Matrix metalloproteinases (MMPs) are the family of proteases that are mainly responsible for degrading extracellular matrix (ECM) components. In the skin, the overexpression of MMPs as a result of ultraviolet radiation triggers an imbalance in the ECM turnover in a process called photoaging, which ultimately results in skin wrinkling and premature skin ageing. Therefore, the inhibition of different enzymes of the MMP family at a topical level could have positive implications for photoaging. Considering that the MMP catalytic region is mostly conserved across different enzymes of the MMP family, in this study we aimed to design a virtual screening (VS) workflow to identify broad-spectrum MMP inhibitors that can be used to delay the development of photoaging. Our in silico approach was validated in vitro with 20 VS hits from the Specs library that were not only structurally different from one another but also from known MMP inhibitors. In this bioactivity assay, 18 of the 20 compounds inhibit at least one of the assayed MMPs at 100 μM (with 5 of them showing around 50% inhibition in all the tested MMPs at this concentration). Finally, this VS was used to identify natural products that have the potential to act as broad-spectrum MMP inhibitors and be used as a treatment for photoaging.

The mechanical behavior of a sandwich structure made of woven glass/epoxy skins and an additive manufactured (AM) core was studied. The AM core was fabricated using polyetherimide and fused deposition modeling. This combination was chosen to increase the out-of-plane strength of an AM aircraft part. Tensile, flexural, and puncture tests were conducted for both the AM core and sandwich structure. The behavior of the beams was governed by the skins; however, the effect of the AM core for one-sided beams was significant. The tensile strength predicted by the simple mixture rule was in good agreement with the experimental results. For the flexural test specimen, sandwich beams were tested under three-point bending. The addition of composite skins increased the elastic modulus and flexural strength in both directions. The prevalent failure modes were skin wrinkling and core failure, and failure of the compressive skin was more likely. Puncture tests were conducted at three different temperatures. Owing to the increased bending stiffness, the sandwich structure did not exhibit ductile-to-brittle transition behavior.

Abstract Out-of-plane joints, between hat (omega) stiffeners and skin panels are asymmetric parts of the composite structure. Studies show that physical-mechanical conditions in these joints significantly affect skin forming quality. In the present article, aimed to investigate the mechanism of the skin wrinkle in the joints of carbon fiber reinforced plastics (CFRP) hat-shaped structure, the pressure testing apparatus based on the Pascal principle is used to surveillance the resin pressure dynamically in out-of-plane joints. In this regard, several influencing factors such as first-order holding time, forming pressure and relative volume of unidirectional fillers are studied. Obtained results show that increasing the first stage holding time can prolong the viscous flow state of the resin, and time to achieve pressure equalization at each detection point, thereby improving the dispersion of the pressure and reducing the possibility of wrinkles. It is found that as the forming pressure increases, the degree of skin wrinkles in the out-of-plane joints ameliorates. Moreover, for fillers with a relative volume within the range of 0–50%, the pressure transfer effect and the skin flatness is relatively dissatisfactory. It is concluded that the filler with a relative volume of 80–120% improves the skin wrinkle in out-of-plane joints.

