Anti-Inflammatory Activity of Ethanol Extract of Marine Sponge Petrosia Sp. by Supression the Level of Tumor Necrosis Factor-alpha

Inflammation is the host's protective response to any stimulus that harms the body. Excessive inflammatory process causes tissue damage. Therefore, an anti-inflammatory agent is needed. The use of natural ingredients, especially sea sponges, is an option to reduce the side effects of anti-inflammatory agents. This utilization is related to the discovery of new agents. So, we tested the effect of the ethanol extract of Petrosia sp. as an anti-inflammatory agent. Animal induced with 1% carrageenan and left for 1 hour. After that the animals were divided into 6 groups (n = 4) and given oral treatment, namely: Group I (normal group); Group II (negative group); Group III (ethanol extract of Petrosia sp. Concentration of 0.05mg/ml); Group IV (ethanol extract of Petrosia sp. Concentration 0.1mg/ml); Group V (ethanol extract of Petrosia sp. Concentration 0.2mg/ml); and Group VI (positive group, Diclofenac Sodium). After 1 hour, the animals were measured for edema volume and plasma TNF-α levels. Based on the research conducted, the ethanol extract of Petrosia sp. decreased edema volume and plasma TNF-α levels in inflammatory mice. The concentration of 0.2mg/mL had a significant effect on the negative control used (p <0.05). On the other hand, Petrosia sp. indicates the presence of alkaloids, flavonoids, and steroids. They may play an important role in the anti-inflammatory process. Thus, it can be concluded that the ethanol extract of Petrosia sp. has anti-inflammatory activity.

In-flammation is the body’s reaction to foreign substances that enter the body with signs of redness, heat, swelling, pain, and impaired organ functions. Curcuma rhizome including medicinal plants that have properties relieve the pain and inflammation of the skin. The purpose of this research was to examine the anti-iflammation effect of Curcuma zedoaria (Berg) Roscoe Rhizome infusion on carrageenan induced paw edema in rats. This research used experimental method with completely randomized design. Twenty five healthy Wistar male rats, 2-3 months old and 150-250 g divided into five groups and each group consist of 5 rats. Rats in group I that served as negative control were given 2.5 mL/200gbw of aquadest. Rats in group II as positive control were given sodium diclofenac with dose of 6.75 mg/kgbw. Group III, IV, and V were given Curcuma zedoaria (Berg) Roscoe Rhizome infusion with dose of 0.625, 1.250 and 2.500 g/kgbw, respectively. Treatments were given per oral 1 hour before injection of 0.1 ml carrageenan 1% subplantar. Rat paw edema volume was measured before and after carrageenan injection. The measurement was done every 0.5 hour for 6 hours observation. Area Under the Curve (AUC) that is calculated from paw edema volume data, was use to calculate percentage of anti-inflammatory effect. Data were analyzed with one way Anova and Least Signifi cant Difference with confi dence level 95%. Infusion of Curcuma zedoaria Rhizome with doses of 0,625, 1,250, and 2,500 g/kgbw had anti-inflammatory effect in Wistar male rats which were induced by Carrageenan 1%. Percentage of anti-inflammation effect (44,16+5,11)%, (48,70+7,05)%, (59,09+9,61)% respectively and the effects were equivalent with positive control.Key words: Anti-inflammation, Curcuma zedoaria (Berg) Roscoe, infusion, Carrageenan.

Traditionally, (Tarenna polycarpa (Miq.) Koord Ex Valeton as known as marbosi-bosi which commonly found in Sibolga, North Sumatera, Indonesia has been used as antidiabetes, cholesterol, anti-inflammatory and antibacterial. Tarenna species has been found its activities as antimicrobial, anti-oxidant and anti-inflammatory activity. The aim of this study was to investigate the anti inflammatory effect ethanol extract of marbosi-bosi leaves (Tarenna polycarpa (Miq.) Koord Ex Valeton in terms of decreased edema volume of male white rat in 1% carrageenan-induced and also to determine the effective dose of extract to decrease the volume of rat paw edema Ethanol extract of marbosi-bosi (Tarenna polycarpa (Miq.) Koord Ex Valeton was obtained by maceration. The antiinflammatory activity test was divided into 5 groups. The Group I (negative control) was given CMC 0.5%, Group II (positive control) was given diclofenac sodium 2,25 mg / kg BW, while Group III, IV and V were  given marbosi-bosi leaf extract at a dose of 50, 100 and 200 mg/kgBW respectively.  Each rat was induced by 1% carrageenan subplantar injection. Examination of antiinflammatory effect was measured by using digital plethysmometer at minute of 30 to minute of 360. The data were analyzed statistically using ANOVA (analysis of variance).The results showed that negative control did not show anti-inflammatory effect had significant differences with other treatment groups. In conclusion, ethanol extract marbosi-bosi (Tarenna polycarpa (Miq.) Koord Ex Valeton has an effective anti-inflammatory activity at a dose of 100 mg / kgBW.

