Cationic and zwitterionic polyelectrolytes are synthesized through atom transfer radical polymerization (ATRP), comprising a polyfluorene backbone with a small fraction of 2,1,3-benzothiadiazole and poly[2-(dimethylamino)ethyl methacrylate] (PDMAEMA) side chains. Due to higher charge density generated from grafted side chains, two polymers show higher water solubility and higher quantum yield. In comparison with cationic polyelectrolytes, zwitterionic polyelectrolytes are stable over a broad pH range from 1 to 13, even in 1 M NaCl solution. The absence of FRET between zwitterionic polymers and dye-labeled ssDNA indicates their ultralow nonspecific adsorption, while cationic polymer shows much stronger nonspecific interactions. The MTT assay of zwitterionic polymers exhibits their minimal cytotoxicity and potential in long-term clinical application. Most importantly, zwitterionic polymer could be efficiently taken up by cells, whereas cationic polymer stains the surface of cell due to membrane disruption generated from positive charges. The results illustrate that conjugated zwitterionic polymer could serve as a novel type of highly efficient ultralow fouling material with low cytotoxicity for labelling cell or potential biomedical applications.

Abstract To induce the water solubility of hexagonal boron nitride (h-BN), we exfoliated and functionalized bulk h-BN with hydroxyl groups (h-BN-OH-n). Short-term studies showed that h-BN-OH-n induced low cytotoxicity in different models: insect haemocytes (in vivo), human erythrocytes and mouse fibroblasts (in vitro). We also demonstrated that Alexa Fluor 647-h-BN-OH-n administered topically to the insects passed through the cuticle barrier and was phagocytosed by haemocytes. Nanoflakes did not affect the haemocyte cell membrane and did not interfere with the phagocytosis of latex beads. Long-term immunoassays showed that h-BN-OH-n, despite not inducing haemocytotoxicity, impaired nodulation, the most important cellular immune response in insects. The haemocytes exposed to h-BN-OH-n and then to bacteria differed in morphology and adhesiveness from the haemocytes exposed only to bacteria and exhibited the same morphology and adhesiveness as the control haemocytes. The h-BN-OH-n-induced decrease in nodulation can therefore result from the reduced ability of haemocytes to recognize bacteria, migrate to them or form microaggregates around them, which can lead to dysfunction of the immune system during pathogen infection. Long-term in vivo studies with animal models are still necessary to unambiguously confirm that h-BN is biocompatible and useful for application as a platform for drug delivery or for bioimaging.

Zwitterionic polymers are suitable for replacing poly(ethylene glycol) (PEG) polymers because of their better antifouling properties, but zwitterionic polymers have poor mechanical properties, strong water absorption, and their homopolymers should not be used directly. To solve these problems, a reversible-addition fragmentation chain transfer (RAFT) polymerization process was used to prepare copolymers comprised of zwitterionic side chains that were attached to an ITO glass substrate using spin-casting. The presence of 4-vinylpyridine (4VP) and zwitterion chains on these polymer-coated ITO surfaces was confirmed using 1H NMR, FTIR, and GPC analyses, with successful surface functionalization confirmed using water contact angle, X-ray photoelectron spectroscopy (XPS), and atomic force microscopy (AFM) studies. Changes in water contact angles and C/O ratios (XPS) analysis demonstrated that the functionalization of these polymers with β-propiolactone resulted in hydrophilic mixed 4VP/zwitterionic polymers. Protein adsorption and cell attachment assays were used to optimize the ratio of the zwitterionic component to maximize the antifouling properties of the polymer brush surface. This work demonstrated that the antifouling surface coatings could be readily prepared using a “P4VP-modified” method, that is, the functionality of P4VP to modify the prepared zwitterionic polymer. We believe these materials are likely to be useful for the preparation of biomaterials for biosensing and diagnostic applications.

Polymethylmethacrylate (PMMA) resin is one of the most commonly used materials for fabricating the base of removable orthodontic appliances. It is known that PMMA resin can promote the adhesion of microbes due to its surface porosity and from long-term use. Vanillin-incorporated PMMA resin has been reported to have antimicrobial effects against Candida albicans. However, the influence of vanillin incorporation on the water sorption and solubility of self-curing orthodontic PMMA resin has not been studied. Objective: To determine the water sorption and solubility of self-curing orthodontic PMMA resin incorporated with different concentrations of vanillin. Materials and methods: Three groups of self-curing orthodontic PMMA resin incorporated with 0.1% and 0.5% vanillin as well as PMMA without vanillin as a control were prepared with ten specimens per group. Water sorption and solubility tests were performed according to ISO 20795-2:2013 specifications. One-way ANOVA and Tukey’s multiple comparison tests at a p<0.05 significance level were used to analyse the data. Results: All three groups met ISO standard requirements. The means of water sorption values showed insignificant differences among the three groups even though the value of the 0.5% vanillin group tended to decrease. Water solubility of the 0.1% vanillin-incorporated group was not significantly different when compared with the control group whereas that of the 0.5% vanillin-incorporated group exhibited a significant lower value when compared with both control and 0.1% vanillin-incorporated groups. This suggests that increasing concentrations of vanillin would decrease the water solubility of PMMA resin. Conclusion: The incorporation of 0.5% vanillin into self-curing orthodontic PMMA resin could significantly reduce its water solubility and showed a trend of less water sorption property. Even with different vanillin concentrations, the water sorption and water solubility properties of PMMA resin were within the ISO standards. Nevertheless, further studies are recommended to determine the amount of residual monomer present in different concentrations of vanillin-incorporated PMMA resin.

Graphene oxide (GO), a 2D nanomaterial, is a promising material for medical application, thanks to its water solubility, antibacterial activity and relatively low cytotoxicity. However, many factors, such as lateral dimension, purity and surface chemistry, may influence its antibacterial activity, its exact mechanism is still unknown. In this work, E. coli was used as model bacterium to determine the antibacterial activity of well-dispersed GO which was obtained by a modified Hummer method and dialyzed to remove the salts and acid used in the oxidation process. After co-culture with GO for 2[Formula: see text]h, up to 90% E. coli cells were inactivated when GO concentration at 8[Formula: see text][Formula: see text]g/mL. The direct interaction was not detected in FE-SEM images and the results of [Formula: see text] potential showed that the interaction between GO and E. coli are repulsive[Formula: see text] Our results showed that GO can produce ROS and inactivate SOD and CAT enzymes in low concentration after co-cultured with E. coli which explained the antibacterial activity of GO in aqueous solution. Meanwhile, GO, with high purity, showed low cytotoxicity towards mammalian cells and did not cause any observable hemoglobin after co-cultured with blood cells. The data presented here prove that GO is effectively inhibit E. coli through inactivating SOD, CAT enzymes and the oxidative stress produced by ROS. Furthermore, the good biocompatibility promised its future application.
