<p>Propolis, bee venom and bee pollen all have been used by humans traditionally for various medicinal purposes. Studies of these products have been limited primarily to antimicrobial, antifungal, anticancer and free radical scavenging properties. The mechanisms of action of these products remain largely unknown. This study investigates the biological effects of propolis, bee venom and bee pollen using chemical genomics and the yeast model organism. These products are screened against genome-wide yeast mutant libraries to determine the genes, proteins, and pathways that are targets of these products. I identified that propolis chelates iron and consequently creates an iron-deficient condition, which results in the upregulation of plasma membrane and vacuolar high-affinity iron transporters to maximise iron acquisition. Bee venom inhibited the biosynthesis of phosphatidylcholine via Opi3p that catalyses the final two steps of phosphatidylcholine biosynthesis within the CDP-ethanolamine pathway. Bee pollen showed a potential effect on GDP-mannose transport in which the GDP-mannose transport mutants confer hypersensitivity against bee pollen treatment.</p>

Ilex paraguariensis A. St. Hil. (Aquifoliaceae), popularly known as “yerba mate,” has great economic and social significance for the population of Southern Latin America. This study was conducted (1) to investigate the phytochemical composition of four different standardized extracts, (2) to investigate its free radical scavenging properties, and (3) to investigate the anti-inflammatory action of I. paraguariensis and its major chemical markers. The chemical profile was achieved by Folin-Ciocalteu, by LC/DAD, and by LC/MS assays, while the antioxidant and anti-inflammatory properties were investigated, respectively, by DPPH assay and by inhibition of nitric oxide (Griess reaction) and TNF-α (ELISA). Our results demonstrated that the IA (aqueous infusion extract) showed higher amounts of total phenolic contents ( 266.62 ± 10.85   mg CAE·g-1 DE), the highest amounts of all six chemical markers (theobromine, 5-O-caffeoylquinic acid, 4-O-caffeoylquinic acid, 3-O-caffeoylquinic acid, caffeine, and rutin), and stronger antioxidant activity ( E C 50 = 54.4 ± 5.14   μ g · m L − 1 ). The IA extract also showed the lowest inhibition of NOx secretion ( 50.10 ± 8.97 % ) as well as inhibition of TNF-α ( 83.33 ± 4.01 % ). Regarding the chemical markers, all compounds showed strong inhibition of NOx secretion, especially theobromine, which was 200x more potent than dexamethasone. Furthermore, TNF-α secretion was also significantly decreased by THEO at 0.033 μM ( 22.15 ± 6.49 % ), NCA at 1.97 μM ( 27.46 ± 3.98 % ), CCA at 0.35 μM ( 39.76 ± 5.73 % ), CGA at 0.56 μM ( 23.58 ± 5.79 % ), CAF at 0.52 μM ( 26.45 ± 5.34 % ), and RUT at 0.16 μM ( 40.18 ± 3.70 % ). Our results suggest that I. paraguariensis and its major chemical markers have strong free radical scavenging properties as well as showed important anti-inflammatory activity and that these compounds in a plant extract may work based on several different mechanisms synergistically, resulting in moderating the immune system.

It is well known that both Fagopyrum esculentum species, buckwheat achenes and buckwheat germs, contain flavonoids, and that they can be considered functional foods. In the present study we have analyzed the total content of polyphenols (TPC) and flavonoids (TFC), as well as the antioxidant activity of buckwheat ahcenes originating from the Balti region, Republic of Moldova, and also of the buckwheat sprouts over seven days of germination. The content of phytochemicals in achenes and germinated buckwheat after three and seven days was determined by HPLC–MS analysis. Using the Folin–Ciocalteu method, we recorded an increase in TPC of 360%, and of 436% in TFC after seven days of buckwheat ahcene germination. We aimed to investigate the free radical scavenging properties of methanolic extracts from ahcenes and sprouted buckwheat. We identified and quantified flavonoids and lignans such as rutin, orientin, isoorientin, vitexin, quercetin, quercitrin, isorhamnetin, lutein, apigenin, catechins, coumestrol—which have countless beneficial effects on human health—using HPLC–MS. FTIR analysis also revealed the accumulation of phenolic compounds during germination. This is the first study on the identification and quantification of phytochemicals from buckwheat achenes and sprouts from the Balti region of the Republic of Moldova.

Black pepper ( Piper nigrum L.) is a popular spice that is grown as tropical and subtropical plant throughout the world. The leaf, flower, fruit, and root are the most important elements of the plant. Asexual or vegetative propagation is becoming highly popular, although the sexual approach is still used for pepper vine cultivation. For mass production of the pepper plant, in vitro culture is also used. The bioactive components contained in them are extremely important because of their therapeutic potential against a number of diseases. They are usually classed as functional foods because, in addition to providing basic nutrition, provide physiological benefits and help to avoid chronic illness. The main component of black pepper is piperine. It has a complex phyto-chemistry includes: Volatile oil, alkaloids, and oleoresins. Because of its free-radical scavenging properties, black pepper and its active components can be prevention and control of tumor growth. Piperine, which can bind and inhibit the SARS-CoV-2 virus that causes the sickness, is present in black pepper and has antibacterial and antiviral effects. Piperine, a key alkaloid component of black pepper, it also helps to cognitive brain function, nutritional absorption, and gastrointestinal health. Black pepper is known as the “King of Spices” as well as the “King of Medicinal Agents,” since it includes a wide variety of bioactive compounds with nutraceutical and pharmacological applications. An overview of the most common applications for black pepper, along with a strong evidence is present in this review.