Abstract. Capsaicin, the main constituent in chili, is an extremely spicy vanillin alkaloid and is found in several Capsicum species in China. Traditionally, it has been used to treat inflammatory diseases such as allergic rhinitis, neuralgia after shingles, refractory female urethral syndrome, spontaneous recalcitrant anal pruritus, and solid tumors. Constant stimulation of the body by inflammatory factors can lead to chronic inflammation. Capsaicin possesses anti-inflammatory activity; however, the underlying mechanism is unknown. We investigated the effect of capsaicin on the secretion of macrophage inflammatory factors in a lipopolysaccharide-induced inflammation model using 56 healthy, SPF grade, BALB/c mice. To this end, mice peritoneal macrophages were isolated and stimulated with lipopolysaccharide (1 μg/mL) and capsaicin (25, 50, 75, or 100 μg/mL) for 24 h. At all concentrations tested, capsaicin significantly promoted the phagocytosis of neutral red dye by macrophages. Furthermore, the gene expression and secretion of inflammatory cytokines significantly increased after induction with lipopolysaccharide (P<0.01); the interleukin (IL)-6 level was 204 μg/mL, tumor necrosis factor (TNF)-α level was 860 μg/mL, and nitric oxide (NO) level was 19.8 μg/mL. However, the treatment with capsaicin reduced their levels (P<0.01) and protein expression of lipopolysaccharide-induced extracellular signal-related kinase 1/2 and p65 (P<0.05). Overall, capsaicin reduced the secretion of inflammatory cytokines (P<0.01), interleukins, TNF-α (P<0.01), and NO by inhibiting the nuclear factor-kappa B and microtubule-associated protein kinase signaling pathways, and thereby reduced lipopolysaccharide-induced inflammatory response in macrophages.

Objectives: The purpose of this study was to investigate the anti-inflammatory effect ethanol extract and fraction of buni leaves (Antidesma bunius L.) in term of decreased edema volume of male white rat in 1% carrageenan-induced and also to determine the effective dose Design: The design of this study was experimental, where the extraction and fraction of buni leaves were tested for inflammation inhibition value ​​in carrageenan-induced white rats.The inflammatory activity test was divided 6 groups. The Group I (negative control) was given CMC 0.5%, Group II (positive control) was given diclofenac sodium 2,25 mg/kg bw (body weight), while Group III, IV, V and VI were given buni leaves extract and fraction at a dose of 100, 200 and 400 mg/kgbw respectively Interventions: The variable that was intervened in this study was the concentration of extract used Main outcome measure: The main measurement results in this study were to know the extracts and fractions which are capable inhibitor volume of edema in the carrageenan-induced rat paw Results: the anti-inflammatory effects from buni leaves exhibited the most effective activity to reduce edema in the rat paw i.e. EEDB 200 mg / kg bw and FEADB 200 mg / kg bw showing the same value of against positive control Conclusion: ethanol extract and ethyl acetate fraction buni leaves(Antidesma bunius L.) has an effective anti-inflammatory activity at a dose of 200 mg/kg bw.

Starting from 6-amino-4-methyl-2H-chromen-2-one 1, a series of some new heterocyclic compounds (2-14) incorporating coumarin moiety was synthesized and assessed for their anti-inflammatory activity using the carrageenan-induced hind paw edema method. The tested coumarin derivatives reduce the edema volume and serum levels of the pro inflammatory cytokines, IL-6 and TNF-α, and have effects on promoting production of anti-inflammatory cytokine, IL-10.

AbstractBackgroundInflammation is one of the most important responses of the body against infection or disease, and it protects tissues from injury; however, it causes redness, swelling, pain, fever and loss of function. The aim of this present study was to evaluate the anti-inflammatory activity of emu oil (Eu) formulated nanofibrous scaffold in HFFF2 fibroblast cells.Materials and methodsEu was formulated successfully in nanofibers through the electrospinning method. Besides, the morphological and structural properties of Eu nanofibres were evaluated using Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The MTT assay (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide) was performed to evaluate the HFFF2 fibroblast cells’ viability. Also, real-time polymerase chain reaction (PCR) was used to evaluate the anti-inflammatory signaling pathway in treated HFFF2 cells with Eu nanofiber.ResultsOur study showed that the Eu nanofiber increased the viability of fibroblast HFFF2 cells (p < 0.05). Also, the expression of interleukin1 (IL1), IL6 and tumor necrosis factor- alpha (TNF-α) pro-inflammatory cytokines genes were significantly decreased in treated HFFF2 cells with Eu nanofiber (p < 0.05).ConclusionsIn conclusion, Eu nanofiber scaffold potentially can reduce the inflammation process through downregulation of IL-1, IL-6 and TNF-α cytokines.

