Cell and tissue interactions are a basic aspect of eukaryotic growth and development. While cell-to-cell interactions involving recognition and incompatibility have been studied extensively in animals, there is no known antigen-antibody reaction in plants and the recognition mechanisms operating in plant grafts have been virtually neglected. An ultrastructural study of the Sedum telephoides/Solanum pennellii graft was undertaken to define possible mechanisms of plant graft incompatibility. Grafts were surgically dissected from greenhouse grown plants at various times over 1-4 weeks and prepared for EM employing variations in the standard fixation and embedding procedure. Stock and scion adhere within 6 days after grafting. Following progressive cell senescence in both Sedum and Solanum, the graft interface appears as a band of 8-11 crushed cells after 2 weeks (Fig. 1, I). Trapped between the buckled cell walls are densely staining cytoplasmic remnants and residual starch grains, an initial product of wound reactions in plants.

The development of thematic textbook learning media based on local wisdom uses the Research and Development (R & D) method. The development stages include: (1) Potentials and problems, (2) Data collection, (3) Product design, (4) Design validation, (5) Design revision, (6) Product testing, (7) Product revision (Sugiyono, 2016). The research was conducted with 33 students from class V SDN 2 Sendang Jepara Regency. Initial product development is a process of making media based on needs analysis. Expert testing or validation was carried out by 2 experts, namely media experts and material experts. The data collection method in this research is observation, interview, and questionnaire. The data analysis technique used descriptive analysis and comparison test of two independent groups, namely using the independent t-test. Based on the results of the trial, this thematic teaching material is very valid, very interesting, effective and can be applied in learning. This can be seen from the results of the validation test from three experts which show that the product is good. The results of the effectiveness test showed that the experimental class students had better learning outcomes than the control class. This means that textbooks based on local wisdom of Jepara Regency are developed effectively

The distribution of sodium-potassium adenosine triposphatase (Na-K-ATPase) activity in kidney sections has been studied by a method based on the hydrolysis of p-nitrophenyl phosphate in alkaline medium containing dimethyl sulfoxide. The products at each stage in the reaction sequence have been subjected to electron probe microanalysis. The initial product was identified as a mixture of KMgPO4 and Mg(PO4)2, and sequential analysis demonstrated the linearity of conversion of this product to a visible form. In human, rabbit and rat kidneys the distribution of activity was found to be essentially identical, with highest levels located in thick ascending limbs and distal convoluted tubules. The initial reaction was completely potassium dependent and was inhibited by ouabain in concentrations reflecting the relative sensitivity of microsomal Na-K-ATPase in each species. Measurement of initial product phosphorus by means of the electron probe is presented as a practical technique for direct quantitation of Na-K-ATPase activity in identified tubule segments.

Palladium(II) ammine complexes enriched in 15N give sharp singlets in their 15N n.m.r . spectra. The 15N chemical shifts depend primarily on the trans donor atom, in a qualitatively similar way to those for platinum(II) ammine complexes. A dimethylformamide solution of Pd(NH3)2Cl2 prepared in the usual way gave a single peak, consistent with the presence of only one isomer (trans). Reactions of increasing amounts of aqueous perchloric acid with Pd(NH3)42+ allowed peaks to be identified for Pd(NH3)3(H2O)2+, cis -Pd(NH3)2(H2O)22+ and Pd(NH3)(H2O)32+. The initial product of the reaction of trans-Pd(NH3)2Cl2 with silver ion is trans-Pd(NH3)2(H2O)22+, which rapidly disproportionates to Pd(NH3)3(H2O)2+ and Pd(NH3)(H2O)32+. With standing, subsequent reactions ultimately produce a solution containing predominantly cis -Pd(NH3)2(H2O)22+ in equilibrium with small amounts of the tri- and mono-ammine complexes, and free ammonia.

Ablation of androgen production through surgery is one strategy against prostate cancer, with the current focus placed on pharmaceutical intervention to restrict androgen synthesis selectively, an endeavor that could benefit from the enhanced understanding of enzymatic mechanisms that derives from characterization of key reaction intermediates. The multifunctional cytochrome P450 17A1 (CYP17A1) first catalyzes the typical hydroxylation of its primary substrate, pregnenolone (PREG) and then also orchestrates a remarkable C17–C20 bond cleavage (lyase) reaction, converting the 17-hydroxypregnenolone initial product to dehydroepiandrosterone, a process representing the first committed step in the biosynthesis of androgens. Now, we report the capture and structural characterization of intermediates produced during this lyase step: an initial peroxo-anion intermediate, poised for nucleophilic attack on the C20 position by a substrate-associated H-bond, and the crucial ferric peroxo-hemiacetal intermediate that precedes carbon–carbon (C-C) bond cleavage. These studies provide a rare glimpse at the actual structural determinants of a chemical transformation that carries profound physiological consequences.

New modes of sterilization of compote from pears in glass containers of SKO 1-82-1000 using high-temperature high-speed heat carriers have been developed. A mathematical model for calculating the duration of the sterilization process depending on the parameters of the heat carrier is obtained. To obtain a mathematical model, we used the method of mathematical planning of the experiment. A technological method for accelerated sterilization of pear compote in a glass jar with a volume of 1 dm3 is proposed. The research made it possible to develop new modes of heat treatment (sterilization) of compote from pears in a glass jar 1-82-1000 at different parameters of hot air and different initial temperatures of the product, °C: 70, 80, 85 and 90. The shortest duration of the thermal sterilization process is achieved at the heat transfer fluid speed of 7,5 m/s at the initial product temperature of 90°C.
