Cercis canadensis, Eastern Redbud, is very Cercis canadensis, Eastern Redbud, is very susceptible to infection by Verticillium Wilt caused bysusceotible to infection by Verticillium Wilt caused by the common soil-borne fungi Verticillium albo-atrum and Verticillium dahliae. Little is known about the inoculum levels, the time required for natural infection to occur and how fast the pathogen travels inside the host species. One-year-old Cercis canadensis seedlings were planted in 7.6 liter (2-gallon) containers with a 1:1:2 soil/sand/perlite mix inoculated with five levels (0, 10, 100, 500, and 1000 microsclerotia/g soil) of V. dahliae prior to planting. At the end of the first growing season, half of the plants were removed from the containers, surface sterilized, dissected and root sections plated out on a Verticillium selective media. The remaining plants were grown for a second season. Infection first occurred in plants which received 100, 500 or 1000 ms/g at the end of the first season. The infection had spread at least 5 cm during the first growing season.

The results of the study of the duration of the juvenile period of indigenous Chechen willow leaf pear genotypes (Pyrus salicifolia Pall.) are given in connection with the acceleration of the breeding process and the use of selected forms in pear breeding for high precocity. The studies were carried out in 2016-2019 at OOO “Orchards of Chechnya” in accordance with the Agreement on creative cooperation with the Russian Research Institute of Fruit Crop Breeding. The work was carried out in accordance with generally accepted programs and methods. The objects of the study were one-year and two-year-old pear seedlings obtained from sowing seeds of selected dwarf and low-growing local Chechen forms of willow pear (P. salicifolia Pall.), laying fruit buds on annual growths and seedlings of Caucasian pear (P. caucasica Fed.), 20 500 pcs. of each specie. The aim of the research was to study the potential of precocity of willow pear seedlings and to reveal of selected forms with the greatest degree of this trait. Stratified seeds were sown in the sowing department of the OOO “Orchards of Chechnya” production nursery in April, 2017. The seedlings were grown according to the common technology in dryland conditions on the plot with chestnut soil. The first fl owering of plants was noted in the spring, 2019. As a result of the research, for the first time on a large number of the experimental material it was found that in the off spring of the indigenous Chechen willow leaf pear genotypes, the selection of a little more than 2% of seedlings with a very short juvenile period (2 years) was possible. They are of great interest in accelerating the breeding process and in the selection of new pear varieties with high precocity. 20 willow leaf pear genotypes were selected for the further use in breeding for high precocity and as sources of the trait of short juvenile period.

Background:The observation of an intraluminal common carotid artery thrombus overlying a wall defect at ultrasonography or angiography is unusual. To our knowledge, there are no previous reports of a free-floating thrombus in the common carotid artery.Case Report:A 45-year-old woman who was previously healthy and on no medications presented with acute hemiparesis and aphasia. Following testing that included carotid duplex and trancranial Doppler ultrasonography, diffusion-weighted magnetic resonance imaging, and digital subtraction angiography, the patient underwent emergency open embolectomy. No underlying wall defect was seen at the time of imaging or surgery. No obvious hypercoagulable state could be identified. Her NIH Stroke Scale score improved from 26 at admission to 2 at three months and 1 at one year.Conclusions:Multimodal imaging may have improved diagnosis and management in this patient with a unique finding. The source of the thrombus remains obscure.

Children are primarily examined by paediatricians. In Nepal, child health is in low priority due to illiteracy, there is total lack of awareness about children’s eye health. The common avoidable causes of childhood blindness are refractive errors, amblyopia, retinopathy of prematurity (ROP), vitamin A deficiency (VAD), xerophthalmia, ophthalmia neonatorum (ON), congenital cataract, and retinoblastoma. Paediatricians could be the key persons for early referral of these children to a paediatric ophthalmologist. Paediatricians can send the patients for eye examination after birth within 6 weeks, at 6 months, at one year and before going to school. In conclusion, early screening and referral by the paediatricians to the paediatric ophthalmologist prevents children from being sightless. Wood Mountain loam was wetted with water or (NH4)2SO4 solution to provide a factorial combination among three moisture and three NH4-N levels. Samples in polyethylene bags were incubated at 2.5-cm depths in fallow, and in an incubator that simulated the diurnal patterns of temperature fluctuation recorded in the field. During the growing season, treatments were sampled regularly for moisture, NO3− and exchangeable NH4-N. Similar determinations were made on in situ samples taken in fallow Wood Mountain loam. The incubator simulated the effects of growing season temperatures on soil N transformations satisfactorily. Pronounced increases or decreases in temperature led to flushes in N mineralization. However, in the 1972 growing season, temperature was suboptimal and temperature changes were generally small. Consequently, when a stepwise multiple regression technique was used to analyze the data, neither ammonification nor nitrification showed a quantitative relationship to temperature. Comparison of the nitrification occurring in laboratory-incubated soils with that occurring in situ led to the conclusion that 70 to 90% of the NO3-N produced in surface soil resulted from wetting and drying. Estimates of potentially ammonifiable soil N(No) and its rate of mineralization (k) were derived from cumulative ammonification by assuming that the laws of first-order kinetics were applicable. In the 10, 15, and 20% moisture treatments the average No was 27, 41, and 82 ppm, respectively. Under the conditions of this study, the time required to mineralize half of No was about 7 wk.

