Rubi Fructus (RF) is known to exert several pharmacological effects including antitumor, antioxidant, and anti-inflammatory activities. However, its antiobesity effect has not been reported yet. This study was focused on the antidifferentiation effect of RF extract on 3T3-L1 preadipocytes. When 3T3-L1 preadipocytes were differentiating into adipocytes, 10–100 μg/mL of RF was added. Next, the lipid contents were quantified by Oil Red O staining. RF significantly reduced lipid accumulation and downregulated the expression of peroxisome proliferator-activated receptorγ(PPARγ), CCAAT0-enhancer-binding proteinsα(C/EBPα), adipocyte fatty acid-binding protein 2 (aP2), resistin, and adiponectin in ways that were concentration dependent. Moreover, RF markedly upregulated liver kinase B1 and AMP-activated protein kinase (AMPK). Interestingly, pretreatment with AMPKαsiRNA and RF downregulated the expression of PPARγand C/EBPαprotein as well as the adipocyte differentiation. Our study shows that RF is capable of inhibiting the differentiation of 3T3-L1 adipocytes through the modulation of PPARγ, C/EBPα, and AMPK, suggesting that it has a potential for therapeutic application in the treatment or prevention of obesity.

Efek Troglitazone terhadap Perubahan Morfologi dan Ekspresi Gen PPAR- γ di Dalam Sel Adiposa 3T3-L1 ABSTRACT3T3-L1 cells are extensively used as a model to study adipogenesis. However, one major concern is the prolonged period of time it takes the cells to differentiate into adipocytes form. To induce this differentiation, the adipogenic induction media is required. In this study, troglitazone, a hypoglycemic agent was added to adipogenic induction media and observed in order to determine the morphological changes and peroxisome proliferator-activated receptor gamma (PPAR-γ) gene expression in 3T3-L1 differentiation. It is generally known that PPAR-ꝩ plays an important role as a transcription factor in adipocyte differentiation. Based on Oil Red O Staining, adipogenic induction with or without troglitazone changed the 3T3-L1 pre-adipocytes into mature round fat cells characterized by red droplet lipids. This cell also had a high absorbance level and degree of droplet accumulation of P≤ 0.05 in each group. In addition, cells treated by troglitazone had the highest PPAR-ꝩ mRNA level (1.9 fold) than those treated by adipogenic induction media without troglitazone or cells un-treated at all. Keywords: 3T3-L1, adipocyte, differentiation, PPAR-ꝩ, troglitazone ABSTRAKSel 3T3-L1 adalah jenis sel yang banyak digunakan dalam studi adipogenesis. Namun, salah satu kelemahan sel tersebut adalah lamanya waktu yang dibutuhkan bagi sel pre-adiposa untuk berdiferensiasi menjadi sel adiposa. Selain itu, dibutuhkan pula media induksi khusus untuk mengubah sel menjadi sel adiposa. Pada penelitian ini, kami mengobservasi fungsi troglitazone, sebagai antidiabetes terhadap perubahan morfologi dan ekspresi gen peroxisome proliferator-activated receptor gamma (PPAR-γ). Telah diketahui bahwa PPAR-ꝩ berperan penting sebagai factor transkripsi dalam diferensasi sel adiposa. Berdasarkan pewarnaan ORO, induksi sel pre-adiposa 3T3-L1 dengan media induksi dengan dan tanpa troglitazone merubah sel preadiposa menjadi sel berbentuk bulat yang dikarakterisasi dengan akumulasi droplet lemak. Nilai absorbansi sel adiposa juga menandakan adanya perbedaan yang signifikan antara kelompok sel yang diberi troglitazone dan tidak, dan sel tanpa diberi media induksi. Sementara, pada kelompok sel yang diberi troglitazone memiliki ekspresi mRNA PPAR-ꝩ (1,9 kali) tertinggi jika dibandingkan dengan sel yang diberi media induksi tanpa troglitazone, dan tanpa media induksi sama sekali.Kata Kunci: 3T3-L1, adiposa, diferensiasi, PPAR-ꝩ, troglitazone

This study was performed in order to investigate the antiobese effects of the ethanolic extract of Veratri Nigri Rhizoma et Radix (VN), a herb with limited usage, due to its toxicology. An HPLC analysis identified jervine as a constituent of VN. By an Oil Red O assay and a Real-Time RT-PCR assay, VN showed higher antiadipogenic effects than jervine. In high-fat diet- (HFD-) induced obese C57BL/6J mice, VN administration suppressed body weight gain. The levels of peroxisome proliferator-activated receptor gamma (PPARγ), CCAAT-enhancer-binding protein alpha (C/EBPα), adipocyte fatty-acid-binding protein (aP2), adiponectin, resistin, and LIPIN1 were suppressed by VN, while SIRT1 was upregulated. Furthermore, VN activated phosphorylation of the liver kinase B1- (LKB1-) AMP-activated protein kinase alpha- (AMPKα-) acetyl CoA carboxylase (ACC) axis. Further investigation of cotreatment of VN with the AMPK agonist AICAR or AMPK inhibitor Compound C showed that VN can activate the phosphorylation of AMPKαin compensation to the inhibition of Compound C. In conclusion, VN shows antiobesity effects in HFD-induced obese C57BL/6J mice. In 3T3-L1 adipocytes, VN has antiadipogenic features, which is due to activating the LKB1-AMPKα-ACC axis. These results suggest that VN has a potential benefit in preventing obesity.

Flubendiamide, a ryanoid class insecticide, is widely used in agriculture. Several insecticides have been reported to promote adipogenesis. However, the potential influence of flubendiamide on adipogenesis is largely unknown. The current study was therefore to determine the effects of flubendiamide on adipogenesis utilizing the 3T3-L1 adipocytes model. Flubendiamide treatment not only enhanced triglyceride content in 3T3-L1 adipocytes, but also increased the expression of cytosine-cytosine-adenosine-adenosine-thymidine (CCAAT)/enhancer-binding protein α and peroxisome proliferator-activated receptor gamma-γ, two important regulators of adipocyte differentiation. Moreover, the expression of the most important regulator of lipogenesis, acetyl coenzyme A carboxylase, was also increased after flubendiamide treatment. Further study revealed that 5-aminoimidazole-4-carboxamide ribonucleotide (AICAR) or A769662, two Adenosine 5′-monophosphate (AMP)-activated protein kinase α activators, subverted effects of flubendiamide on enhanced adipogenesis. Together, these results suggest that flubendiamide promotes adipogenesis via an AMPKα-mediated pathway.

Chicken preadipocytes cultured in cocktail supplement with rosiglitazone resulted in a marked increase in lipid droplet accumulation, glycerol-3-phosphate dehydrogenase (GPDH) activity and mRNA expression of adipocyte fatty acid-binding protein (aP2), G0/G1 switch gene 2 (G0S2), peroxisome proliferator-activated receptor γ (PPARγ) and lipolysis. The present study provides a novel induction method for in vitro chicken preadipocyte differentiation.