   EUTECTIC MIXTURE OF LOCAL ANAESTHETIC (EMLA) STIMULATED SKIN WRINKLING TEST IN NORMAL POPULATIONABSTRACTIntroduction: Peripheral neuropathy (PN) could reduce life quality among the affected individuals, while early detection is difficult, apparently healthy individuals could also be affected by this disorder. Stimulated skin wrinkling by eutectic mixture of local anaesthetic test (SSW-EMLA) and brief peripheral neuropathy screening (BPNS) are simple non- invasive examination to detect PN.Aims: To find out the percentage of asymptomatic neuropathy in the normal population using BPNS and SSW-EMLA.Methods: A cross-sectional study on a population that is considered as normal who come to R.D. Kandou Hospital Manado consecutively between October 2015 and September 2016. The normal population was based on modified healthy subject criteria by Lauria et al in a study about skin neural fiber morphometry. Brief peripheral neuropathy screening and SSW-EMLA were performed in all subjects. The Chi-square or Fisher exact test were used to assess between-group differences. P-value of 0.05 was considered statistically significance.Results: We enrolled 99 subjects, most of them were male (61.1%) and the median age was 31 years old. Asymptomatic neuropathy was found in 25.2% of the subjects. The highest neuropathy to non-neuropathy ratio was found in the >40 years old group (0,75). Stimulated skin wrinkling by eutectic mixture of local anaesthetic test could detect more cases compared to BPNS (24.2% vs 7.1%) and this finding was statistically significant.Discussion: The percentage of asymptomatic neuropathy in the normal population was 25.2% and mostly detected by SSW-EMLA compared to BPNS significantly.Keywords: Asymptomatic neuropathy, BPNS, normal population, SSW-EMLAABSTRAKPendahuluan: Neuropati perifer dapat menurunkan kualitas hidup penderitanya, namun deteksi dini masih sulit dilakukan, padahal orang-orang yang terlihat sehat juga dapat mengalaminya. Salah satu pemeriksaan non-invasif sederhana yang dapat dilakukan adalah stimulated skin wrinkling by eutectic mixture of local anaesthetic test (SSW-EMLA) selain skrining brief peripheral neuropathy screening (BPNS) yang biasa digunakan.Tujuan: Mengetahui persentase neuropati asimtomatik pada populasi normal menggunakan BPNS dan SSW- EMLA.Metode: Penelitian potong lintang terhadap populasi yang dianggap normal yang berobat di Instalasi Rawat Jalan RSUP Prof. Dr. R.D. Kandou secara konsekutif antara bulan Oktober 2015 sampai September 2016. Kriteria populasi normal berdasarkan modifikasi dari kriteria subjek sehat yang dibuat oleh Lauria dkk dalam penelitian tentang morfometri serabut saraf dermal kulit manusia. Subjek dilakukan pemeriksaan BPNS dan SSW-EMLA, lalu dinilai perbedaan antar kelompok dengan uji Chi-square atau Fisher’s exact. Nilai p=0,05 dijadikan batas kemaknaan statistik.Hasil: Terdapat 99 subjek yang sebagian besar adalah laki-laki (61,1%) dan median usia adalah 31 tahun. Neuropati asimtomatik ditemukan pada 25,2% subjek. Rasio mengalami dan tidak mengalami neuropati tertinggi ditemukan pada kelompok usia >40 tahun (0,75). Uji SSW-EMLA mampu mendeteksi lebih banyak kasus dibanding BPNS (24,2% vs7,1%) secara bermakna.Diskusi: Didapatkan 25,2% subjek neuropati asimtomatik pada populasi normal yang dominan ditemukan dengan uji SSW-EMLA dibandingkan BPNS secara bermakna.Kata kunci: BPNS, neuropati asimtomatik, populasi normal, SSW-EMLA

<p class="abstract"><strong>Background:</strong> Ageing is a continuous process and efforts are being made to reverse or rather slow the process. It is evident through changes in the skin, hair and nails. Due to changes in these, aesthetic appearance is compromised. Choline-stabilized orthosilicic acid (ch-OSA®) has shown results in improving skin, hair and nail health. The aim of the present study was to assess these effects in the Indian population taking oral ch-OSA®.</p><p class="abstract"><strong>Methods:</strong> Randomly selected 39 participants were assigned to 4 groups to study alopecia, hair quality, skin health and nail health. Ch-OSA®was administered for 5 months. Assessment was made at baseline, 2, 3 and 5 months. After this ch-OSA®was stopped and further assessment was made at 8 months.<strong></strong></p><p class="abstract"><strong>Results:</strong> There was a significant improvement in terms of all the 11 parameters related to alopecia and hair quality from baseline to 8 months (p&lt;0.001). Most of the participants reported grade-1 skin wrinkling at 5 or 8 months. Skin hydration also improved significantly after the ch-OSA<sup>®</sup> administration. Greater percentage of participants reported of reduced roughness and dyschromia at the end of the study. Nail parameters were also significantly improved. No side-effect was reported by the participants throughout study.</p><p class="abstract"><strong>Conclusions:</strong> Oral intake of ch-OSA<sup>®</sup> showed significant beneficial effects on the health of hair, skin and nails with good tolerability.</p>
