-2 events per 1000 patients per year[7, 8]. Therefore, many patients will present with new VTE and a recent exposure to the AZ vaccine, requiring consideration of investigation for TTS. Recent data suggests that PF4 antibodies can be seen in up to 8% of patients without thrombosis but following AZ vaccination[9]. We hypothesised in patients with recent AZ vaccination, new VTE but with a normal platelet count, that the incidence of a PF4 antibody is similar to this background rate of PF4 positivity. If confirmed, then presence of a normal platelet count despite new VTE and recent vaccination may exclude TTS without the need for PF4 antibody testing. We present our preliminary data on the rates of PF4 antibody positivity amongst patients with VTE, recent AZ vaccination and a normal platelet count at presentation. Aim and Methods To assess the incidence of PF4 ELISA positive results in patients with confirmed VTE, recent vaccination (within 4-42 days) with the first dose of AZ vaccine, and platelet count greater than 150x10 9/L. A retrospective audit of cases referred with suspected TTS to Monash Pathology, Melbourne, Victoria, and New South Wales Health Pathology at Royal Prince Alfred Hospital and St George Hospital sites Sydney, New South Wales, Australia, for testing for anti PF4 antibodies from 1 st April to 31 st July 2021. Patient sera were tested for the Anti-PF4 antibody using the STAGO Asserachrom HPIA IgG ELISA (Asnières sur Seine, France). For patients with a positive PF4 antibody test additional testing was sought for either the presence of platelet activating antibodies with a flow cytometry-based assay or the presence of spontaneous serotonin release without heparin in the serotonin release assay. Results From April 1 st to July 31 st 350 tests were run on 332 patients. 91 patients met our criteria, of whom 51 were female and 40 male, with a median age of 73 years. Median platelet count at presentation was 226x10 9/L, and median D dimer values were 10 times the upper limit of normal. 86 patients had either DVT, PE or both, including 2 with upper limb DVT, and 5 patients had PE with concurrent arterial events (1 axillary artery thrombosis, 3 arterial strokes, 1 coronary artery thrombosis). Further details are presented in table 1. 82 patient samples tested negative for anti-PF4 antibodies by ELISA, 5 were positive, and were 4 weak positive/equivocal (see table 2 for further details). Of the positive results, 3 had functional testing available, of which 2 were negative, and 1 showed discordant results, with a positive SRA but negative flow cytometry. None of the weak positive/equivocal cases had functional testing results available. Of the negative ELISA results, 5 patients had functional testing results available, of which 4 were negative. One of these cases had positive testing by flow cytometry, but negative by SRA (case included in table 2). Conclusion In our Australian cohort of patients with their first dose of AZ vaccine and new VTE within 4-42days, but a normal platelet count (therefore not fulfilling the clinical criteria of TTS), the incidence of a positive PF4 antibody test was 9/91 (9.9%, 95% CI 3.7-15.9%) and only one had evidence of platelet activating antibodies. This observed rate is similar to that observed in healthy patients without thrombosis who received AZ vaccination as described by Thiele et. al., 2021. Further confirmation in a larger cohort of VTE patients is required, but if confirmed, then PF4 ELISA testing in patients with VTE and normal platelet count post AZ vaccine may not be required, and should give clinicians confidence to institute routine management. Abstract Palm oil fields with slopes require special wheels for transportation for transporting fresh fruit bunches (FFB), the special wheels in question are modified wheels from the existing cage wheel, the modified wheels can adjust the slope conditions on the land with the lug angle adjustment mechanism. The objective of this study was to test the functional and performance of the modified lug wheel as a means of transporting FFB on sloping land using a hand tractor on a pivot type trailer. Functional testing on the modified lug wheel was carried out by observing changes in the angle of the lugs (0, 15, 30, 45)0 by lifting one of the tractor shafts first using a jack, after which the locking pins were released. The angle change rim can be moved clockwise or anticlockwise by hand according to the desired lug angle. Performance testing on the modified lug wheel is carried out by observing and calculating the slip on the wheel. Functional testing on trailer pivots is carried out by observing all components of the towing system when installed and operated, to determine whether the trailer towing system can function properly. Performance testing on the pivot trailer is done by observing and calculating the turning radius when the tractor turns. Performance testing is also carried out by observing the stability of the two-wheel tractor and trailer. The slope of the land and the angle of the lugs greatly affect wheel slip, the higher the slope on a land, the higher the possibility of wheel slippage during operation. The test results show that the use of modified lug wheels shows better performance at 30° lug angle, as seen from the smaller slip value at 30° lug angle.

Background: Differences in postoperative recovery after anterior cruciate ligament reconstruction (ACLR) between men and women have been demonstrated in the adult population. Sex-based differences have been incompletely investigated in adolescents, which represent the subpopulation most affected by ACL injury. Purpose/Hypothesis: The purpose of this study was to compare the 6-month postoperative functional recovery after ACLR between adolescent boys and girls. It was hypothesized that significant differences in postoperative strength, dynamic balance, and functional hop test performance would be seen between the sexes. Study Design: Cohort study; Level of evidence, 3. Methods: Included in this study were athletes aged 12 to 19 years with closed or closing growth plates who underwent ACLR with hamstring autograft between May 2014 and May 2018 at a single institution. All athletes had undergone strength and functional testing between 5 and 8 months postoperatively. Exclusion criteria were previous knee surgery (contralateral or ipsilateral knee), concomitant injury/surgery other than meniscal tear/repair, allograft supplementation, and incomplete medical records. The limb symmetry index (LSI) for strength (measured with handheld dynamometer), as well as dynamic Y-balance and functional hop test performance, was compared between groups. To account for differences in physical characteristics between the sexes, 1-way between-group multivariate analysis of covariance was used to analyze the data. Results: Overall, 543 patients (211 boys, 332 girls) were included. There was no significant difference in age, body mass index, incidence of concomitant meniscal pathology, use of regional anesthesia, or time to functional testing between cohorts. Female athletes demonstrated a statistically significantly greater deficit in quadriceps strength LSI compared with male athletes (boys, +3.4%; girls, –2.3%; P = .011). Both male and female athletes demonstrated 33% hamstring strength deficits, with no statistically significant sex-based differences in dynamic balance or functional hop testing. Conclusion: Female athletes demonstrated greater quadriceps strength deficits than male athletes at 6 months after ACLR with hamstring autograft. Severe hamstring strength deficits persisted in both male and female patients at this time point. The correlation of such deficits to risk of ACL retear warrants continued study in the adolescent population and may support a delay in return to sports, which has been suggested in the more recent literature.