Extra virgin Olive oil is extracted from fruit that can be used as anti-inflammatory agent. This research aimed to test the potential of extra virgin olive oil in reducing edema volume and TNF-α plasma in carrageenan-induced rats. This research was purely experimental research with the post test control group design. A total of twenty eight Wistar rats were divided randomly into four treatment groups. Group I was a control negative group, while the group II, III, and IV were orally administered with extra virgin Olive oil at the dose of 0.9 ; 1.8 ; 2.7 mL/day, respectively. Paw edema was measured one hour before the rats was induced to carrageenan and every hour until four hours after it was induced to carrageenan. TNF-α plasma was measured at four hours. Analysis of the data was done by calculating the presentation of edema inhibition in every group, then the data was statistically analyzed by Anova, Repeated Anova, LSD and Kruskal Wallis test with 95% confidence interval. The result showed that extra virgin olive oil has an anti-inflammatory effect. The highest decrease in edema volume percentage was in group III (14.21%). There was a significant difference in the edema volume of all treatment groups at each time of the experiment with TNF-α value (p < 0.05). In conclusion, the administration of extra virgin olive oil can lower the volume of carrageenan-induced edema in rats depend on the dose. Also, the administration of extra virgin olive oil can be dose-dependent in reducing the levels of TNF-α in carrageenan-induced edema in rats.

Dipterocarpus tuberculatus Roxb. has been used traditionally as a remedy for many diseases, especially inflammation. Therefore, we analyzed and explored the mechanism of the anti-inflammatory effect of a Dipterocarpus tuberculatus Roxb. ethanol extract (Dt-EE). Dt-EE clearly and dose-dependently inhibited the expression of pro-inflammatory cytokines such as IL-6, TNF-α, and IL-1β in lipopolysaccharide (LPS)-treated RAW264.7 cells. Also, Dt-EE suppressed the activation of the MyD88/TRIF-mediated AP-1 pathway and the AP-1 pathway related proteins JNK2, MKK4/7, and TAK1, which occurred as a result of inhibiting the kinase activity of IRAK1 and IRAK4, the most upstream factors of the AP-1 pathway. Finally, Dt-EE displayed hepatoprotective activity in a mouse model of hepatitis induced with LPS/D-galactosamine (D-GalN) through decreasing the serum levels of alanine aminotransferase and suppressing the activation of JNK and IRAK1. Therefore, our results strongly suggest that Dt-EE could be a candidate anti-inflammatory herbal medicine with IRAK1/AP-1 inhibitory and hepatoprotective properties.

AbstractFisetin is a naturally occurring flavonoid that possesses several pharmacological benefits including anti-inflammatory activity. However, its precise anti-inflammatory mechanism is not clear. In the present study, we found that fisetin significantly inhibited the expression of proinflammatory mediators, such as nitric oxide (NO) and prostaglandin E2 (PGE2), and cytokines, such as interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α), in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Additionally, fisetin attenuated LPS-induced mortality and abnormalities in zebrafish larvae and normalized the heart rate. Fisetin decreased the recruitment of macrophages and neutrophils to the LPS-microinjected inflammatory site in zebrafish larvae, concomitant with a significant downregulation of proinflammatory genes, such as inducible NO synthase (iNOS), cyclooxygenase-2a (COX-2a), IL-6, and TNF-α. Fisetin inhibited the nuclear localization of nuclear factor-kappa B (NF-κB), which reduced the expression of pro-inflammatory genes. Further, fisetin inactivated glycogen synthase kinase 3β (GSK-3β) via phosphorylation at Ser9, and inhibited the degradation of β-catenin, which consequently promoted the localization of β-catenin into the nucleus. The pharmacological inhibition of β-catenin with FH535 reversed the fisetin-induced anti-inflammatory activity and restored NF-κB activity, which indicated that fisetin-mediated activation of β-catenin results in the inhibition of LPS-induced NF-κB activity. In LPS-microinjected zebrafish larvae, FH535 promoted the migration of macrophages to the yolk sac and decreased resident neutrophil counts in the posterior blood island and induced high expression of iNOS and COX-2a, which was accompanied by the inhibition of fisetin-induced anti-inflammatory activity. Altogether, the current study confirmed that the dietary flavonoid, fisetin, inhibited LPS-induced inflammation and endotoxic shock through crosstalk between GSK-3β/β-catenin and the NF-κB signaling pathways.