The purpose of this research is to determine the impact of the turf-only substrate and turf–perlite in the ratio 2:1 and of growth regulators in the quality of adventive roots ( the number and length) of well lignified one-year old branches without fruit buds in the Bluecrop cultivar (Vaccinium corymbosum L.) taken at the end of the latent period before budding at the February 15 th during the -2015 growing season. In order to support the increase of the number of roots and their length the hardwood cuttings are treated with different IBA and NAA concentrations (1500, 3000, 4500 ppm), while a part of cuttings were untreated control. The number and the length of roots have increased in relation to the increase of concentration from 1500 to 3000 ppm followed by a decline of these values in concentrations over 3000 ppm. Respectively, the number of roots (8) and the higher values of root length (4.6 cm) are achieved in the turf–perlite substrate, IBA 3000 ppm (compared to the turf-only substrate). The presence of perlite helps the aeration of the substrate and supports biochemical and physiological processes which lead to the inducing of adventive roots. Regarding the number and length of roots an important variation for (p<0.05) was observed between different concentrations of IBA and NAA. In general the effect of IBA was a lot better than the effect of NAA.

Major advances have been made in understanding the dynamics of humoral immunity briefly after the acute coronavirus disease 2019 (COVID-19). However, knowledge concerning long-term kinetics of antibody responses in convalescent patients is limited. During a one-year period post symptom onset, we longitudinally collected 162 samples from 76 patients and quantified IgM and IgG antibodies recognizing the nucleocapsid (N) protein or the receptor binding domain (RBD) of the spike protein (S). After one year, approximately 90% of recovered patients still had detectable SARS-CoV-2-specific IgG antibodies recognizing N and RBD-S. Intriguingly, neutralizing activity was only detectable in ~43% of patients. When neutralization tests against the E484K-mutated variant of concern (VOC) B.1.351 (initially identified in South Africa) were performed among patients who neutralize the original virus, the capacity to neutralize was even further diminished to 22.6% of donors. Despite declining N- and S-specific IgG titers, a considerable fraction of recovered patients had detectable neutralizing activity one year after infection. However, neutralizing capacities, in particular against an E484K-mutated VOC were only detectable in a minority of patients one year after symptomatic COVID-19. Our findings shed light on the kinetics of long-term immune responses after natural SARS-CoV-2 infection and argue for vaccinations of individuals who experienced a natural infection to protect against emerging VOC.