: Polyphenolic phytoconstituents have been widely in use worldwide since ages and are categorised as secondary metabolites of plants. The application of polyphenols such as quercetin, resveratrol. curcumin as nutritional supplement has been researched widely. The use of polyphenols, and specifically quercetin for improving the memory and mental endurance have shown significant effects among rats. Even though similar results has not been resonated among human but encouraging preclinical results have encouraged researchers to explore other polyphenols to study the effects as supplements among athletes. The phytopharmacological research has elucidated the use of natural polyphenols to prevent and treat various physiological and metabolic disorders owing to its free radical scavenging properties, anti-inflammatory, anti-cancer and immunomodulatory effects. In spite of the tremendous pharmacological profile, one of the most dominant problem regarding the use of polyphenolic compounds is their low bioavailability. Nanonization is considered as one of the most prominent approaches among many. This article aims to review and discuss the molecular mechanisms of recently developed nanocarrier-based drug delivery systems for polyphenols and its application as drugs and supplements. Nanoformulations of natural polyphenols are bioactive agents, such as quercetin, kaempferol, fisetin, rutin, hesperetin, and naringenin epigalloccatechin-3-gallate, genistein, ellagic acid, gallic acid, chlorogenic acid, ferulic acid, curcuminoids and stilbenes is expected to have better efficacy. These delivery systems are expected to provide higher penetrability of polyphenols at cellular levels and exhibit a controlled release of the drugs. It is widely accepted that natural polyphenols do demonstrate significant therapeutic effect. However, the hindrances in their absorption, specificity and bioavailability can be overcome using nanotechnology.

The present study was carried out to elucidate the in vitro free radical scavenging potential of various extracts of Tabebuia roseo-alba leaves. Assays for radical scavenging, such as DPPH, ABTS+, hydroxyl, hydrogen peroxide, superoxide, nitric oxide assay and reducing power activity were performed using standard protocols and the results were compared with standard ascorbic acid. Among the various extracts used the ethanolic extract of T. roseo-alba exhibited efficient scavenging potential with lowest EC50 value proving its antioxidant potential. Leaves of T. roseo-alba have strong free radical scavenging properties and thus can be used as a potential antioxidant to resolve diseases that are associated with oxidative stress including diabetes and other age related disorders.

Kombucha is a fermented beverage. Its consumption has significantly increased during the last decades due to its perceived beneficial effects. For this reason, it has become a highly commercialized drink that is produced industrially. However, kombucha is still also a homemade beverage, and the parameters which, besides its organoleptic characteristics, define the duration of its potential beneficial properties over time, are poorly known. Therefore, this study aimed to determine the effect of 9-month storage at 4 °C with 30-day sampling on the pH, total phenolic, and flavonoid contents, free radical scavenging properties of kombucha fermented from black tea. Our results highlighted that, after four months, the phenolic content decreased significantly from the initial value of 234.1 ± 1.4 µg GAE mL−1 to 202.9 ± 2.1 µg GAE mL−1, as well its antioxidant capacity tested by two in vitro models, DPPH, and ABTS assays. Concomitantly, the pH value increased from 2.82 to 3.16. The novel findings of this pilot study revealed that kombucha from sugared black tea can be stored at refrigerator temperature for four months. After this period the antioxidant properties of kombucha are no longer retained.

Croton ferrugineus Kunth is an endemic species of Ecuador used in traditional medicine both for wound healing and as an antiseptic. In this study, fresh Croton ferrugineus leaves were collected and subjected to hydrodistillation for extraction of the essential oil. The chemical composition of the essential oil was determined by gas chromatography equipped with a flame ionization detector and gas chromatography coupled to a mass spectrometer using a non-polar and a polar chromatographic column. The antibacterial activity was assayed against three Gram-positive bacteria, one Gram-negative bacterium and one dermatophyte fungus. The radical scavenging properties of the essential oil was evaluated by means of DPPH and ABTS assays. The chemical analysis allowed us to identify thirty-five compounds representing more than 99.95% of the total composition. Aliphatic sesquiterpene hydrocarbon trans-caryophyllene was the main constituent with 20.47 ± 1.25%. Other main compounds were myrcene (11.47 ± 1.56%), β-phellandrene (10.55 ± 0.02%), germacrene D (7.60 ± 0.60%), and α-humulene (5.49 ± 0.38%). The essential oil from Croton ferrugineus presented moderate activity against Candida albicans (ATCC 10231) with an MIC of 1000 μg/mL, a scavenging capacity SC50 of 901 ± 20 µg/mL with the ABTS method, and very strong antiglucosidase activity with an IC50 of 146 ± 20 µg/mL.
