New morphological criteria and molecular characterization of black aspergilli aggregate from corn, sorghum and wheat grains

Abstract Echinococcus granulosus is the causative agent of cystic echinococcosis, which has serious impacts on human and/or animal health, resulting in significant economic losses. Echinococcus granulosus comprises a number of intra-specific variants or strains at the genetic level. In Saudi Arabia, few studies were performed on genetic variations in Echinococcus species. Therefore, the present study aimed to investigate the phenotypic and genetic characterization of hydatid cysts harboured by sheep and camels in Al-Madinah Al-Munawarah. Samples of hydatid cysts were collected from local sheep (n = 25) and camels (n = 8). The morphological criteria of protoscoleces were investigated. To investigate the molecular characterization, random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR), single-stranded conformation polymorphism (SSCP) were carried out. DNA was extracted from individual fertile cysts and subjected to RAPD-PCR analysis (using five arbitrary primers) and PCR amplification of cytochrome c oxidase I (cox1) and 12S ribosomal ribonucleic acid (12S rRNA) genes. The PCR products were subjected to SSCP analysis for genetic discrimination in E. granulosus isolates. In addition, partially sequencing of the mitochondrial DNA cox1 genes was achieved for assessing the phylogenetic positions of collected isolates using some global published sequence data of cox1 genes. The rostellar hooks of camel and local sheep isolates show remarkable variability in their dimensions. Five distinct SSCP patterns were identified in the 12S rRNA gene, showing intraspecific variations in E. granulosus of camels and local sheep. Sequencing of (cox1) genes of both local sheep and camels exhibit high similarity with those of the same gene (E. granulosus sensu stricto) published in NCBI BLAST.

AbstractTwo tapeworm specimens collected in northeast China in 2009 and 2011 were identified as Diphyllobothrium latum based on morphological criteria. Molecular methods were used to confirm their identity and analyze genetic variations compared with published data for this species. Species identity was confirmed by molecular characterization of the 18S rDNA partial sequence, complete sequences of internal transcribed spacers (ITSs) and 5.8S rDNA, and partial sequences of mitochondrial cytochrome c oxidase subunit 1 (cox1) and mitochondrial NADH dehydrogenase subunit 5 (nad5). PCR amplification and sequence analysis of 18S rDNA (1472 bp), ITS regions (1218 bp), cox1 (885 bp), and nad5 (1028 bp) revealed that these four sequences showed more than 99% identity to reference sequences for D. latum, confirming that this species is D. latum. To date, a total of 12 diphyllobothriosis cases have been documented in China. This study represents the first molecular characterization of D. latum in China, providing molecular evidence of human diphyllobothriosis in China.

This paper is a contribution to the morphological and molecular characterization of the cyanobacterium Brasilonema roberti-lamii from populations found in Central Mexico. The general growth form and the morphological, morphometric and ecological characteristics of the populations studied clearly correspond to those described for Brasilonema roberti-lamii (basionym: Tolypothrix roberti-lamii) from the French Antilles. Based on molecular data from DNA sequencing of the16S rRNA gene and the IGS of the cpcB-cpcA phycocyanin operon (cpcBA-IGS), we propose that the populations that we studied are closely related to those of other Brasilonema species, including B. octagenarum UFV-OR1, UFV-E1 and HA4187-MV1-p1F, Brasilonema sennae CENA 114, B. tolantongensis, B. terrestre CENA 116, B. angustatum HA4187-MV1-B2+p1F and HA4187-MV1-B2+p1H and B. bromeliae SPC951. Our findings support the transference of Tolypothrix roberti-lamii, which was made based exclusively on morphological criteria, to Brasilonema. The use of molecular analyses in addition to traditional morphological and ecological criteria, known as polyphasic approach, is a good alternative to describe taxa of cyanobacteria, mainly at the genus and species levels.