A disease in the koi carp (Cyprinus carpio koi) and the common carp (Cyprinus carpio carpio), caused by the herpesvirus and accompanied by a high mortality rate, has spread across numerous fish ponds all over the world since 1998, resulting in massive mortality and significant financial losses. The herpesvirus-like virus, called the koi herpesvirus (KHV) has been isolated and identified from the koi and the common carp in the course of the incidences of massive mortalities. The first appearance of a disease with a high mortality in the common and the koi carp caused by the koi herpesvirus (KHV) was described in 1998 in Israel and the United States of America (USA). Since that time, a large number of cases of outbreaks of this disease have been confirmed throughout the world, including the USA, Israel, and a large number of European countries. The deaths occurred seasonally, in late spring or early autumn, when the water temperature was from 18-28?C. The most important factor of the environment that affects the occurrence and gravity of this disease is the water temperature. This disease is currently considered one of the factors that present the biggest threat to populations of the common and the koi carp. Diseased fish are disoriented, their movements uncoordinated, their breathing rapid, gills swollen, and they have local skin lesions. The virus was isolated from tissue of diseased fish and cultivated on a KF-1 (koi fin cells) cell line. Electronic microscopy examinations revealed virus identical viral particles of the Herpesviridae family. Analyses of the virion polypeptide and DNA established differences between the KHV and the previously known herpesvirus of the Cyprinida family, Herpesvirus cyprini (CHV), and the virus of the channel catfish (Channel catfish virus - CCV). In the years 2004 and 2005, high mortality was established among one-year and two-year carp fry on three fish ponds. At two ponds, the deaths occurred among one year and two-year carp fry during the spring period, when the water temperature was over 18?C. During the autumn period, mortality was recorded among one-year carp fry at water temperatures above 23?C. On the grounds of the determined pathomorphological changes and the high mortality during the period of the year when the temperature was above 18?C, we suspect that KHV is also present in fish ponds in Serbia, even though the virus itself has not been isolated. The objective of this work is briefly to present the relevant data on this disease which is inflicting significant losses to carp production, to show the current distribution of this disease, the diagnostic methods, and the possibilities for the prevention and control of KHV.

The enhanced transmissibility and immune evasion associated with emerging SARS-CoV-2 variants demands the development of next-generation vaccines capable of inducing superior protection amid a shifting pandemic landscape. Since a portion of the global population harbors some level of immunity from vaccines based on the original Wuhan-Hu-1 SARS-CoV-2 sequence or natural infection, an important question going forward is whether this immunity can be boosted by next-generation vaccines that target emerging variants while simultaneously maintaining long-term protection against existing strains. Here, we evaluated the immunogenicity of INO-4800, our synthetic DNA vaccine candidate for COVID-19 currently in clinical evaluation, and INO-4802, a next-generation DNA vaccine designed to broadly target emerging SARS-CoV-2 variants, as booster vaccines in nonhuman primates. Rhesus macaques primed over one year prior with the first-generation INO-4800 vaccine were boosted with either INO-4800 or INO-4802 in homologous or heterologous prime-boost regimens. Both boosting schedules led to an expansion of antibody responses which were characterized by improved neutralizing and ACE2 blocking activity across wild-type SARS-CoV-2 as well as multiple variants of concern. These data illustrate the durability of immunity following vaccination with INO-4800 and additionally support the use of either INO-4800 or INO-4802 in prime-boost regimens.

The objective of this study was to compare the structure and efficacy in terms of retarding cuticular transpiration of leaf cuticles from eastern redbud (Cercis canadensis L.) and dull-leaf and glossy-leaf Mexican redbud [Cercis canadensis var. mexicana (Rose) M. Hopk.]. Leaves of Mexican redbud exhibited several xeromorphic characteristics compared to eastern redbud: a smaller, thicker leaf with thicker cuticles, more cuticular wax, a higher specific leaf mass, and greater hydrated water content on a leaf area basis. Mexican redbuds with a glossy leaf differed from those with a dull leaf only in a thicker adaxial cuticle lacking wax crystallite on the surface. Epicuticular wax crystallite were present on the abaxial surface of all leaves examined. Detached leaves of eastern redbud had a higher water loss rate than those of Mexican redbud only on a dry mass basis, not on a leaf area basis. There was no difference in the rate of water loss by detached leaves of glossy-leaf and dull-leaf Mexican redbuds after 4 hours.

Abstract A white flowering Eastern redbud (Cercis canadensis var. alba L.) has been successfully micropropagated. Two node explants collected from the initial flush of spring growth were cultured on woody plant medium (WPM). Increased shoot multiplication occurred at 10,15 and 20 μM (2.3, 3.4 and 4.5 ppm) benzyladenine (BA). Microshoots were rooted in vitro on half strength WPM with a 15-day treatment of 100 and 300 μM (18.6 and 55.9 ppm) α-naphthaleneacetic acid (NAA) or 100 and 300 μM (20.3 and 60.9 ppm) indolebutyric acid (IBA) prior to being moved to full strength WPM without growth regulators. Percentage rooting and the mean number of roots per cutting were comparable between NAA and IBA treated microcuttings, however, the subsequent root morphology differed between the two treatments. NAA treated plants developed a coarse, unbranched root system, while IBA treated cuttings developed a more desireable fine, branched root system. Rooted microshoots were successfully acclimated to greenhouse conditions.
